    Notes: Abstract The various types of autoradiographic patterns occurring in salivary gland nuclei ofD. melanogaster following a short pulse of tritiated thymidine have been described and the probable order of their sequence within a polytenic replication cycle assigned. On the basis of that assignment, the distribution of those patterns in the nuclei of salivary glands of 160 staged larvae and prepupae has been assessed and the following interpretations made: Throughout the third larval instar and prepupal period there is a continuing decrease in frequency of labeled nuclei. Within the larval period that decrease is reflected in a proportional decrease in label patterns characteristic of both initiation and propagation phases of the replication cycle. At the beginning of the prepupal period there is a rise in the proportion of labeled cells displaying initiation type patterns followed by an abrupt cessation of such patterns. It is proposed that termination of initiation of polytenic replication is amongst the physiologic manifestations of the induction of pupation.

    Notes: Abstract The chromosomes of the mouse have been identified by specific banding patterns revealed by the Feulgen stain. Comparison of the patterns of the Feulgen-stained karyotype with those of acetic-saline-Giemsa stain and quinacrinemustard-fluorescence demonstrates a high order of similarity among the three, with the localization of Feulgen dense bands and regions closely paralleling that of Giemsa dark and fluorescence bright bands. Since the stained substrate of the Feulgen reaction is known to be DNA, it is suggested that all three banding methods reveal the distribution of DNA or of some moiety that closely follows DNA distribution in metaphase chromosomes. The preparative procedure of the Feulgen banding method consists of a 15 to 20 minute exposure to PO4 buffer at pH 10 and a prolonged (60–72 hrs) exposure to 12xSSC. Omission or curtailment of either step results in preparations with chromosome sets that are not karyotypable, although some stain differentiation is produced. HCl extraction prior to the preparative treatment blocks banding, but acid extraction following the preparative treatment, either that of the HCl hydrolysis of the Feulgen reaction of that of an almost fourfold extension of the standard hydrolysis time, does not obliterate bands already formed. By extrapolation from biochemical studies of chromatin, it is postulated that the localization of Feulgen dark and light stain, representing relative DNA densities, reflects the regional protein association of the DNA; the Feulgen dense regions may result from aggregation of a specific class of histones by the alkaline buffer with consequent condensation of the DNA bound to those histones; the Feulgen pale or negative regions may represent those in which non-aggregated proteins, histone and non-histone, have been solubilized in the saline incubation, rendering the DNA of those regions subject to diffusion or vulnerable to fragmentation in the Feulgen hydrolysis.

    Notes: Abstract This study describes the sequential alternation of compaction and decompaction in the chromosomes of the Chinese hamster oocyte from diakinesis to metaphase II. A series of micrographs show that the compact metaphase I chromosomes become greatly extended as they enter and pass through anaphase I. Once polarized, the presumptive oocyte chromosomes become exceedingly compact and form a tightly packed mass, each chromosome assuming contours to accomodate “dovetailing” with its neighbors, while the chromosomes consigned to the polar body remain extended and show signs of the incipient deterioration. Prior to ovulation, the chromosomes of the mass separate and begin to decompact, in part at least, by the previously postulated mechanism of uncoiling. Following ovulation, the chromosomes are greatly extended and, as the metaphase II complement, remain in that state until the advent of fertilization. — Evidence that the compaction patterns are ordered and chromosome specific is presented by observation of the two smallest chromosomes of the complement. At telophase I those chromosomes are markedly different in size and arm ratio; at metaphase II the differences are less pronounced and at mitotic metaphase the two smallest chromosome pairs are so similar in morphology as to be indistinguishable. It is proposed, therefore, that those two chromosomes differ in their fundamental morphology as revealed at the exceedingly compact state of telophase I oocyte chromosomes. Their subsequently established resemblance at mitotic metaphase may be due to allocycly on the part of one or both, resulting in two chromosomes of apparantly similar length and arm ratio.

