    Notes: Abstract The aerobic nitrogen-fixing cyanobacterium, Cyanothece sp. BH68K produces non-mucoid variants defective in exopolysaccharide (EPS) production at a high frequency. The EPS-producing wild-type colonies (EPS+) have a characteristic smooth and shiny appearance which allows them to be easily distinguished from the EPS- variants. When grown on agar plates lacking a source of combined nitrogen, the EPS- variants exhibited a yellow phenotype typical of nitrogen starvation. These EPS- variants showed varying degrees of reversion back to the EPS+ phenotype. After reversion, they exhibited normal diazotrophic growth on agar plates. Alcian blue and ruthenium red staining indicated that the EPS is an acidic polysaccharide, which is present as a loose network around the cell, and which can be completely removed by low speed centrifugation. The accumulation of EPS takes place mainly during the stationary phase. All EPS- variants failed to produce any EPS. Analysis of growth of wild-type and EPS- variants revealed that EPS production is beneficial for diazotrophic growth on solid medium, but not in liquid medium. In addition, EPS phenotypic alteration may have some advantage in the dispersal of cells from one place to another in the natural environment.

    Notes: Abstract The coordinate expression of stress-specific genes is a common response of all organisms to altered environmental conditions. In cyanobacteria, the physiological consequences of stress are often reflected in both the ultrastructure of the cell and in photosynthesis-related properties. This review will focus on the alterations in cyanobacterial pigment-protein organization which occur under different growth conditions, and how several molecular genetic aproaches are being used in this laboratory to investigate the regulatory mechanisms underlying these alterations. We will discuss in detail the response to iron starvation, and present a testable hypothesis for the mechanism of thylakoid reorganization mediated by this response.

    Notes: Abstract Understanding the chemical speciation of metals in solution is necessary for evaluating their toxicity and mobility in soils. Soil samples from the Powder River Basin, Wyoming were extracted with distilled deionised H2O. Soil water extracts were subjected to chemical speciation to determine the relative distribution and chemical forms of copper (Cu), zinc (Zn) and lead (Pb) in acidic environments. As pyrite oxidised, the pH decreased from 6.6 to 2.4, concentration of dissolved sulfate (ST) increased from 259 to 4,388 mg L-1 and concentration of dissolved organic carbon (DOC) decreased from 56.9 to 14.4 mg L-1. Dissolved Cu concentrations ranged from 0.06 to 0.42 mg L-1 and dissolved Zn concentrations ranged from 0.084 to 4.60 mg L-1. Dissolved concentrations of Pb were found to be 0.003 to 0.046 mg L-1. Chemical speciation indicated that at near neutral pH, dissolved metal concentration in soil water extracts was dominated by DOC- metal complexes. At low pH, dissolved metal concentration in soil water extracts was dominated by free ionic forms (e.g. Cu2+, Zn2+, Pb2+) followed by ion pairs (e.g. CuSO4 0, ZnSO inf4 sup0 , PbSO in4 sup0 ). Results obtained in this study suggest that as soil pH decreased, the availability and mobility of metal ions increased due to the chemical form in which these metal ions are present in soil solutions.

    Notes: Abstract The combustion of coal in power plants generates solids (e.g., fly ash, bottom ash) and flue gas (e.g., SO x , CO2). New Clean Air Act mandated reduction of SO x emissions from coal burning power plants. As a result, a variety of Clean Coal Technologies (CCT) are implemented to comply with these amendments. However, most of the CCT processes transfer environmentally sensitive elements (e.g., As, Cd, Pb, Se) from flue gas to CCT ash. The objective of this study was to determine the effect of a pressurized CO2 treatment on the chemistry of CCT ash. Three CCT ash samples, produced from lime injection, atmospheric fluidized bed combustion, and sodium carbonate injection processes were reacted under different CO2 pressure treatment conditions. Treated and untreated samples were subjected to various experiments including, X-ray diffraction (XRD) analysis, calcium carbonate solubility studies, and trace element extraction studies. Factors influencing the efficiency of a CO2 treatment for CCT ash samples include combustion process, moisture, CO2 concentration, and pressure. The CO2 pressure treatment resulted in the precipitation of calcite in CCT ash samples, and thus lowered the pH and the concentration of extractable trace elements (e.g., Cd, Pb, Cr, As, Se). Furthermore, we found that CO2 pressure treatment was more effective for lime injection and atmospheric fluidized bed combustion processed samples than for sodium carbonate injection processed samples.

    Notes: Abstract The plasmid distribution of several clonal isolates of the unicellular, diazotrophic, cyanobacterium Cyanothece sp. has been analyzed. The Cyanothece isolates contain three to four plasmids ranging in size from 4.8 kb to 40 kb. The plasmid profiles of three Cyanothece strains (BH63, BH68, BH93) indicated that strains BH68 and BH93 were closely related and that strain BH63 may be more distantly related. A small 4.8-kb plasmid (pSE480), from the clonal isolate Cyanothece sp. strain BH68F, has been subcloned and restriction mapped. Ten restriction sites have been mapped, five of which are unique and suitable for further subcloning. Southern hybridization revealed that this plasmid was present in two out of five clonal isolates of strain BH68 and in one isolate of strain BH93. A 10-kb plasmid from strain BH68F (pSE1000) was found in all of the BH68 isolates and was absent in the BH93 isolate, Cyanothece sp. strain BH93A. No notable physiological changes were observed in the absence of either the 4.8-kb or 10-kb plasmids. Therefore, these plasmids remain cryptic. Further analysis of these plasmids may provide insight into the function of these plasmids and will allow the construction of shuttle vectors for gene transfer experiments.

    Notes: Abstract The unicellular cyanobacterium, Cyanothece sp. BH68K, is capable of performing both oxygen-sensitive nitrogen fixation and oxygenic photosynthesis within a single cell. To understand the oxygen protection mechanisms of nitrogenase, mutants defective in nitrogen fixation (Nif-) were isolated by use of diethyl sulfate as a mutagen. Out of 24 mutants screened, 6 mutants could not express nitrogenase activity under aerobic conditions, but expressed activity under anaerobic conditions (Fox-); 4 mutants showed no activity under both aerobic and anaerobic conditions (Fix-); and the remaining mutants were impaired in both aerobic and anaerobic nitrogenase activity (Imp). Respiratory oxygen consumption and photosynthetic oxygen evolution were analyzed in the wild-type and in two Fox- mutants. In the wild-type the appearance of high aerobic nitrogenase activity was correlated with an increase in dark respiration, whereas no such increase was seen in the Fox- mutants. We propose that in Fox- mutants, respiratory oxygen consumption plays an important role in maintaining aerobic nitrogenase activity.
