    Notes: Abstract. Ca2+-regulated motility is essential to numerous cellular functions, including muscle contraction. Systems with troponin C, myosin light chain, or calmodulin as the Ca2+ receptor have evolved in striated muscle and other types of cells to transduce the cytoplasm Ca2+ signals into allosteric conformational changes of contractile proteins. While these Ca2+ receptors are homologous proteins, their coupling to the responding elements is quite different in various cell types. The Ca2+ regulatory system in vertebrate striated muscle represents a highly specialized such signal transduction pathway consisting of the troponin complex and tropomyosin associated with the actin filament. To understand the molecular mechanism in the Ca2+ regulation of muscle contraction and cell motility, we have revealed a preserved ancestral close linkage between the genes encoding two of the troponin subunits, troponin I and troponin T, in the genome of mouse. The data suggest that the troponin I and troponin T genes may have originated from a single locus and evolved in parallel to encode a striated muscle-specific adapter to couple the Ca2+ receptor, troponin C, to the actin–myosin contractile machinery. This hypothesis views the three troponin subunits as two structure–function domains: the Ca2+ receptor and the signal transducing adapter. This model may help to further our understanding of the Ca2+ regulation of muscle contraction and the structure–function relationship of other potential adapter proteins which are converged to constitute the Ca2+ signal transduction pathways governing nonmuscle cell motility.

    Notes: A procedure of thermo-mechanical treatment (TMT) was carried out to 2519 aluminumalloy to improve its properties. The influences of cold deformation ratio, ageing temperature andageing time of the TMT on the mechanical properties and electrochemical corrosion resistance of thisalloy were investigated. The results show that after TMT the tensile strength of 2519 aluminum alloycan be improved obviously but its electrochemical corrosion resistance decreases slightly. Theoptimum processing parameters of the TMT for 2519 aluminum alloy can be described as: solidsolution at 530°Cfor 0.5h, then cold deform with a ratio of 15% followed by aging at 150°C for10h

    Notes: Abstract The new color reactions of titanium (IV) with six 2,6,7-trihydroxyl-9-phenylsubstituted-3-fluorone derivatives (THPF) have been studied in the presence of nitrilotriacetic acid (NTA) and cetyltrimethylammonium bromide (CTMAB). The effects of secondary ligands such as NTA, ascorbic acid, hydroxylamine and mandelic acid on the ternary complex of titanium (IV) have been reported. It was found that the system of titanium (IV)-2,6,7-trihydroxyl-9-(2′,4′-dichlorophenyl)-3-fluorone (DCPF)-NTA-CTMAB quaternary complex is the most sensitive in these systems studied. The apparent molar absorptivity of the complex at 576 nm is 1.9 × 105 1·mol−1·cm−1. The stoichiometric ratio of the Ti (IV)-NTA-DCPF-CTMAB complex is estimated to be 1 ∶ 1 ∶ 3 ∶ 3. The proposed method proves to be satisfactory for the direct determination of titanium in alloy steels, silicate minerals and aluminium alloys. The method is highly sensitive and selective.

    Notes: Summary Nebulin, a giant myofibrillar protein with size variants from 700 to 900 kDa in various skeletal muscles, has been proposed to constitute a set of inextensible filaments anchored at the Z-line and coextensive with actin filaments. To elucidate the architectural organization of this fourth set of myofilaments in the skeletal muscle sarcomere, we performed immunoelectron microscopic localization of epitope profiles of a number of site-specific monoclonal antibodies against cloned human nebulin fragments of known sequence loci. Monoclonal antibody N113, which is directed to fragment ND8 at approximately 300 residues away from the C-terminus, labelled the edges of Z-lines in both human quadriceps muscle and rabbit psoas muscle. Monoclonal antibody N101, which is directed to fragment NB5 near the N-terminal side, is localized to a single locus at 0.89 μm from the Z-line in human quadriceps muscle and 0.80μm from the Z-line in rabbit psoas muscle. Additionally, monoclonal antibody N109, which is directed to fragment NA3 on the carboxy side of the adjacent fragment NB5, is localized at 0.76 μm away from the Z-line in rabbit psoas muscle. This one-to-one correspondence between epitope loci and sequence loci demonstrates that a single nebulin polypeptide spans the length of the thin filament with its C-terminus anchored at the Z-line. The epitope spacings of site-specific antibodies are consistent with the notion that the nebulin filament is uniform in mass density along its length. We conclude that the thin filament, as defined morphologically by electron microscopy, is a composite filament of the conventional actin thin filament (actin/tropomyosin/troponin) and coextensible nebulin polypeptides which act as full-length molecular templates that regulate or stabilize colaterally the actin filament in the skeletal muscle sarcomere.

    Notes: Abstract Neuronal cdc2-like kinase, nclk, is a heterodimer of cyclin dependent protein kinase 5, cdk5, and a 25 kDa subunit derived from a novel, neuron-specific, 35 kDa protein: p35. The characterization and regulation of nclk will be summarized in this minireview. The activity of nclk appears to be governed by highly complex regulatory mechanisms including protein-protien interaction, protein phosphorylation and isoforms. The histone H1 kinase activity of nclk is absolutely dependent of the interaction between the 25 kDa subunit and the catalytic subunit, cdk5. In addition, nclk interacts with other cellular proteins to form macromolecular complexes. The kinase activity of nclk is inhibitedin vitro by the phosphorylation reactions of a weel-like protein tyrosine kinase and a protein serine/threonine kinase from bovine thymus. Northern blot analysis has revealed the existence of two populations of p35 mRNA of 2 and 4 kb. A novel cDNA encoding a p35 homologous protein has been obtained from a human hippocampus library.

    Notes: The coating flow of a reactive fluid on an inclined plane is studied. The fluid viscosity steadily increases due to a sustained cross-linking reaction. Using well-known long wave approximations and a simple kinetic relation, we find that the linear stability of a uniform coating is affected not only by the inclination angle but also by the reaction order. In the weakly nonlinear limit, a generalized Kuramoto-Sivashinsky (K-S) equation is derived. Two interesting cases are identified in the limit of creeping flow. One is when surface tension is moderate and a generalized Burger's equation results, and the other is when the surface tension is large and the traditional K-S equation is recovered when the inclination angle is greater than 90°. The solution in this case represents a shock-like traveling wave down the incline and it appears to be unique for reacting coating flows.
