Syntheses with α-Metalated Isocyanides, XLVIII; Phosphorus Analogs of Amino Acids and Peptides, V. - Synthesis of 1-Aminoalkylphosphonic Acids by Alkylation of α-Metalated Diethyl IsocyanomethylphosphonateDiethyl 1-isocyanomethylphosphonate (3) can be monoalkylated or bialkylated with alkyl halides (using potassium tert-butoxide or sodium hydride in methylene dichloride as bases). The higher diethyl 1-isocyanoalkylphosphonates 5, on acid hydrolysis, yield either diethyl 1-aminoalkylphosphonates 7 or 1-aminoalkylphosphonic acids 8. - 1-Amino-1-phenylalkylphosphonic acids 10 are obtained analogously starting from diethyl α-isocyanobenzylphosphonate (9).
Notes:
Isocyanmethylphosphonsäure-diethylester (3) läβt sich baseninduziert (Kalium-tert-butylat oder Natriumhydrid in Methylendichlorid) zu höheren 1-Isocyanalkylphosphonsäure-diethylestern 5 mono- oder dialkylieren. Diese ergeben bei der sauren Hydrolyse wahlweise 1-Aminoalkylphosphonsäure-diethylester 7 oder 1-Aminoalkylphosphonsäuren 8. - Analog lassen sich aus α-Isocyanbenzylphosphonsäure-diethylester (9) unter Kettenverlängerung 1-Amino-1-phenylalkylphosphonsäuren (10) synthetisieren.
