    Description: The ongoing increase of CO2 in the atmosphere is inducing a progressive lowering of marine water pH that is predicted to decrease to 7.8 by the end of this century. In marine environment, physical perturbation may affect reproduction, which is crucial for species' survival and strictly depends on gamete quality. The effects of seawater acidification (SWAc) on gamete quality of broadcast spawning marine invertebrates result largely from experiments of gamete exposure while the SWAc impact in response to adult exposure is poorly investigated. Performing microcosm and in field experiments at a naturally acidified site, we investigated the effects of adult SWAc exposure on sperm quality parameters underlying fertilization in Mytilus galloprovincialis. These animals were exposed to pH 7.8 over 21 days and collected at different times to analyze sperm parameters as concentration, motility, viability, morphology, oxidative status, intra- and extra-cellular pH and mitochondrial membrane potential. Results obtained in the two experimental approaches were slightly different. Under field conditions, we found an increase in total sperm motility and mitochondrial membrane potential on days 7 and 14 from the start of SWAc exposure whereas, in microcosm, SWAc group showed an increase of total motility on day 14. In addition, sperm morphology and intracellular pH were affected in both experimental approaches; whereas oxidative stress was detected only in spermatozoa collected from mussels under natural SWAc. The overall analysis suggests that, in mussels, SWAc toxic mechanism in spermatozoa does not involve oxidative stress. This study represents the first report on mussel sperm quality impairment after adult SWAc exposure, which may affect fertilization success with negative ecological and economic consequences; it also indicates that, although naturally acidified areas represent ideal natural laboratories for investigating the impact of ocean acidification, microcosm experiments are necessary for examining action mechanisms.

    Notes: Abstract Two kinds of surveys on marine environment quality were carried out along the Neapolitan Bay from the winter 1977 to 1980. The first was a general investigation along 60 km of coast, embracing several municipalities. Its purpose was to identify the most consistently polluted areas. The second one was a more limited but prolonged monitoring along about 12 km of coast entirely included in the municipality of Naples. At the beginning of this latter inspection two fecally contaiminated areas were found. These showed an increase in pollution during summer. However during the three years of surveillance a general improvement of the water quality was observed, perhaps due to the use of undersea pipes for the discharge of sewage far from the coast. As a consequence in the summer of 1980, 78% of the sampling points exihibited values ofEscherichia coli below limits prescribed in the Italian standard for bathing water (100E. coli/100 ml), while only 38% of the stations in the summer of 1978 could be considered suitable for bathing.

    Notes: Abstract In the summer of 1980, 78 sediment samples were collected in the gulf of Naples. Samples were analyzed for total and fecal coliforms, fecal streptococci, sulfite reducer clostridia and total plate count. Over almost all of the area surveyed fecal pollution was widespread, but different indicators exhibited different distribution patterns. The presence of fecal streptococci was found prevalent with the respect to that of fecal coliforms.

    Notes: Abstract In the summer of 1982 a microbiological monitoring of marine sediments was conducted in the southern part of the gulf of Naples to assess fecal contamination. Ninty nine sediment samples were analyzed for coliforms, fecal streptococci, sulfite reducers and total plate counts. The southern part of the gulf was less polluted than other parts of the bay following the coliforms patterns. On the contrary different consideration could be drawn following the distribution of enterococci and clostridia: according to these two microbial parameters this area could seem more polluted than other zones of the gulf. Possible explanations are presented.

    Notes: Abstract This paper compares three different methods to determine faecal pollution in shellfish. Two of these are related to MPN (Most Probable Number) technique and the last one is a direct count. The different results which may be obtained using these different techniques are shown. The relevance that introduction of the intervalve water may have on the bacterial titres in shellfish is presented. The shellfish utilized in this work came from two locations situated in the same bay. Simultaneous analyses on flesh, intervalve water and shellfish growing water show the different microbial titre that these three elements may have at the same time.

    Notes: In this report we show that the first event of activation in the human oocyte, the Fertilization current (FC), is a slow transient outward current of 300 pA, which induces a gradual hyperpolarization of the plasma membrane from -20mV to -60mV, 60-120 min after insemination, followed by a repolarization to -20mV. Activation currents (AC) of 600-2,500 pA, induced by exposure to the calcium ionophore A23187 or by microinjection of InsP3 into the cytosol, are also outward. The AC are inhibited by preloading oocytes with EGTA suggesting they are calcium dependent. Since AC are 2-10-fold the amplitude of the FC the fertilizing spermatozoon in the human only activates a portion of the primary elements stored in the oocyte for triggering metabolic depression. Oocyte activation in the human resembles that in the hamster rather than other mammals or invertebrates studied to date. © 1994 Wiley-Liss, Inc.

    Notes: Using a specific protein binding assay we have shown that a spermatozoon of the ascidian Ciona intestinalis contains 1.58 ± 0.74 × 10-19 moles of inositol 1,4,5-tri-phosphate (InsP3), while a human spermatozoon contains 6.4 ± 0.14 × 10-19 moles. Induction of the acrosome reaction (AR) in both species, by exposure to the calcium ionophore A23187, does not significantly alter levels of InsP3, suggesting that phosphatidylinositol (PI) turnover is not necessary for the calcium ionophore induced AR. Furthermore, PI turnover in ascidian spermatozoa appears to be insensitive to lithium and phorbol ester. The high intracellular concentration of InsP3 in spermatozoa, corresponding to 50-200 μM, suggests it may play a role in egg activation. © 1993 Wiley-Liss, Inc.

    Notes: Neomycin, injected into ascidian oocytes to a final concentration of 10-50 mM, inhibits both the fertilization current and the surface contraction, showing that phosphoinositide hydrolysis is required for these early activation events. Sperm-activated fertilization currents are not inhibited in the presence of 100 μg/ml intracellular heparin, suggesting that these currents are not directly gated by InsP3. The sulfhydryl reagent thimerosal at 100 μM, in contrast, significantly increases the fertilization current presumably by sensitizing the channel receptor. Since heparin inhibits the surface contraction, InsP3 receptors are shown to play a role in the propagation of the activation response in ascidian oocyte. Depleting intracellular calcium stores by microinjecting 50 mM EGTA into oocytes does not activate fertilization channels; however, subsequent fertilization of these EGTA loaded oocytes leads to a significantly larger and faster fertilization current. Thus in contrast to somatic cells studied to date, second messenger operated plasma membrane channels in ascidian oocytes are not gated by calcium released from intracellular stores. © 1994 Wiley-Liss, Inc.

    Notes: In this paper we report on the mechanism of action of bonellin on sea urchin eggs. Bonellin acts in the same way as porphyrins, ie, by causing the crosslinking of membrane proteins and the peroxocompounds formation. A possible role of membrane organization related to cleavage formation is discussed.
