    Notes: Abstract The response of bone mass to long-term treatment with estrogen and progesterone in patients with complete androgen-insensitivity syndrome (AIS) is unknown. We report a 17-year-old female patient (karyotype 46 X, Y) with AIS studied during a 4-year period. Bone mineral density (BMD) measured by dual X-ray absorptiometry in lumbar spine and proximal femur was sharply reduced at the initial visit, and remained unchanged during long-term follow-up on hormone replacement therapy with estrogens and progestin. Bone metabolism markers were all in the normal range. The lack of significant increase in BMD highlights the importance of androgens on bone physiology that cannot be balanced in spite of an appropriate estrogenic milieu.

    Notes: Abstract The expression pattern of a peroxidase – (EC 1.11.1.7) encoding gene, TPX1, was studied in NaCl-adapted and unadapted tomato (Lycopersicon esculentum) cells growing in suspension culture. We followed the transcript level during the cell growth cycle and under different salt treatments. There was a cell-cycle-dependent TPX1 expression, the maximum level of transcripts coinciding with the period of cell enlargement in both salt-adapted and unadapted tomato cells. However, enzyme activity of the encoded isoperoxidase was only detected in the culture medium of the salt-adapted cells. NaCl treatment decreased TPX1 transcripts in both cell lines, but the threshold concentration for this down-regulation was higher in adapted cells. In conclusion, salt-adapted tomato cells show significant changes in comparison with unadapted cells in the expression of a gene encoding a cell-wall-targeted peroxidase.

    Notes: Summary The crown light environments of saplings of two Costa Rican rain forest tree species were simultaneously compared. The species, Dipteryx panamensis (Pitt.) Record & Mell., a relatively shade-intolerant species, and Lecythis ampla Miers, a shade-tolerant species, have contrasting growth and branching patterns. Quantum sensors were placed throughout the crowns of saplings up to 2.5 m tall and quantum fluxes were recorded with microloggers for seven-day periods. The shade-intolerant species had total quantum flux densities 35% larger than those of the shade-tolerant species, but totals for both species were less than 2% of full sun. More than 90% of the quantum flux densities measured within the crowns of both species were less than 25 μmol m-2s-1. Lateral light was an important component of daily quantum flux totals; for saplings of both species, the half-hour with the maximum average irradiance for the day frequently occurred in mid-morning or midafternoon. Despite dissimilar crown and leaf display, there was no difference in the overall variability of irradiance within the crowns of the two species. However, quantum fluxes received within the crowns differed substantially in both species. Within-crown locations differed significantly from day to day because of variation in weather conditions. Daily total quantum flux densities and totals expressed as a percent of full sun were significantly correlated with height growth over the previous 12 months.

    Notes: Abstract   We examined the effects of pollen selection for rapid pollen-tube growth on progeny vigor. First, we crossed a wild gourd (Cucurbita texana) to a cultivated zucchini (Cucurbita pepo cv `Black Beauty') to produce an F1 and then an F2 generation. Half of the F1 seeds were produced by depositing small loads of C. texana pollen onto the stigmas of C. pepo. These small pollen loads were insufficient to produce a full complement of seeds and, consequently, both the fast- and the slow-growing pollen tubes were permitted to achieve fertilization. An F2 generation was then produced by depositing small loads of F1 pollen onto stigmas of F1 plants. The F2 seeds resulting from two generations of small pollen loads are termed 'the non-selected line' because there was little or no selection for pollen-tube growth rate on these plants. The other half of the F1 and F2 seeds were produced by depositing large pollen loads (〉10000 pollen grains) onto stigmas and then allowing only the first 1% or so of the pollen tubes that entered the ovary to fertilize the ovules. We did this by excising the styles at the ovary at 12–15 h after pollination. The resulting F2 seeds are termed `the selected line' because they were produced by two generations of selection for only the fastest growing pollen tubes. Small pollen loads from the F2 plants, both the selected and the non-selected lines, were then deposited onto stigmas of different C. pepo flowers, and the vigor of the resulting seeds was compared under greenhouse and field conditions. The results showed that the seeds fertilized by pollen from the selected line had greater vegetative vigor as seedlings and greater flower and fruit production as mature plants than the seeds fertilized by pollen from the non-selected line. This study demonstrates that selection for fast pollen-tube growth (selection on the microgametophyte) leads to a correlated increase in sporophyte (progeny) vigor.

    Notes: Summary This study examines the assumption of the pollen competition hypothesis that genetic differences among microgametophytes lead to differences in pollen performance and result in non-random fertilization. In addition, we examined the assumption that pollen performance is genetically correlated with sporophyte vigor due to an overlap in gene expression between the two stages of the life cycle. The results from a pollen mixture experiment in which two cultivars of common zucchini were used show that the ability to sire seeds is nonrandom with respect to the cultivar of the pollen donor plant. The proportion of the progeny sired by the two cultivars is not independent of the region of the fruit where the seeds are produced. The progeny sired by the yellow cultivar outperformed the progeny sired by the green cultivar in a greenhouse study. In addition, the progeny sired by the yellow cultivar from the stylar region of the fruit germinated faster and had more leaf area than the progeny sired by the same cultivar from the peduncular end of the fruit. Thus, the most vigorous progeny are obtained from the stylar region of the fruit where the ovules are fertilized by the most vigorous microgametophytes.

    Notes: Abstract. Changes of soluble and ionically bound peroxidase and indoleacetic acid (IAA) oxidase activities were followed during peach seed development. Soluble peroxidase activity was located mainly in the embryo plus endosperm tissue, whereas wall ionically bound activities were found predominantly in the integument tissue. The different peroxidase isoenzymes present in the extracts were characterized by polyacrylamide gel electrophoresis and isoelectric focusing; the main soluble isoenzyme of embryo plus endosperm tissue was an anionic isoperoxidase of R F 0.07. Basic ionically bound isoenzymes were located only in the integument tissue, but two soluble anionic isoenzymes of R F 0.23 and 0.51 were also present in this tissue. In parallel, peroxidase protein content was estimated specifically using polyclonal antibodies. The kinetic data and the changes of seed IAA oxidase activity during fruit development suggested that basic peroxidase isoenzymes from ionically bound extracts of integument might be involved in IAA degradation.
