2-Methoxy-and 2-(Methylthio)pyrimidinesO-Methylisourea hydrochloride (1a) reacts with the β-diketones 2a and b in boiling ethanol to yield the 2-hydroxypyrimidines 3a and b under exchange of the methoxy group. However, the methoxy group is retained in the synthesis of the pyrimidines 5a and b accomplished in an alkaline medium at room temperature. Reaction of S-methylisothiourea sulfate (6) with 2a and c under the same conditions leads to the formation of the 2-(methylthio)pyrimidines 7a and b. In the synthesis of the pyrimidines 10a and b starting from the β-diketone 2d the carbinol amines 9a and b are isolated as intermediates.

Bei der Umsetzung von O-Methylisoharnstoff-hydrochlorid (1a) mit den β-Diketonen 2a und b in siedendem äthanol bilden sich unter Austausch der Methoxygruppe die 2-Hydroxypyrimidine 3a und b. Hingegen bleibt die Methoxygruppe erhalten bei der zu den Pyrimidinen 5a und b führenden Kondensation in alkalischem Medium bei Raumtemperatur. Unter diesen Reaktions-bedingungen werden auch bei der Umsetzung des S-Methylisothioharnstoff-sulfats (6) mit den β-Diketonen 2a und c die 2-(Methylthio)pyrimidine 7a und b erhalten. Mit dem β-Diketon 2d lassen sich auf dem zu den Pyrimidinen 10a und b führenden Wege die Carbinolamine 9a und b als Zwischenprodukte isolieren.
