    Notes: Conditions were established for routine cinemicrographic examination of sperm incorporation by living zona-free mouse eggs employing oil immersion objectives and Nomarski optics. Initial sperm attachment to the egg plasma membrane, which was reversible and appeared to require flagellar activity, involved localized areas of the head corresponding approximately to the position of the equatorial segment. Penetrating sperm lay flat on the egg and, during incorporation, appeared to sink into the egg cytoplasm, accompanied by short bursts of flagellar activity and subsequent rotation of the flagellum around its insertion point. Ensuing sperm head decondensation involved dissociation of individual particulate structures and a dramatic localized clearing in the egg cytoplasm. The normalcy of the penetration process and the potential applicability of this approach was attested to by the observations that polar body extrusion, male and female pronuclear formation, and migration through the egg cytoplasm in preparation for syngamy occurred in several sequences followed for extended time periods.

    Notes: The surface configuration of zona-free mouse ovarian eggs before and after germinal vesicle breakdown (GVBD) and of tubal eggs before and after insemination was studied by scanning electron microscopy. The surface of germinal vesicle-intact eggs was characterized by uniformly distributed, dense (2-3μm2 and slender (0.1 × 0.8μm) microvilli. Following GVBD, the mouse oolemma underwent dramatic changes which persisted essentially unmodified in uninseminated tubal eggs. These changes consisted of an apparent conversion of the oolemma from a microvillar to a smooth or ruffled configuration with approximately two ruffle-like micro-projections (0.6 × 0.4 μm) perμm2 of surface. In most eggs, both ruffled and smooth configurations ere present, creating dramatic polarities in surface topography similar to those documented previously by TEM [Nicosia et al, 1977]. Following insemination and presumed sperm-egg fusion, microvilli reappeared either from smooth areas or from the upper edge of ruffled microprojections. Zona removal appeared to induce limited reduction in the size of microvilli in some eggs. The present results emphasize the labile and dynamic nature of the egg surface and suggest a close relationship among nuclear, cytoplasmic, and surface events during the final stages of egg maturation and during fertilization.

    Notes: The kinetics of spontaneous and induced acrosomal loss have been studied in human sperm incubated in capacitating and noncapacitating media. Acrosomal status was quantitated using indirect immunofluorescence with a monoclonal antibody. The response of sperm to induction by calcium ionophores was time dependent reaching a maximum after 6 hours of incubation under capacitating conditions. The inducible population slowly decreased in size through the balance of a 24-hour incubation. The time-dependent development of ionophore responsiveness by sperm exposed to capacitating conditions corroborates the idea that only capacitated cells can respond to undergo acrosomal loss in response to ionophore. In contrast, only a small, constant percentage of sperm incubated under noncapacitating conditions responded to ionophore. Substitution experiments involving the addition or deletion of human serum albumin suggest that albumin is not absolutely required for capacitation but is essential for the maintenance of motility. Polyvinyl alcohol can be substituted for serum albumin, but it does not support capacitation or motility as well as HSA. These studies may provide a basis for optimizing capacitating conditions for human sperm in vitro as well as for diagnosing fertility or fertility potential based on measurements of spontaneous and ionophore induced acrosomal loss under defined culture conditions.

    Notes: A procedure was developed for isolation of plasma membrane (PM) preparations from unfertilized mouse eggs. Zona-free mouse eggs prepared by the method of Boldt and Wolf (Gamete Res 13:213-222, 1986) were labeled with 125I-concanavalin A (ConA) prior to sonication and fractionation on iso-osmotic self-generated Percoll density gradients. Experiments using the ConA-specific sugar α-methylmannoside (αMM) indicated that 125I-ConA bound specifically to the egg PM. Greater than 95% of 125I-ConA binding to zona-free eggs was blocked in the presence of 0.1 M αMM, and incubation of eggs in αMM after 125I-ConA labeling caused release of 85-90% of bound label. Fractionation of 125I-ConA-labeled eggs by Percoll density gradient centrifugation yielded a single radioactive peak at density = 1.025, corresponding to egg PM material. Prolonged incubation of 125I-ConA-labeled eggs or egg sonicates prior to fractionation did not alter the location of the radioactive peak, indicating that 125I-ConA did not label other organelles. As a control, human erythrocytes were labeled with 125I-ConA and fractionated under identical experimental conditions and yielded a single radioactive peak at density (1.020) comparable to that observed for 125I-ConA-labeled eggs. These results indicate that 125I-ConA can be used as a specific marker to support PM isolation from small numbers of zona-free mouse eggs.