    Notes: Summary The pattern of metaphase chromosome association in the human complement was studied by two methods of statistical analysis of interchromosomal distances. Those methods included ring analysis in which a characteristic position of the centromere of each chromosome relative to the center of a two-dimensional representation of a metaphase complement was defined, and estimation of the capacity for associativity of each of three regions of each chromosome: the centromere (c) and the ends of each arm (p,q). The following information was obtained: 1. In general, the distance from the center is directly related to chromosome size. 2. The most notable deviation from that size-related progression is displayed by the X chromosomes. The markedly peripheral position of the X is characteristic of both X's of the female and the single X of the male. 3. The relative associativity of each chromosome of the complement is, in general, inversely related to size with an additional preferential capacity of associativity displayed by the acrocentric chromosomes. Analyses of the different inter-regional classes established that the supplementary associativity factor of the acrocentric chromosomes was inherent in their pericentromeric and p-arm regions and excluded the ends of the q arms from participation in that factor. 4. Those analyses demonstrated that the specific morphology or ‘geometry’ of the acrocentric chromosomes contributes little to their high relative associativity. In addition to the tendency for the c/p regions of the acrocentric chromosomes to associate with each other, presumably because of their common function in nucleolar organization, those regions also displayed a propensity to associate with the distal regions of the arms of other chromosomes. A molecular basis for that propensity other than that of ribosomal DNA is postulated to be that of other fractions of highly reiterated DNA sequences. 5. Analysis of the relative associativities of each of the three regions of the Y chromosome revealed that the Yq displays a much stronger capacity to associate with the c's of other chromosomes than does the Yc or Yp.

    Notes: The basic nuclear proteins of a fraction of elongating spermatids from human tests and of a fraction of motile spermatozoa from the ejaculate, separated by ion-exchange chromatography, were compared. Analysis by acetic acid-urea polyacrylamide gel electrophoresis (PAGE) showed that, in both fractions, four proteins of lower mobility were coeluted with protamine 1 by 23% guanidinium chloride (GuCI) while protamine 2 alone was eluted by 50% GuCI. Treatment with alkaline phosphatase identified those four proteins as phosphorylated protamines, and cyanogen bromide (CNBr) treatment of the dephosphorylated protamines distinguished them as variants of protamine 2 and not of protamine 1. Thus far, phosphorylated forms of protamine 1 have not been detected in either spermatids or spermatozoa. Those observations indicate that protamine 2 functions in the cycle of phosphorylation-dephosphorylation, which is essential to the process of sperm chromatin condensation, while the role of protamine 1 in human spermiogenesis is not yet defined.The presence of phosphorylated protamine in motile, presumably mature spermatozoa appears to be characteristic of human sperm but not of the sperm of other mammals and is probably the basis for the heterogeneity of chromatin condensation frequently observed in human spermatozoa.

    Notes: Previous studies have demonstrated that a consistent maximum number of polytenic replication cycles occurs in the salivary gland nuclei of a wild-type strain of D. melanogaster. Since that number is achieved within the larval period, the DNA synthesis of the prepupal period is believed to be that of propagation of the final cycle. Photometric determinations have been made, in this study, of the salivary gland nuclei of larvae and prepupae of the tu-h strain in which the larval period has been extended as a consequence of delay or failure of pupation. The DNA values indicate that a higher maximum number of polytenic replications is achieved in such nuclei. It is inferred, thereby, that initiation of polytenic replication is a function of the larval state and, since it is terminated by the intervention of metamorphosis, a hormonal dependence is suggested.

    Notes: A comparison has been made, by Feulgen photometry, of the polytene nuclei of the salivary glands of a wild-type strain of Drosophila melanogaster grown at 17°C and 25°C, respectively. Despite the fact that the time period of the larval stage was more than doubled at the lower temperature, the DNA values were the same in magnitude and similar in distribution of replication classes at each of the stages studied. The data have been interpreted as indicating that, so long as the larval state prevails, initiation of polytenic replication occurs upon completion of the previous cycle.
