    Notes: The dynamics of several prototypical molecular systems after excitation with femtosecond laser pulses at 155 nm has been studied in pump–probe experiments. The vacuum ultraviolet (VUV) pump pulses with a pulse width of 350–450 fs were generated by near-resonant four-wave difference frequency mixing in argon. The careful analysis of the time-dependent ion signals has allowed us to determine the lifetime of the excited molecular states down to about 30 fs. The extremely short lifetime of water molecules excited to the repulsive A˜ state has been directly observed for the first time: τD≤20 fs. For molecular oxygen highly excited in the Schumann–Runge band, a decay time of 40±20 fs was obtained. The lifetimes of ethylene and chloroethylenes as well as of benzene and toluene reaching from 40 up to 180 fs are primarily caused by internal conversion. The decay times τD=(1.9±0.1) and τD=(90±20) ps obtained for carbon disulfide and nitric oxide, respectively, are due to predissociation of the VUV excited states. © 1999 American Institute of Physics.

    Notes: Biochemical and genetic techniques have provided considerable insight into the structure-function relationship of one of the ADP-ribosyl transferases produced by Pseudomonas aeruginosa, exotoxin A. Exotoxin A contains a typical prokaryotic signal sequence which, in combination with the first 30 amino-terminal amino acids of the mature protein, is sufficient for exotoxin A secretion from P. aeruginosa. Determination of the nucleotide sequence and crystalline structure of this prokaryotic toxin allowed a molecular model to be constructed. The model reveals three structural domains of exotoxin A. Analysis of the identified domains shows that the amino-terminal domain (domain I) is involved in recognition of eukaryotic target cells. Furthermore, the central domain (domain II) is involved in secretion of exotoxin A into the periplasm of Escherichia coli. Evidence also implicates the role of domain II in translocation of exotoxin A from the eukaryotic vesicle which contains the toxin after it becomes internalized into susceptible eukaryotic cells via receptor-mediated endocytosis. The carboxy-terminal portion of exotoxin A (domain III) encodes the enzymatic activity of the molecule. The structure of this domain includes a cleft which is hypothesized to be the catalytic site of the enzyme. Several residues within domain III have been identified as having a direct role in catalysis, while others are hypothesized to play an important structural role.

    Notes: The yield of exotoxin A from Pseudomonas aeruginosa has been shown to be strain-dependent. Exotoxin A production requires the presence of the positive regulatory gene, regA. We cloned the regA genetic locus from the prototypical P. aeruginosa strain PAO1 and examined its ability to influence exotoxin A yields compared to the same region cloned from the hyper-toxin-producing strain, PA103. The P. aeruginosa regA mutant strain, PA103-29, containing the PAO1 regA locus in trans produced approximately five to seven times less extracellular exotoxin A than PA103-29 containing the regA locus cloned from the hyper-toxigenic strain, PA103. Nucleotide sequence analysis of the PAO1 regA locus revealed several differences, the most striking of which was the absence of a second open reading frame that was present in the analogous PA103 DNA. In addition, an amino acid substitution was found at position 144 of RegA (Thr in PAO1 and Ala in PA103). Recombinant molecules were constructed to test the contribution of each of these changes in nucleotide sequence on extracellular exotoxin A yields. The amino acid substitution in the PAO1 RegA protein was found not to affect overall exotoxin A yields. In contrast, the presence of the second open reading frame immediately downstream of the PA103 regA gene was found to influence extracellular exotoxin A yields. This open reading frame encodes a gene which we call regB. Nucleotide sequence analysis indicates that regB is 228 nucleotides in length and encodes a protein of 7527 Daltons. Our data suggest that regB is required for optimal exotoxin A production and its absence in strain PAO1 partially accounts for the difference in yield of extracellular exotoxin A between P. aeruginosa strains PAO1 and PA103.

    Notes: Abstract The effects of the insecticide lambda-cyhalothrin (Karate) on non-target arthropods in winter wheat were studied throughout two successive seasons in 1998 and 1999. The study particularly focussed on the crop in the growing season after insecticide application (also winter wheat) for detection of potential long-term effects and for determination of the suitability of different sampling methods.The investigations were based on the assumption that arthropod immigration from surrounding areas is limited in large fields. For this reason a simple approach seemed to be feasible. Two plots of equal size (10 ha, adjusted to each other) were defined in a 100 ha field and designated control and treatment plots. Ten sampling points were established on each plot. The following monitoring methods were utilised: visual counting, sweep netting and pitfall trapping. In the first year of investigation, countings and catches were carried out 2 days prior to insecticide application and 2, 16, 30 and 44 days after application. In the next year, they were carried out 365 days and 384 days after insecticide application in the successive crop of winter wheat.At the time of the first sampling prior to insecticide application, the two plots showed significant differences with respect to arthropod density or activity, particularly in visual counting and to a minor degree to sweep netting and pitfall trapping. Lower densities or activities were observed in the plot reserved for treatment. Measures for mathematical equalisation of the results of population densities before pesticide treatment should be considered.After insecticide application, the densities or activities of non-target arthropods decreased, particularly in visual counting and sweep netting. After one year, these effects disappeared to a large extent. Several groups of arthropods reached even higher levels in the treated plot than in the untreated one. The pitfall traps revealed weak activity-decreasing effects in carabids and spiders in the treated plot, but the opposite tendency for staphylinids. Hence, it seems that the conditions in a large field are less homogeneous, and that smaller scale conditions can support processes of recovery in non-target populations.

    Notes: Summary Compared to doxorubicin, equimolar epirubicin toxicity is reduced by about 50% by the epimerization of a hydrogen and hydroxyl group at the 4′ position of the anthracycline sugar moeity. The circadian timing of doxorubicin administration markedly affects its lethal and sub-lethal bone marrow and gut toxicities in mice, as well as the severity of its clinical toxicity. We tested whether the timing of administration of equitoxic epirubicin doses similarly affected the toxicological response in female CD2F1 mice. A large and highly reproducible effect of the circadian stage of administration was documented with best drug tolerance occurring during the first half of the daily rest (light) span of the animals. In addition to this circadian rhythm, a significant seasonal effect was found with significantly fewer deaths occurring after epirubicin was given in the Summer, as compared to the Winter. Safest circadian timing for epirubicin is statistically significantly earlier in the day than for doxorubicin, while their seasonal patterns are quite similar.
