Zusammenfassung In den Extremitäten und Köpfen von Rattenembryonen des Tages 16–20 wurden die Aktivität und die Lokalisation einer Phosphatase untersucht. Die Enzyme aus Köpfen und Extremitäten sind gleich und wurden als unspezifische alkalische Phosphatase charakterisiert. Das Enzym kommt partikelgebunden im Sediment und in löslicher Form im 30000 g-Überstand vor. Die Phosphataseaktivität findet sich als Bleiphosphatniederschlag intrazellulär in Vesikeln und Granula der Osteoblasten der desmalen, perichondralen und chondralen Osteogenese. Nach seiner Sekretion läßt sich das Enzym extrazellulär in der Interzellularsubstanz des verkalkenden Vorknorpels und im Osteoid während der Osteogenese nachweisen. Es besteht eine enge räumliche Beziehung zwischen Bleiphosphatniederschlägen und den ersten Apatit-Einlagerungen.

Summary From days 16 to 20 we examined the activity and localisation of a phosphatase in the heads and extremities of rat foetuses. The enzymes from heads and extremities are identical and were characterized as an unspecific alkaline phosphatase. This enzyme could be found particle-bound in the sediment as well as in the 30000 g-supernatant. The phosphatase-activity was found as intracellular lead phosphate deposits in the vesicles and granules of the osteoblasts of the desmal, perichondrial and intercartilaginous osteogenesis. After secretion the enzyme could be found extracellularly in the intercellular matrix of the calcifying precartilage and in the osteoid during osteogenesis. There is a close spatial relationship between lead phosphate deposits and the first apatite depositions.
