    Description: Precise dissection of cells with ultrashort laser pulses requires a clear understanding of how the onset and extent of ablation (i.e., the removal of material) depends on pulse energy. We carried out a systematic study of the energy dependence of the plasma-mediated ablation of fluorescently-labeled subcellular structures in the cytoskeleton and nuclei of fixed endothelial cells using femtosecond, near-infrared laser pulses focused through a high-numerical aperture objective lens (1.4 NA). We find that the energy threshold for photobleaching lies between 0.9 and 1.7 nJ. By comparing the changes in fluorescence with the actual material loss determined by electron microscopy, we find that the threshold for true material ablation is about 20% higher than the photobleaching threshold. This information makes it possible to use the fluorescence to determine the onset of true material ablation without resorting to electron microscopy. We confirm the precision of this technique by severing a single microtubule without disrupting the neighboring microtubules, less than 1 micrometer away. c2005 Optical Society of America.

    Description: The approach used in establishing the predicted aerodynamic uncertainties and the process used in applying these uncertainties during the design of the Orbiter flight control system and the entry trajectories are presented. The flight test program that was designed to verify the stability and control derivatives with a minimum of test flights is presented and a comparison of preflight predictions with preliminary flight test results is made. It is concluded that the approach used for the Orbiter is applicable to future programs where testing is limited due to time constraints or funding.

    Description: The major aerodynamic design challenge at the beginning of the United States Space Transportation System (STS) research and development phase was to design a vehicle that would fly as a spacecraft during early entry and as an aircraft during the final phase of entry. The design was further complicated because the envisioned vehicle was statically unstable during a portion of the aircraft mode of operation. The second challenge was the development of preflight aerodynamic predictions with an accuracy consistent with conducting a manned flight on the initial orbital flight. A brief history of the early contractual studies is presented highlighting the technical results and management decisions influencing the aerodynamic challenges. The configuration evolution and the development of preflight aerodynamic predictions will be reviewed. The results from the first four test flights shows excellent agreement with the preflight aerodynamic predictions over the majority of the flight regimes. The only regimes showing significant disagreement is confined primarily to early entry, where prediction of the basic vehicle trim and the influence of the reaction control system jets on the flow field were found to be deficient. Postflight results are analyzed to explain these prediction deficiencies.

    Description: The present conference gives attention to topics in the application of fracture mechanics, subcritical crack growth phenomena, fracture testing methods, ductile fracture behavior, and fracture mechanisms and their analysis. Specific papers treat the resistance curve approach to composite materials characterization, fracture toughness in ductile iron and cast steel, hold-time effects in elevated temperature fatigue crack propagation, creep crack growth under nonsteady conditions, viscoplastic fatigue in a superalloy at elevated temperatures, fracture testing with arc bend specimens, one-point bend impact test application, and a compact mode II fracture specimen. Also discussed are the computation of stable crack growth using the J-integral, the use of plastic energy dissipation to characterize crack growth, the extension of surface cracks under cyclic loading, the minimum time criterion for crack instability in structural materials, dynamic crack propagation and branching under biaxial loading, and boundary layer effects in cracked bodies.

    Description: The Shuttle Program development schedule and the management decision to perform an orbital, manned mission on the first launch resulted in a requirement to develop realistic aerodynamic uncertainties for the preflight aerodynamic predictions. This paper addresses the methodology in developing two types of aerodynamic uncertainties. One involves the ability to reproduce aerodynamic results between various wind tunnel tests. The second addresses the difference between preflight aerodynamic predictions and flight results derived from analysis of past aircraft programs. Both types of uncertainties for pitching moment, lateral-directional stability, rudder power, and aileron power are presented. In addition, the application of uncertainties to flight control design and fight test planning is briefly reviewed.

    Description: The investigation was conducted in the NASA/Ames Research Center 12-foot Pressure Tunnel. The model was a Langley-built 0.015-scale SSV orbiter model with remote independently operated left and right elevon surfaces. The objective of the test was to generate a detailed aerodynamic data base for the current shuttle orbiter configuration. Special attention was directed to definition of nonlinear aerodynamic characteristics by taking data at small increments in angle of attack, angle of sideslip, and elevon position. Six-component aerodynamic force and moment and elevon position data were recorded over an angle of attack range from -4 deg to 24 deg at angles of sideslip of 0 deg and + or - 4 deg. Additional tests were made over an angle of sideslip range from -6 deg to 6 deg at selected angles of attack. The test Mach numbers were 0.22 and 0.29 and the Reynolds number was varied from 2.0 to 8.5 million per foot.

    Description: Soft X ray flare events were observed with the S-056 X-ray telescope that was part of the ATM complement of instruments aboard SKYLAB. Analyses of these data are reported. The observations are summarized and a detailed discussion of the X-ray flare structures is presented. The data indicated that soft X-ray emitted by a flare come primarily from an intense well-defined core surrounded by a region of fainter, more diffuse emission. An analysis of flare evolution indicates evidence for preliminary heating and energy release prior to the main phase of the flare. Core features are found to be remarkably stable and retain their shape throughout a flare. Most changes in the overall configuration seem to be result of the appearance, disappearance or change in brightness of individual features, rather than the restructuring or reorientation of these features. Brief comparisons with several theories are presented.