    Notes: This study was performed to evaluate the effects of mechanical stimulation and sucrose treatment on the oocyte activation process. Fresh and aged human oocytes were exposed to sucrose and zonae were dissected with microneedles before fixation and quantitative analysis of cortical granules by transmission electron microscopy. Examination of the mean number of cortical granules/analyzed segment revealed no significant differences between control oocytes or oocytes treated with sucrose or sucrose treatment followed by zona dissection. A significant decline in the number of cortical granules/segment was observed for oocytes undergoing prolonged culture after dissection (P 〈 0.05). Thus, zona dissection and sucrose exposure of freshly aspirated mature human oocytes do not result in classical oocyte activation.

    Notes: The anionic fluorescein dye, Erythrosin B, inhibited the penetration of mouse eggs in vitro. High concentrations of the dye (0.5-1.0 mM) inhibited the penetration of cumulus-intact, cumulus-free, and zona-free eggs at sperm concentrations of approximately 2 × 105 cells per milliliter. Examination of gamete viability indicated that the dye did not cause gross, irreversible changes in either amete, and we therefore concluded that dye-dependent inhibition may have resulted from dye association with cell membranes. The dye concentration required to give 50% inhibition, apparent I50, of 500 μM obtained for zonafree eggs was artificially elevated by the presence of bovine serum albumin (BSA) (2%) in the culture medium. A corrected I50 of 11 μM was estimated in the absence of BSA.

    Notes: Studies of sperm-egg fusion using zona-free mouse eggs are impaired by the procedures used for removal of the zona pellucida. Methods involving proteolytic digestion or mechanical aspiration through micropipettes are limited in that proteases can adversely affect fertility and mechanical removal often results in low egg yields. An efficient procedure for preparation of zona-free mouse eggs was developed using a combined enzymatic (chymotrypsin) mechanical approach (CT-M procedure). Zona-intact eggs, obtained after hyaluronidase treatment, were exposed to 0.001% α-chymotrypsin in medium containing 0.5% bovine serum albumin (BSA). Brief (2 minute) exposure to chymotrypsin under these conditions caused pronounced zona distention in a majority (80-90%) of the eggs, facilitating mechanical removal and resulting in a high yield of zona-free eggs. Eggs prepared by the CT-M method displayed identical penetration levels relative to mechanically denuded eggs. CT-M prepared eggs also showed sperm concentration dependent penetration levels and demonstrated a plasma membrane block to polyspermy, qualities previously observed in mechanically prepared eggs [Wolf DP, 1978, Dev Biol 64:1-10]. Eggs could be exposed to 0.001% CT for zona distention over a 2-10-minute time period with no detrimental effects on fertility. The effect of chymotrypsin was also studied by treating zona-free eggs for 30 minutes over a 1-1,000-μg/ml range of enzyme, and a concentration-dependent reduction in penetration levels was observed. These results indicate that the CT-M method is a useful procedure for the isolation of large numbers of zona-free mouse eggs.

    Notes: Unfertilized oocytes of the surf clam, Spisula solidissima, have resting membrane potentials of -18 ± 7 mV (n = 20). Within five seconds of sperm addition, an electrophysiologically detectable response was apparent, which was characterized by a rapid and prolonged depolarrization depolarization followed four to five minutes post-insemination by the beginning of the beginning of a steady hyperpolarization to approximatelv -70 mV. This final hyperpolarization was completed within ten minutes of sperm addition. The initial rapid depolarization following insemination may result from a transient increase in sodium conductance, and it may be crucial in preventing polyspermy, since the degree of polyspermy in Spisula oocytes was sensitive to external sodium ion concentrations. Evidence was obtained that changes in intracellular pH are essential for oocvte activation. Using germinal vesical breakdown (GVB) as a marker for activation, it was shown that agents that raise intracellular pH (ammonia and procaine) induced GVB, whereas agents that lower intracellular pH pH (Na-acetate or Na-propionate seawater) inhibited GVB.
