    Description: This study was designed to elucidate details of the structure and formation process of the alternate lamellar pattern known to exist in lamellar bone. For this purpose, we examined basic internal lamellae in femurs of young rats by transmission and scanning electron microscopy, the latter employing two different macerations with NaOH at concentrations of 10 and 24%. Observations after the maceration with 10% NaOH showed that the regular and periodic rotation of collagen fibrils caused an alternation between two types of lamellae: one consisting of transversely and nearly transversely cut fibrils, and the other consisting of longitudinally and nearly longitudinally cut fibrils. This finding confirms the consistency of the twisted plywood model. The maceration method with 24% NaOH removed bone components other than cells, thus allowing for three-dimensional observations of osteoblast morphology. Osteoblasts extended finger-like processes paralleling the inner bone surface, and grouped in such a way that, within a group, the processes arranged in a similar direction. Transmission electron microscopy showed that newly deposited fibrils were arranged alongside these processes. For the formation of the alternating pattern, our findings suggest that: (1) osteoblasts control the collagen fibril arrangement through their finger-like process position; (2) osteoblasts behave similarly within a group; (3) osteoblasts move their processes synchronously and periodically to promote alternating different fibril orientation; and (4) this dynamic sequential deposition of fibrils results in the alternate lamellar (or twisted plywood) pattern.

    Description: Dissociation of an acid molecule in aqueous media is one of the most fundamental solvation processes but its details remain poorly understood at the distinct molecular level. Conducting high-pressure treatments of an open-cage fullerene C 70 derivative with hydrogen fluoride (HF) in the presence of H 2 O, we achieved an unprecedented encapsulation of H 2 O·HF and H 2 O. Restoration of the opening yielded the endohedral C 70 s, that is, (H 2 O·HF)@C 70 , H 2 O@C 70 , and HF@C 70 in macroscopic scales. Putting an H 2 O·HF complex into the fullerene cage was a crucial step, and it would proceed by the synergistic effects of "pushing from outside" and "pulling from inside." The structure of the H 2 O·HF was unambiguously determined by single crystal x-ray diffraction analysis. The nuclear magnetic resonance measurements revealed the formation of a hydrogen bond between the H 2 O and HF molecules without proton transfer even at 140°C.

    Description: New transport equations for chemical reaction rate and its mean value in turbulent flows have been derived and analyzed. Local perturbations of the reaction zone by turbulent eddies are shown to play a pivotal role even for weakly turbulent flows. The mean-reaction-rate transport equation is shown to involve two unclosed dominant terms and a joint closure relation for the sum of these two terms is developed. Obtained analytical results and, in particular, the closure relation are supported by processing two widely recognized sets of data obtained from earlier direct numerical simulations of statistically planar 1D premixed flames associated with both weak large-scale and intense small-scale turbulence.

    Description: A new multilayer-coated varied line-spaced grating, JS4000, was fabricated and tested for extending the upper limit of a grating X-ray spectrometer for electron microscopy. This grating was designed for 2–3.8 keV at a grazing incidence angle of 1.35°. It was revealed that this new multilayer structure enables us to take soft-X-ray emission spectra continuously from 1.5 to 4.3 keV at the same optical setting. The full-width at half maximum of Te-L α1,2 (3.8 keV) emission peak was 27 eV. This spectrometer was applied to indium tin oxide particles and clearly resolved Sn-L α (3444 eV) and In-L β1 (3487 eV) peaks, which could not be resolved by a widely used energy-dispersive X-ray spectrometer.

