Zusammenfassung Die Unzulänglichkeiten des traditionell verwendeten Schwefelsäure/Wasserstoffperoxid-Gemisches bei der Mineralisation des Reststickstoffes im Serum werden mit einem anderen Digestionsmittel, der Perchlorsäure verglichen. In ihr wurde ein Reagens von hervorragenden Eigenschaften gefunden, das eine schnelle, einfache und quantitative Digestion auch komplizierter Stoffe ermöglicht. So kann man quantitativ Salze der ÄDTA mineralisieren, die ein verzweigtes Molekül mit hohem Molekulargewicht besitzt. Die Vorteile sind folgende: Man kann bei verhältnismäßig niedrigen Temperaturen arbeiten; die Ergebnisse werden nicht von der Inkonstanz der Badtemperatur beeinflußt; die Gefahr des Überhitzens der Analysenproben ist ausgeschlossen; im Zeitintervall von 5–20 min bleibt der Stickstoffgehalt unverändert; das Reagens muß nicht quantitativ beseitigt werden. Die Fehlergrenze bei dieser Methode liegt im Konzentrationsbereich von 25–200 mg N/100 ml bei ungefähr 5%. Bei geeigneter Wahl von Konzentration und Verdünnung bietet Perchlorsäure keine größere Gefahr als andere Digestionsmittel.

Abstract The unsatisfying results of the mostly used mineralisation mixture, sulphuric acid and hydrogen peroxide, are compared with the digestion reagent perchloric acid. This reagent has very good properties, and makes possible a simple, quick digestion of the most complicated compounds. Thus, it is possible to mineralise high concentrations of EDTA salts. The advantages of this kind of digestion are: It is possible to perform the examination at relatively low temperatures. The results are not influenced by the inconstancy of the bath. There is no danger of over-heating samples; in the range of 5–20 min the content of nitrogen does not change. It is not necessary to remove the digestion reagent, because oxidation capacity is lost by digestion. A very simple technique and great quickness of the determination is offered. The method was succesfully applied in the determination of non-protein nitrogen in serum with very satisfying results, the average deviation being 5% within the concentration range of 25–200 mg N/100 ml. Using suitable relations and dilutions the danger to be expected is not greater than with other digestion mixtures.
