    Description: Prototype galectins, endogenously expressed animal lectins with a single carbohydrate recognition domain, are well-known regulators of tissue properties such as growth and adhesion. The earliest discovered and best studied of the prototype galectins is Galectin-1 (Gal-1). In the Gallus gallus (chicken) genome, Gal-1 is represented by two homologs: Gal-1A and Gal-1B, with distinct biochemical properties, tissue expression, and developmental functions. We investigated the origin of the Gal-1A/Gal-1B divergence to gain insight into when their developmental functions originated and how they could have contributed to vertebrate phenotypic evolution. Sequence alignment and phylogenetic tree construction showed that the Gal-1A/Gal-1B divergence can be traced back to the origin of the sauropsid lineage (consisting of extinct and extant reptiles and birds) although lineage-specific duplications also occurred in the amphibian and actinopterygian genomes. Gene synteny analysis showed that sauropsid gal-1b (the gene for Gal-1B) and its frog and actinopterygian gal-1 homologs share a similar chromosomal location, whereas sauropsid gal-1a has translocated to a new position. Surprisingly, we found that chicken Gal-1A, encoded by the translocated gal-1a , was more similar in its tertiary folding pattern than Gal-1B, encoded by the untranslocated gal-1b , to experimentally determined and predicted folds of nonsauropsid Gal-1s. This inference is consistent with our finding of a lower proportion of conserved residues in sauropsid Gal-1Bs, and evidence for positive selection of sauropsid gal-1b , but not gal-1a genes. We propose that the duplication and structural divergence of Gal-1B away from Gal-1A led to specialization in both expression and function in the sauropsid lineage.

    Description: Total Worker Health® (TWH) frameworks call for attention to organizational leadership in the implementation and effectiveness of TWH approaches. It is especially important to study this within in the small business environment where employees face significant health, safety, and well-being concerns and employers face barriers to addressing these concerns. The purpose of this study was to gain a better understanding of how small business leaders perceive employee health, safety, and well-being in the context of their own actions. We conducted semi-structured interviews with 18 small business senior leaders and used a qualitative coding approach to analyze the transcripts to determine the frequency with which leaders discussed each code. When we asked leaders about their leadership practices for health, safety, and well-being, leaders reflected upon their business (65%), themselves (28%), and their employees (7%). Leaders rarely discussed the ways in which they integrate health, safety, and well-being. The interviews demonstrate that small business leaders care about the health of their employees, but because of the perceived value to their business, not to employees or themselves. Thus, they may lack the knowledge and skills to be successful TWH leaders. The present study supports a need for continued small business TWH leadership research.

    Description: Without exogenous rock-derived nutrient sources, terrestrial ecosystems may eventually regress or reach a terminal steady state, but the degree to which exogenous nutrient sources buffer or slow to a theoretical terminal steady state remains unclear. We used strontium isotope ratios ( 87 Sr/ 86 Sr) as a tracer and measured 87 Sr/ 86 Sr values in aeolian dust, soils, and vegetation across a well-constrained three million year semi-arid substrate age gradient to determine: 1) whether the contribution of atmospheric sources of rock-derived nutrients to soil and vegetation pools varied with substrate age, and 2) to determine if the depth of uptake varied with substrate age. We found that aeolian-derived nutrients became increasingly important, contributing as much as 71% to plant-available soil pools and tree ( Pinus edulis ) growth during the latter stages of ecosystem development in a semi-arid climate. The depth of nutrient uptake increased on older substrates, demonstrating that trees in arid regions can acquire nutrients from greater depths as ecosystem development progresses presumably in response to nutrient depletion in the more weathered surface soils. Our results demonstrate that global and regional aeolian transport of nutrients to local ecosystems is a vital process for ecosystem development in arid regions. Furthermore, these aeolian nutrient inputs contribute to deep soil nutrient pools, which become increasingly important for maintaining plant productivity over long time scales.