    Description: Aberrant transcriptional regulation in the brain is thought to be one of the key components of the pathogenesis and pathophysiology of neuropsychiatric disorders. Heat shock factors (HSFs) modulate cellular homeostasis through the control of gene expression. However, the roles of HSFs in brain function have yet to be elucidated fully. In the present study, we attempted to clarify the role of HSF1-mediated gene regulation in neuronal and behavioral development using HSF1-deficient (HSF1−/−) mice. We found granule neurons of aberrant morphology and impaired neurogenesis in the dentate gyrus of HSF1−/− mice. In addition, HSF1−/− mice showed aberrant affective behavior, including reduced anxiety and sociability but increased depression-like behavior and aggression. Furthermore, HSF1 deficiency enhanced behavioral vulnerability to repeated exposure to restraint stress. Importantly, rescuing the HSF1 deficiency in the neonatal but not the adult hippocampus reversed the aberrant anxiety and depression-like behaviors. These results indicate a crucial role for hippocampal HSF1 in neuronal and behavioral development. Analysis of the molecular mechanisms revealed that HSF1 directly modulates the expression of polysialyltransferase genes, which then modulate polysialic acid–neural cell adhesion molecule (PSA-NCAM) levels in the hippocampus. Enzymatic removal of PSA from the neonatal hippocampus resulted in aberrant behavior during adulthood, similar to that observed in HSF1−/− mice. Thus, these results suggest that one role of HSF1 is to control hippocampal PSA-NCAM levels through the transcriptional regulation of polysialyltransferases, a process that might be involved in neuronal and behavioral development in mice.

    Description: Optical pulses at THz and mid-infrared frequencies tuned to specific vibrational resonances modulate the lattice along chosen normal mode coordinates. In this way, solids can be switched between competing electronic phases and new states are created. Here, we use vibrational modulation to make electronic interactions (Hubbard-U) in Mott-insulator time dependent. Mid-infrared optical pulses excite localized molecular vibrations in ET-F2TCNQ, a prototypical one-dimensional Mott-insulator. A broadband ultrafast probe interrogates the resulting optical spectrum between THz and visible frequencies. A red-shifted charge-transfer resonance is observed, consistent with a time-averaged reduction of the electronic correlation strength U. Secondly, a sideband manifold inside of the Mott-gap appears, resulting from a periodically modulated U. The response is compared to computations based on a quantum-modulated dynamic Hubbard model. Heuristic fitting suggests asymmetric holon-doublon coupling to the molecules and that electron double-occupancies strongly squeeze the vibrational mode. Scientific Reports 4 doi: 10.1038/srep03823

    Description: Database obtained earlier in 3D Direct Numerical Simulations (DNS) of statistically stationary, 1D, planar turbulent flames characterized by three different density ratios σ is processed in order to investigate vorticity transformation in premixed combustion under conditions of moderately weak turbulence (rms turbulent velocity and laminar flame speed are roughly equal to one another). In cases H and M characterized by σ = 7.53 and 5.0, respectively, anisotropic generation of vorticity within the flame brush is reported. In order to study physical mechanisms that control this phenomenon, various terms in vorticity and enstrophy balance equations are analyzed, with both mean terms and terms conditioned on a particular value c of the combustion progress variable being addressed. Results indicate an important role played by baroclinic torque and dilatation in transformation of average vorticity and enstrophy within both flamelets and flame brush. Besides these widely recognized physical mechanisms, two other effects are documented. First, viscous stresses redistribute enstrophy within flamelets, but play a minor role in the balance of the mean enstrophy Ω ¯ within turbulent flame brush. Second, negative correlation u ′ · ∇ Ω ′ ¯ between fluctuations in velocity u and enstrophy gradient contributes substantially to an increase in the mean Ω ¯ within turbulent flame brush. This negative correlation is mainly controlled by the positive correlation between fluctuations in the enstrophy and dilatation and, therefore, dilatation fluctuations substantially reduce the damping effect of the mean dilatation on the vorticity and enstrophy fields. In case L characterized by σ = 2.5, these effects are weakly pronounced and Ω ¯ is reduced mainly due to viscosity. Under conditions of the present DNS, vortex stretching plays a minor role in the balance of vorticity and enstrophy within turbulent flame brush in all three cases.
