    Description: Allopolyploidization, the combination of the genomes from two different species, has been a major source of evolutionary innovation and a driver of speciation and environmental adaptation. In plants, it has also contributed greatly to crop domestication, as the superior properties of many modern crop plants were conferred by ancient allopolyploidization events. It is generally thought that allopolyploidization occurred through hybridization events between species, accompanied or followed by genome duplication. Although many allopolyploids arose from closely related species (congeners), there are also allopolyploid species that were formed from more distantly related progenitor species belonging to different genera or even different tribes. Here we have examined the possibility that allopolyploidization can also occur by asexual mechanisms. We show that upon grafting--a mechanism of plant-plant interaction that is widespread in nature--entire nuclear genomes can be transferred between plant cells. We provide direct evidence for this process resulting in speciation by creating a new allopolyploid plant species from a herbaceous species and a woody species in the nightshade family. The new species is fertile and produces fertile progeny.     Description: Tissue grafting includes applications ranging from plant breeding to animal organ transplantation. Donor and recipient are generally believed to maintain their genetic integrity, in that the grafted tissues are joined but their genetic materials do not mix. We grafted tobacco plants from two transgenic lines carrying different marker and reporter genes in different cellular compartments, the nucleus and the plastid. Analysis of the graft sites revealed the frequent occurrence of cells harboring both antibiotic resistances and both fluorescent reporters. Our data demonstrate that plant grafting can result in the exchange of genetic information via either large DNA pieces or entire plastid genomes.     Description: Double-stranded RNAs (dsRNAs) targeted against essential genes can trigger a lethal RNA interference (RNAi) response in insect pests. The application of this concept in plant protection is hampered by the presence of an endogenous plant RNAi pathway that processes dsRNAs into short interfering RNAs. We found that long dsRNAs can be stably produced in chloroplasts, a cellular compartment that appears to lack an RNAi machinery. When expressed from the chloroplast genome, dsRNAs accumulated to as much as 0.4% of the total cellular RNA. Transplastomic potato plants producing dsRNAs targeted against the beta-actin gene of the Colorado potato beetle, a notorious agricultural pest, were protected from herbivory and were lethal to its larvae. Mit einer empfindlichen Analysenmethode, die auf die Reaktion CO+HgO→CO2+Hg basiert und den CO-Gehalt auf Grund der Absorption des freigesetzten Hg bei 2537 Å ermittelt, wurden im Gasraum über wachsenden Kulturen von Saccharomyces cerevisiae, S. oviformis, Escherichia coli, Aerobacter aerogenes, Pseudomonas spec. und Lactobacillus brevis 0.4–2.6 ppm CO nachgewiesen. Bei Lactobacillus arabinosus, Bacillus cereus var. mycoides und Aspergillus niger war eine CO-Bildung nicht meßbar. 2. Bei S. cerevisiae war die CO-Bildung bei Konzentrationen von 10–50 g Glucose pro Liter Medium am größten. Außerdem wurde die CO-Bildung proportional zum anfänglichen Sauerstoffgehalt im Gasraum über den Kulturen gefördert. 3. Mit 14C-markierter Glucose wurde nachgewiesen, daß CO aus Glucose entsteht. 4. Die CO-Bildung der untersuchten Mikroorganismen ist so gering, daß sie keine Bedeutung für den Kreislauf dieses Spurengases in der Atmosphäre hat.
    Notes: Summary 1. Growing cultures of Saccharomyces cerevisiae, S. oviformis, Escherichia coli, Aerobacter aerogenes, Pseudomonas spec. and Lactobacillus brevis produce trace amounts of CO (0.4–2.6 ppm) that can be detected in the gas space above the cultures using a sensitive analytical method based on the reaction CO+HgO→CO2+Hg. The liberated Hg is quantitatively measured by atomic absorption at 2537 Å. No CO could be detected above cultures of Lactobacillus arabinosus, Bacillus cereus var. mycoides and Aspergillus niger. 2. The maximum CO production by Saccharomyces was obtained with concentrations of 10–50 g glucose per liter medium. The amount of CO formed increased with the oxygen concentration in the gas space above the cultures. 3. Using 14C-glucose it was shown that S. cerevisiae forms CO from glucose. 4. The formation of CO by the microorganisms investigated is very small. The ratio of CO/CO2 produced is much smaller than in environmental air. Therefore it can be concluded that the production of CO by these microorganisms has probably no significance for the atmospheric cycle of this trace gas.