    Description: Gastric neuroendocrine tumours (NETs) arise from enterochromaffin-like cells, which are located in oxyntic glands within the stomach. Type I tumours represent 70–80% of gastric NETs and are associated with hypergastrinaemia, chronic atrophic gastritis and achlorhydria. Gastrin is involved in the endocrine regulation of gastric acid production. Most type I gastric NETs are sporadic, have a good prognosis and their genetic basis are unknown. We performed an exome sequencing study in a family with consanguineous parents and 10 children, five of whom were affected by type I gastric NET. Atypical clinical traits included an earlier age of onset (around 30 years), aggressiveness (three had nodal infiltration requiring total gastrectomy and one an adenocarcinoma) and iron-deficiency rather than megaloblastic anaemia. We identified a homozygous missense mutation in the 14th exon of the ATP4A gene (c.2107C〉T), which encodes the proton pump responsible for acid secretion by gastric parietal cells. The amino acid p.Arg703Cys is highly conserved across species and originates a change of one of the transmembrane domains that avoids the liberation of protons from cells to stomach. This is consistent with the achlorhydria that was observed in the affected individuals. No germline or somatic mutations in the ATP4A gene were found in sporadic gastric NET patients. Based on the results of this large family, it seems that this atypical form of gastric NET has an earlier age of onset, behaves more aggressively and has atypical clinical traits that differentiated from other studied cases.

    Description: The radiative properties of mineral dust aerosol during the GERB Intercomparison of Long-wave and Short-wave (GERBILS) are presented. GERBILS consisted of aircraft flights over land areas between Mauritania and Niger during June 2007. During one case of large aerosol optical depth (AOD=1.0 at 0.55 μ m), a short-wave spectrometer measured sky radiances versus scattering angle that are compared to modelled data. The modelling used phase functions of spheres, spheroids and irregular-shaped particles using T-matrix and ray-tracing methods. Irregular particles provided the most satisfactory solution. In another case of full short-wave and long-wave radiative closure, measurements and modelled clear sky conditions allowed calculation of the direct radiative effect (DRE) at high and low level. The modelled AOD (0.92) required to simulate the measured spectral irradiances agrees with the aircraft AOD (0.79) within measurement uncertainty. The simulated irradiances are less sensitive to particle shape than radiances. However, it is shown through modelling of the surface and top-of-atmosphere (TOA) DRE over all daylight hours that significant differences exist at TOA due to variation in the asymmetry parameter. The TOA short-wave diurnally averaged DRE was modelled as between 0 and –20 W m −2 depending on particle shape. A long-wave interferometer measured downwelling and upwelling radiances to derive surface emissivity across the window region. Measured nadir brightness temperatures from high level show signature of dust. A drop in brightness temperature of 14K was determined using modelled pristine-sky spectra. The modelled outgoing long-wave DRE due to dust from this case was +14 W m −2 averaged over 24 h, or +17 W m −2 per unit AOD. Modelling studies illustrate the sensitivity to aerosol refractive index and size distribution for both short-wave and long-wave DREs. Considering the full spectrum, a refractive index dataset from the literature has been selected that best represents the Saharan dust encountered during GERBILS.     Description: Abstract Understanding how altered soil organic carbon (SOC) availability affects microbial communities and their function is imperative in predicting impacts of global change on soil carbon (C) storage and ecosystem function. However, the response of soil microbial communities and their function to depleted C availability in situ is unclear. We evaluated the role of soil C inputs in controlling microbial biomass, community composition, physiology, and function by (1) experimentally excluding plant C inputs in situ for 9 yr in four temperate forest ecosystems along a productivity gradient in Oregon, USA; and (2) integrating these findings with published data from similar C‐exclusion studies into a global meta‐analysis. Excluding plant C inputs for 9 yr resulted in a 13% decrease in SOC across the four Oregon sites and an overall shift in the microbial community composition, with a 45% decrease in the fungal : bacterial ratio and a 13% increase in Gram‐positive : Gram‐negative bacterial ratio. Although gross N mineralization decreased under C exclusion, decreases in gross N immobilization were greater, resulting in increased net N mineralization rates in all but the lowest‐productivity site. Microbial biomass showed a variable response to C exclusion that was method dependent; however, we detected a 29% decrease in C‐use efficiency across the sites, with greater declines occurring in less‐productive sites. Although extracellular enzyme activity increased with C exclusion, C exclusion resulted in a 31% decrease in microbial respiration across all sites. Our meta‐analyses of published data with similar C‐exclusion treatments were largely consistent with our experimental results, showing decreased SOC, fungal : bacterial ratios, and microbial respiration, and increased Gram‐positive : Gram‐negative bacterial ratio following exclusion of C inputs to soil. Effect sizes of SOC and respiration correlated negatively with the duration of C exclusion; however, there were immediate effects of C exclusion on microbial community composition and biomass that were unaltered by duration of treatment. Our field‐based experimental results and analyses demonstrate unequivocally the dominant control of C availability on soil microbial biomass, community composition, and function, and provide additional insight into the mechanisms for these effects in forest ecosystems.

