    Notes: Value of information (VOI)analytic techniques are used to evaluate the benefit of performing animal bioassays to provide information about the cancer potency of specific chemical compounds. These tools allow the identification of the conditions in which the cost of reducing uncertainty about potency, by performing a subchronic or chronic bioassay, is justified by the benefit of having improved information for making control decisions. The decision analytic results are readily scaled to apply to a range of human contact rates (exposures)and a variety of control strategies. The sensitivity of results to uncertainty about animal to human extrapolation and the design of the bioassay is explored. An evaluation of the possible gains in general understanding about the mechanisms of carcinogenicity resulting from chronic bioassays is beyond the scope of this approach.

    Notes: Abstract A gene encoding a type I topoisomerase (TOP1) was isolated from Candida albicans, sequenced, and expressed in Saccharomyces cerevisiae. The TOP1 gene was identified from a C. albicans genomic library by hybridization with the product of a polymerase chain reaction with degenerate primer sets encoding regions conserved in other TOP1 genes. A clone containing an open reading frame of 2463 bp and predicted to encode a protein of 778 amino acids with sequence similarity to eukaryotic type I topoisomerases was identified. The C. albicans TOP1 gene restored camptothecin sensitivity and increased the topoisomerase activity in S. cerevisiae, indicating that the DNA fragment encodes a functional C. albicans topoisomerase I.

    Notes: Abstract The electrical properties of unfertilized eggs of Fucus serratus L. were characterized using voltage clamp and current clamp with single electrodes. The plasma membrane of the unfertilized egg is excitable. Depolarizing the egg in current clamp induced a transient depolarizing voltage response, the amplitude of which was dependent on the presence of external Ca2+ or Ba2+ and was blocked by La3+. The repolarizing phase was blocked by tetraethylammonium ions. Repeated stimulation at frequencies greater than 0.5 Hz caused a transient loss of excitability. Voltage-clamp experiments revealed that an inward current with an activation threshold of -35 mV underlies the depolarizing phase of the voltage response. This current showed rapid activation and slow inactivation. The current was blocked by La3+ and could be carried by Ca2+ and Ba2+ but not by Sr2+ or Na+. Further depolarization to values more positive than-5 mV induced a slowly activating outward K+ current in addition to the inward current, which corresponded to the repolarizing phase of the voltage response. This K+ current showed little or no inactivation during stimulation and slow deactivation on return to the resting potential. Hyperpolarizing the egg elicited an inward current. On fertilization, the Fucus egg generates a depolarizing “fertilization potential”. Voltage-clamp experiments revealed an inward fertilization current underlying the fertilization potential. Within 15 min of fertilization a dramatic, irreversible increase in resting K+ permeability developed. The roles of the plasma-membrane channels in generation of the fertilization potential and egg activation are discussed.

    Notes: Summary A 1.3-kb segment of Escherichia coli DNA containing the regulatory gene, araC, and the promoter of the araBAD operon was amplified by the polymerase chain reaction (PCR) and cloned into pUC18, resulting in plasmid pKB130 that produced the α fragment of β-galactosidase upon addition of L-arabinose (L-ara). A synthetic gene for human immunodeficiency virus (HIV)-1 preprotease was placed downstream of the araBAD promoter in pKB130 to create a translational fusion inducible by addition of L-ara. The fusion protein correctly autoprocessed in vivo to yield a mature 99-amino-acid HIV-1 protease, which was found predominantly in inclusion bodies. This material could be refolded to an active form, which was purified to homogeneity. A small fraction of the protease was expressed in vivo as a soluble active form, which allowed the monitoring of expression during fermentation by a rapid and simple whole cell assay employing an HIV-1 protease-specific fluorogenic substrate.

    Description: Voltage-gated sodium channels underlie action potential generation in excitable tissue. To establish the evolutionary mechanisms that shaped the vertebrate sodium channel a-subunit (SCNA) gene family and their encoded Nav1 proteins, we identified all SCNA genes in several teleost species. Molecular cloning revealed that teleosts have eight SCNA genes, comparable to the number in another vertebrate lineage, mammals. Prior phylogenetic analyses had indicated that teleosts and tetrapods share four monophyletic groups of SCNA genes and that tandem duplications selectively expanded the number of genes in two of the four mammalian groups. However, the number of genes in each group varies between teleosts and tetrapods suggesting different evolutionary histories in the two vertebrate lineages. Our findings from phylogenetic analysis and chromosomal mapping of Danio rerio genes indicate that tandem duplications are an unlikely mechanism for generation of the extant teleost SCNA genes. Instead, analysis of other closely mapped genes in D. rerio supports the hypothesis that a whole genome duplication was involved in expansion of the SCNA gene family in teleosts. Interestingly, despite their different evolutionary histories, mRNA analyses demonstrated a conservation of expression patterns for SCNA orthologues in teleosts and tetrapods, suggesting functional conservation.
