    Notes: We have investigated the Ca2+-dependent interactions of villin, a protein of the intestinal microvillar core, with actin by monitoring resonance energy tranfer between fluorescently labeled actin subunits. In the presence of elevated free Ca2+(∼20 μM), villin affects both the nucleation and the elongation phases of actin polymerization. Consistent with previous reports, villin stimulates the nucleation process and will form stable nuclei under depolymerization conditions. Compared to the control, the net rate of polymerization is slightly inhibited at low con-centrations of villin (villin/actin ∼ 1:400) but is stimulated at higher concentrations (villin/actin 〉 1:100). Villin also significantly increases the critical concentration of actin polymerization. Addition of either villin or villin-actin complexes induces depolymerization of preassembled actin filaments. This villin-induced depolymerization is reversible upon removal of free Ca2+ or upon the addition of phalloidin. The exchange of actin subunits at steady state is inhibited at low concentrations of villin (villin/actin ∼ 1:200) but is stimulated at higher concentrations (villin/actin ∼ 1:50). None of the above effects is observed at 〈 10-8 M free [Ca2+].

    Notes: The 21 S complex of enzymes for DNA synthesis in the combined low salt nuclear extract-post microsomal supernatant from HeLa cells [Malkas et al. (1990) Biochemistry 29:6362-6374] was purified by poly (ethylene glycol) precipitation, Q-Sepharose chromatography, Mono Q Fast Protein Liquid Chromatography (FPLC), and velocity gradient centrifugation. The procedure gives purified enzyme complex at a yield of 45%. The 21 S enzyme complex remains intact and functional in the replication of simian virus 40 DNA throughout the purification. Sedimentation analysis showed that the 21 S enzyme complex exists in the crude HeLa cell extract and that simian virus 40 in vitro DNA replication activity in the cell extract resides exclusively with the 21 S complex. The results of enzyme and immunological analysis indicate that DNA polymerase α-primase, a 3′,5′ exonuclease, DNA ligase I, RNase H, and topoisomerase I are associated with the purified enzyme complex. Denaturing polyacrylamide gel electrophoresis of the purified enzyme complex showed the presence of about 30 polypeptides in the size range of 300 to 15 kDa. Immunofluorescent imaging analysis, with antibodies to DNA polymerase α,β and DNA ligase I, showed that polymerase α and DNA ligase I are localized to granular-like foci within the nucleus during S-phase. In contrast, DNA polymerase β, which is not associated with the 21 S complex, is diffusely distributed throughout the nucleoplasm.

    Description: Contact inhibition of locomotion (CIL), the repulsive response of cells upon cell–cell contact, has been the predominant paradigm for contact-mediated responses. However, it is difficult for CIL alone to account for the complex behavior of cells within a multicellular environment, where cells often migrate in cohorts such as sheets, clusters, and streams. Although cell–cell adhesion and mechanical interactions play a role, how individual cells coordinate their migration within a multicellular environment remains unclear. Using micropatterned substrates to guide cell migration and manipulate cell–cell contact, we show that contacts between different regions of cells elicit different responses. Repulsive responses were limited to interaction with the head of a migrating cell, while contact with the tail of a neighboring cell promoted migration toward the tail. The latter behavior, termed contact following of locomotion (CFL), required the Wnt signaling pathway. Inhibition of the Wnt pathway disrupted not only CFL but also collective migration of epithelial cells, without affecting the migration of individual cells. In contrast, inhibition of myosin II with blebbistatin disrupted the migration of both individual epithelial cells and collectives. We propose that CFL, in conjunction with CIL, plays a major role in guiding and coordinating cell migration within a multicellular environment.

    Description: Abstract Tin fluorophosphate (TFP) glass, which can be used to manufacture a phosphor‐in‐glass (PiG) for achieving high‐power white light‐emitting diodes (w‐LEDs), has attracted a great deal of attention because of its low‐melting point. Mn2+‐doped ultralow glass transition temperature (~122°C) Sn–F–P–O glasses were prepared to achieve broadband visible light emission from 390 to 720 nm. By controlling the concentration of MnO, the emission color of the TFP glass can be adjusted from blue/cool white to warm white/red. In particular, 0.2 mol% MnO‐doped TFP glass, which yields bright and warm white light and has ultralow glass transition temperature and thermal stability, has a promising application prospect in the field of high‐power w‐LEDs.

    Description: Incremental ecological service value is an important assessment indicator for ecosystem restoration projects, such as the interconnected river system network project in western Jilin Province, China. In this paper, a fuzzy two-stage chance-constrained stochastic programming (FTSCCP) model is proposed for the optimization of regional incremental ecological service value through adjustments to the original water diversions and ecological water supplement schemes for ecosystem restoration under different management requirements and scenarios. The proposed method can effectively address uncertainties such as fuzzy parameters, probability distributions, and random variables in optimizing processes simultaneously. The results reveal that, under the requirement of meeting the original plan, a decrease in restoration scale would mainly appear under low-flow scenarios, and for the other scenarios, the original plan would be fully realized with an increase in flood utilization and a decrease in normal water supplementation. For the requirement of maximizing the incremental ecological service value, the adjustments would mainly be increases in water supplementation for reed wetlands and marsh wetlands and decreases in water surface area, including fish ponds and crab ponds. The ecological service values would be 11,256.79 million CNY (Chinese Yuan), 16,465.11 million CNY and 17,639.59 million CNY under low-, normal- and high-flow scenarios, respectively, with increases of 3.2%, 6.9% and 14.5% compared to the values based on the requirement of meeting the original plan. The results would be helpful for guiding regional ecosystem restoration and water resource management in the western Jilin Province.

    Description: Restoring natural wetlands with conservation projects is an urgent task for human well-being. This paper introduces the Interval linear programming (ILP) method in wetland restoration projects for the first time and builds an optimization model. The purpose of the optimization model is to find an optimal restoration measures allocation pattern that can minimize the total investment in wetland restoration projects and obtain additional ecological environment and socio-economic benefits. The optimization model can also decrease the influence of interval uncertainty in the system by expressing the executed solution as interval numbers with an upper bound and a lower bound. The result of the optimization model for the wetland restoration project indicated a range of 6.84%–15.43% reduction on comparison with the original scheme which verified the effectiveness and validity of this optimization model. Our findings indicate that higher ecological and social benefits of wetland restoration projects can be achieved with lower restoration investment on the application of the reasonable and optimal restoration measures allocation pattern by the optimization model. The results of interval solutions can provide guidance for project managers to select a satisfactory decision-making plan by adjusting the decision variables in the interval solutions according to the practical situation. It can be seen that reeds were suggested to be planted over 46.75 km2, with the same lower bound and higher bound. Meanwhile, populus euphratica, and dryland willow were recommended to be planted in a mixed forest pattern within the interval of 30.54 km2 to 37.25 km2, and so forth. With the optimal solutions obtained from the model, the total project investment would be in the range of 2193.14 (104 CNY) to 2416.01 (104 CNY). Future improvements of our optimization model in wetland restoration projects should consider other kinds of uncertainties in the system such as stochastic uncertainties, fuzzy uncertainties, and integrated uncertainties.
