    Description: Annual precipitation for the last 2,500 years was reconstructed for northeastern Qinghai from living and archaeological juniper trees. A dominant feature of the precipitation of this area is a high degree of variability in mean rainfall at annual, decadal, and centennial scales, with many wet and dry periods that are corroborated by other paleoclimatic indicators. Reconstructed values of annual precipitation vary mostly from 100 to 300 mm and thus are no different from the modern instrumental record in Dulan. However, relatively dry years with below-average precipitation occurred more frequently in the past than in the present. Periods of relatively dry years occurred during 74-25 BC, AD 51-375, 426-500, 526-575, 626-700, 1100-1225, 1251-1325, 1451-1525, 1651-1750 and 1801-1825. Periods with a relatively wet climate occurred during AD 376-425, 576-625, 951-1050, 1351-1375, 1551-1600 and the present. This variability is probably related to latitudinal positions of winter frontal storms. Another key feature of precipitation in this area is an apparently direct relationship between interannual variability in rainfall with temperature, whereby increased warming in the future might lead to increased flooding and droughts. Such increased climatic variability might then impact human societies of the area, much as the climate has done for the past 2,500 years.

    Description: The development of bottom simulating reflectors (BSRs) and the gas hydrate stability zone (GHSZ) in continental margins is related to recent depositional and tectonic processes. This interrelation is important for understanding the potential resource of gas hydrate deposits. The purpose of this study is to understand the influence of such activity on the marine gas hydrate system of the seaward extension of the Trujillo Basin, Peru Margin. Here, we analyze near-seafloor heat flow probe data, high-resolution multichannel seismic (MCS) profiles, swath bathymetry and ocean floor observation system (OFOS) images. Based on our results, we identify three main physiographical subregions in the study area: (1) an area with turbidites, continuous BSRs and low-to-moderate near-seafloor heat flow (7-33 mW/m**2), (2) an area with sediment waves of turbidity origin, a mix of continuous and patchy BSRs, and moderate near-seafloor heat flow (26-39 mW/m**2), (3) an area with extensional faulting, patchy BSRs and moderate-to-high near-seafloor heat flow (52-110 mW/m**2). We conclude that sediment flow processes are the result of recent depositional controls, and faulting is the result of recent tectonic activity. Near-seafloor heat flow around chemoherms is moderate. Estimates of BSR-derived heat flow show differences to near-seafloor heat flow. This difference points to advection of fluids occurring at the seafloor. Alive Calyptogena and tube worms were observed in these venting zones. Based on our analysis, we conclude that: (1) recent depositional processes control the development of continuous BSRs and may restrict heat transfer through the GHSZ in the form of fluid venting at the seafloor, (2) recent tectonic processes control the development of patchy BSRs and allow heat transfer through the GHSZ along faults, which is manifested as fluid venting at the seafloor.

    Notes: Abstract The structures of bacterial communities were studied in activated sludge samples obtained from the aerobic and anaerobic zones of a wastewater treatment plant showing enhanced phosphorous removal. Samples were analyzed by in situ hybridization with oligonucleotide probes complementary to selected regions of the 16S and 23S ribosomal RNA (rRNA) characteristic for defined phylogenetic entities (genera and larger groups). The microbial community structures revealed by molecular techniques were compared with the compositions of culturable bacterial communities, obtained from the characterization of 255 isolates from tryptone-soy (TS) agar and R2A agar. These isolates were characterized by 89 physiological tests and their cellular fatty acid patterns, and identified. Culture-dependent techniques indicated the following distribution: different Aeromonas spp. (2.7–8.3% on R2A agar; 45.0–63.7% on TS agar), Acinetobacter spp. (5.4–9.0% on R2A agar; 5.0–9.1% on TS agar), Pseudomonas spp. (up to 10% on R2A agar) and Shewanella putrefaciens (up to 3.0% on R2A agar), all members of the gamma subclass of Proteobacteria, were isolated most frequently. The relatively rare isolates of the beta subclass were identified as Acidovorax spp., Alcaligenes spp., and Comamonas spp., The Gram-positive bacteria (high DNA G+C) were assigned mainly to Arthrobacter spp., Microbacterium spp., and Mycobacterium phlei. In order to assess the in situ abundance of the most frequently isolated genus, Aeromonas, two rRNA-targeted oligonucleotide probes were developed. The two gamma proteobacterial genera Aeromonas and Acinetobacter constituted less than 5% of all bacteria. In situ, Proteobacteria belonging to the beta subclass and high G+C Gram-positive bacteria were dominant. From filamentous bacteria, Sphaerotilus spp. and Leptothrix spp. could be detected occasionally. In addition, one sample contained a high proportion of the morphologically distinct filaments of Microthrix parvicella. As for the genus Acinetobacter, the relative abundance of the most frequently gamma-proteobacterial genus Aeromonas was overestimated by the intrinsic selectivity of cultivation. Cultivation on nutrient-rich medium (TS-agar) especially supported an enhanced isolation of bacteria belonging to these two genera.

    Notes: Abstract Hereditary multiple exostoses (EXT) is a genetically heterogeneous bone disorder caused by genes segregating on human chromosomes 8, 11, and 19 and designated EXT1, EXT2 and EXT3, respectively. Recently, the EXT1 gene has been isolated and partially characterized and appears to encode a tumor suppressor gene. We have identified six mutations in the human EXT1 gene from six unrelated multiple exostoses families segregating for the EXT gene on chromosome 8. One of the mutations we detected is the same 1-bp deletion in exon 6 that was previously reported in two independent EXT families. The other five mutations, in exons 1, 6, 9, and the splice junction at the 3′ end of exon 2, are novel. In each case, the mutation is likely to result in a truncated or nonfunctional EXT1 protein. These results corroborate and extend the previous report of mutations in this gene in two EXT families, and provide additional support for the EXT1 gene as the cause of hereditary multiple exostoses in families showing linkage to chromosome 8.