    Description: Malaria, caused by Plasmodium sp, results in almost one million deaths and over 200 million new infections annually. The World Health Organization has recommended that artemisinin-based combination therapies be used for treatment of malaria. Artemisinin is a sesquiterpene lactone isolated from the plant Artemisia annua. However, the supply and price of artemisinin fluctuate greatly, and an alternative production method would be valuable to increase availability. We describe progress toward the goal of developing a supply of semisynthetic artemisinin based on production of the artemisinin precursor amorpha-4,11-diene by fermentation from engineered Saccharomyces cerevisiae, and its chemical conversion to dihydroartemisinic acid, which can be subsequently converted to artemisinin. Previous efforts to produce artemisinin precursors used S. cerevisiae S288C overexpressing selected genes of the mevalonate pathway [Ro et al. (2006) Nature 440:940–943]. We have now overexpressed every enzyme of the mevalonate pathway to ERG20 in S. cerevisiae CEN.PK2, and compared production to CEN.PK2 engineered identically to the previously engineered S288C strain. Overexpressing every enzyme of the mevalonate pathway doubled artemisinic acid production, however, amorpha-4,11-diene production was 10-fold higher than artemisinic acid. We therefore focused on amorpha-4,11-diene production. Development of fermentation processes for the reengineered CEN.PK2 amorpha-4,11-diene strain led to production of 〉 40 g/L product. A chemical process was developed to convert amorpha-4,11-diene to dihydroartemisinic acid, which could subsequently be converted to artemisinin. The strains and procedures described represent a complete process for production of semisynthetic artemisinin.

    Description: The wings of archaic Odonatoidea from the mid-Carboniferous of Argentina show features analogous to "smart" mechanisms in modern dragonflies that are associated with the agile, versatile flight necessary to catch prey in flight. These mechanisms act automatically in flight to depress the trailing edge and to facilitate wing twisting, in response to aerodynamic loading.     Description: Continuing degradation of coral reef ecosystems has generated substantial interest in how management can support reef resilience. Fishing is the primary source of diminished reef function globally, leading to widespread calls for additional marine reserves to recover fish biomass and restore key ecosystem functions. Yet there are no established baselines for determining when these conservation objectives have been met or whether alternative management strategies provide similar ecosystem benefits. Here we establish empirical conservation benchmarks and fish biomass recovery timelines against which coral reefs can be assessed and managed by studying the recovery potential of more than 800 coral reefs along an exploitation gradient. We show that resident reef fish biomass in the absence of fishing (B0) averages approximately 1,000 kg ha(-1), and that the vast majority (83%) of fished reefs are missing more than half their expected biomass, with severe consequences for key ecosystem functions such as predation. Given protection from fishing, reef fish biomass has the potential to recover within 35 years on average and less than 60 years when heavily depleted. Notably, alternative fisheries restrictions are largely (64%) successful at maintaining biomass above 50% of B0, sustaining key functions such as herbivory.
