    Description: This brief article provides some of the background to the publication of The Freshwater Algal Flora of the British Isles (John et al. 2002). This publication updates West and Fritsch's A Treatise on British Freshwater Algae (2nd edition) which was published in 1927. Taxonomic experts on the major freshwater algal groups were approached and almost all agreed to collaborate. The book which has taken more than 10 years to complete is illustrated with over 2000 line drawings and there are 20 half tone plates.

    Notes: We present the results of a phylogenetic study, based on amplified ribosomal DNA restriction analysis of the rDNA operon, of 37 Arthrospira (‘Spirulina’) cultivated clonal strains from four continents. In addition, duplicates from different culture collections or markedly different morphotypes of particular strains established as clonal cultures were treated as separate entries, resulting in a total of 51 tested cultures. The strain Spirulina laxissima SAG 256.80 was included as outgroup. The 16S rRNA genes appeared too conserved for discrimination of the strains by amplified ribosomal DNA restriction analysis, and thus the internally transcribed spacer was selected as molecular taxonomic marker. The internally transcribed spacer sequences situated between the 16S and the 23S rRNA were amplified by polymerase chain reaction and yielded amplicons of about 540 bp. Direct use of cells for polymerase chain reaction seemed to inhibit the amplification reaction. This was overcome by the design of a crude lysis protocol and addition of bovine serum albumin in the polymerase chain reaction mix.The amplicons were digested with four restriction enzymes (EcoRV, HhaI, HinfI, MseI) and the banding patterns obtained were analyzed. Cluster analysis showed the separation of all the strains into two main clusters. No clear relationships could be observed between this division into two clusters and the geographic origin of the strains, or their designation in the culture collections, or their morphology.

    Notes: Genomic rearrangements involving amplification of metallothionein (MT) genes have been reported in metal-tolerant eukaryotes. Similarly, we have recently observed amplification and rearrangement of a prokaryotic MT locus, smt, in cells of Synechococcus PCC 6301 selected for Cd tolerance. Following the characterization of this locus, the altered smt region has now been isolated from a Cd-tolerant cell line, C3.2, and its nucleotide sequence determined. This has identified a deletion within smtB, which encodes a trans-acting repressor of smt transcription. Two identical palindromic octanucleotides (5′-GCGATC-GC-3′) traverse both borders of the excised element. This palindromic sequence is highly represented in the smt locus (7 occurrences in 1326 nucleotides) and analysis of the GenBank/EMBL /DDB J DNA Nucleotide Sequence Data Libraries reveals that this is a highly iterated palindrome (HIP1) in other known sequences from Synechococcus strains (estimated to occur at an average frequency of once every c. 664 bp). HIP1 is also abundant in the genomes of other cyanobacteria. The functional significance of smtB deletion and the possible role of HIP1 in genome plasticity and adaptation in cyanobacteria are discussed.

    Notes: Abstract Standard methods for sampling, fixation and quantification of organisms in highly acid environments with pH values at or below 3 are presented. Some important problems are discussed, together with recommendations on how to handle the material. Examples of specific problems are included, especially the effects of fixatives on the cell dimensions of species of Chlamydomonas, Oxytricha and Actinophrys, and on cell numbers of Actinophrys. Mixed populations of heterotrophic protists should be preserved with Lugol's solution, because other fixatives do not permit the recognition of heliozoans.

    Notes: Abstract There is an increasing awareness of the need to assess the impact of nutrient enrichment on river ecosystems separately from the impacts of organic effluents. A range of methods have been proposed and some have moved from the development stage to practical use by water management organizations. The methods can be applied to broad surveys or provide baseline information to assess possible future change. In the latter case it is recommended that several different methods are used, especially where it is important to get reliable information on the long-term impact of improvements in effluent quality. Estimates of biomass measured as chlorophyll a have often been used for phytoplankton and sometimes also for benthic communities. However, a lot of care is needed in applying this method, because of the range of factors besides nutrient concentration which can influence values. Approaches based on the whole community have been developed by a number of research groups, usually involving semiquantitative estimates of abundance. There has also been a rapid increase in the use of indices based on the relative proportions of epilithic diatom species. The methodologies used by a number of research groups in Europe are broadly similar, making it possible to compare results between different regions. The development of indices based on macrophyte floristic composition in relation to river nutrient status is also under development, especially in France and UK. However, interpretation of the results is complicated where long-term changes are taking place in nutrient concentrations in the water, because of the varying contributions of sediment and water to different species of rooted plant. Bioassays can be especially helpful where it is desired to establish whether either N or P is limiting for a population of community.

    Notes: Abstract The phosphomonoesterase activity of a microbialcommunity dominated by Chroococcidiopsis in arock sample from the Antarctic dry desert was comparedwith that of a non-axenic culture of thecyanobacterium isolated from the same rock in order toestablish whether the organism differed in itsproperties when immobilized in the rock. The pHoptimum (9.5) was the same, but the response totemperature and light both differed. In the case oftemperature, the optimum was much higher in culture. Light flux at the low value of 8 μmol photonm-2 s-1 was slightly inhibitory for acrushed rock sample, but enhanced activity of thecultured material. Although materials both showeda typical (apparent) Michaelis-Menten relationship,the values for Km and Vmax differed. Itis suggested that study of natural immobilized systemsis important for the development of procedures to useimmobilized phototrophs for practical purposes.
