    Notes: Abstract A role for cytosolic free Ca2+ (Ca2+i) in the regulation of growth of Papaver rhoeas pollen tubes during the self-incompatibility response has recently been demonstrated [Franklin-Tong et al. Plant J. 4:163–177 (1993); Franklin-Tong et al. Plant J. 8:299–307 (1995); Franklin-Tong et al. submitted to Plant J.]. We have investigated the possibility that Ca2+i is more generally involved in the regulation of pollen tube growth using confocal laser scanning microscopy (CLSM). Data obtained using Ca2+ imaging, in conjunction with photolytic release of caged inositol 1,4,5-trisphosphate [Ins(1,4,5)P3], point to a central role of the phosphoinositide signal transduction pathway in the control of Ca2+ fluxes and control of pollen tube growth. These experiments further revealed that increases in cytosolic levels of Ins(1,4,5)P3 resulted in the formation of distinct Ca2+ waves. Experiments using the pharmacological agents heparin, neomycin and mastoparan further indicated that Ca2+ waves are propagated, at least in part, by Ins(1,4,5)P3-induced Ca2+ release rather than by simple diffusion or by “classic” Ca2+-induced Ca2+ release mechanisms. We also have data which suggest that Ca2+ waves and oscillations may be induced by photolytic release of caged Ca2+. Ratio-imaging has enabled us to identify an apical oscillating Ca2+ gradient in growing pollen tubes, which may regulate normal pollen tube growth. We also present evidence for the involvement of Ca2+ waves in mediating the self-incompatibility response. Our data suggest that changes in Ca2+i and alterations in growth rate/patterns are likely to be closely correlated and may be causally linked to events such as Ca2+-induced, or Ins(1,4,5)P3-induced wave formation and apical Ca2+ oscillations.

    Notes: Summary Analysis by gas chromatography revealed the presence of small amounts of squalene, but not lanosterol nor ergosterol in Pythium paroecandrum, P. ultimum, P. graminicola, and P. arrhenomonas. However, when acetate-14C was used as a precursor for sterols, even squalene was not found in P. graminicola. The deficiency in the sterol synthesizing mechanism may therefore be at or before the squalene forming step. Both squalene and ergosterol were present in the mycelium of Rhizoctonia solani, as shown by both gas chromatography and by the incorporation of acetate-14C into ergosterol. The absence of ergosterol in Pythium and its presence in Rhizoctonia is consistant with the resistance to the antibiotic filipin of Pythium species and the sensitivity of R. solani.

    Notes: Abstract Deposition of micrometer-scale tungsten, molybdenum and platinum lines with the aid of the uv light from an argon-ion laser was investigated. Tungsten and molybdenum were deposited from their corresponding carbonyls and platinum from an acetylacetonate compound. High-quality metal conductors could be achieved for tungsten and platinum using a combined photolytic and pyrolytic, or “hybrid”, deposition scheme. The resistivity for these two deposited metals was approximately twice the bulk value. For molybdenum the deposition process was fast and dominated by the pyrolysis; the corresponding line resistivity was slightly higher than for the two other metals.

    Notes: The paper reports a study into set-up time reduction and mistake proofing methods in a small company involved in the machining of precision components in small batches with high variety for the aerospace industry. The company has made some set-up reductions mainly using work study related methods and in one manufacturing cell by the use of the Single Minute Exchange of Die (SMED) methodology. Mistake proofing devices in the form of fouling pins and offset holes have been developed for the family of components manufactured in this cell. Until recently the set-up times were not measured and worse still were considered as productive hours. As a consequence, there was a lack of awareness and motivation amongst operational personnel to reduce set-up times and knowledge of SMED was limited to a small group of individuals. This, along with the lack of investment in mechanisms to aid set-up time reductions and prevent errors, has restricted the use of this type of methods and technology. However, there is evidence that the demands made by the company's major customer will lead to increased efforts to put into place these types of changes.

    Notes: The bitter component, limonin, was quantified in orange and grapefruit juices after preliminary separation on a C-18 cartridge, or by direct injection of the juice onto a C-18 or C-8 high performance liquid chromatographic (HPLC) column. The lower limit of detection for limonin was about 1 ppm on a 2.1 mm i.d. microbore column and 2 ppm on a 4.6 mm i.d. analytical column. In a few orange juice samples, the preliminary separation step was necessary to eliminate interfering substances prior to HPLC separation.

    Notes: Eighteen volatile compounds have been identified in a methylene chloride extract of a distilled fraction from loquat fruit. Individual compounds were identified by comparison of gas chromatographic retention times and mass spectra with those for authentic compounds. Major components phenyl ethyl alcohol, 3-hydroxy-2-butanone, phenylacetaldehyde and hexen-1-ols, and minor components, ethyl acetate, methyl cinnamate and β-ionone probably contribute to the fruity-floral flavor of the fruit.
