    Description: We present the first synthesis of global peatland extent through the last glacial cycle (130 ka) based on 〉975 detailed stratigraphic descriptions from exposures, soil pits, and sediment cores. Buried peats are defined as organic-rich sediments overlain by mineral sediments. Also included are deposits rich in wetland macrofossils indicated a local peatland environment. The dataset includes location (lat/long), chronologic information (when available), a description of the buried peat sediment, overlying and underlying sediments, whether geochemical information is available, and the original references.

    Description: These data describe peat initiation and development in the Falkland Islands, South Atlantic. The datasets presented represent a combination of primary data collection and our compilation of previously-published data. The 'falklands_basal_dates' file lists radiocarbon dates which can be considered to represent peat initiation. Data are for: site name, location (northing/easting), radiocarbon date details (labID/age/error), basal depth, data source (reference given where data is secondary) and a brief description of the basal stratigraphy. The 'falklands_core_properties' dataset gives details of peat properties for cores from ten sites across East Falkland sampled in this study. Data are for: site codes (locations as listed in 'falklands.basal.dates), bulk density (determined by water displacement and drying at 105C), loss on ignition (combustion at 550C), %carbon (elemental analysis) and modelled age using Bacon. The 'falklands_depths' dataset gives measured peat depth for 371 sites. Depths listed are means of variable numbers of individual site measurements with a maximum measurable depth of 250cm. Full details of all datasets are given in the accompanying paper. Collectively the data presented here give the first synthesis of peat age and properties in the Falkland Islands.

    Notes: The morphological response, as measured by changes to mycelial area, radial extension and border fractal dimension, of the oomycete Achlya bisexualis to Cu, Co, Hg, Zn and Cd at concentrations of between 0.05 and 3 mM is described. All of the metals decreased mycelial area and radial extension. Border fractal dimension increased in the presence of Cu, Co and Hg with individual hyphae extending out beyond the mycelial margin. In the presence of 3 mM Hg these hyphae displayed spiral growth. Zn and Cd had no effect on border fractal dimension. We suggest that all of the metals slow growth and that Cu, Co and Hg may also disrupt the relationship between tip growth and branching at the edge of the mycelium.

    Description: Glacial−interglacial variations in CO2 and methane in polar ice cores have been attributed, in part, to changes in global wetland extent, but the wetland distribution before the Last Glacial Maximum (LGM, 21 ka to 18 ka) remains virtually unknown. We present a study of global peatland extent and carbon (C) stocks through the last glacial cycle (130 ka to present) using a newly compiled database of 1,063 detailed stratigraphic records of peat deposits buried by mineral sediments, as well as a global peatland model. Quantitative agreement between modeling and observations shows extensive peat accumulation before the LGM in northern latitudes (〉40°N), particularly during warmer periods including the last interglacial (130 ka to 116 ka, MIS 5e) and the interstadial (57 ka to 29 ka, MIS 3). During cooling periods of glacial advance and permafrost formation, the burial of northern peatlands by glaciers and mineral sediments decreased active peatland extent, thickness, and modeled C stocks by 70 to 90% from warmer times. Tropical peatland extent and C stocks show little temporal variation throughout the study period. While the increased burial of northern peats was correlated with cooling periods, the burial of tropical peat was predominately driven by changes in sea level and regional hydrology. Peat burial by mineral sediments represents a mechanism for long-term terrestrial C storage in the Earth system. These results show that northern peatlands accumulate significant C stocks during warmer times, indicating their potential for C sequestration during the warming Anthropocene.

    Description: Hematophagous organisms produce a suite of salivary proteins which interact with the host’s coagulation machinery to facilitate the acquisition and digestion of a bloodmeal. Many of these biomolecules inhibit the central blood-clotting serine proteinase thrombin that is also the target of several clinically approved anticoagulants. Here a bioinformatics approach is used to identify seven tick proteins with putative thrombin inhibitory activity that we predict to be posttranslationally sulfated at two conserved tyrosine residues. To corroborate the biological role of these molecules and investigate the effects of amino acid sequence and sulfation modifications on thrombin inhibition and anticoagulant activity, a library of 34 homogeneously sulfated protein variants were rapidly assembled using one-pot diselenide-selenoester ligation (DSL)-deselenization chemistry. Downstream functional characterization validated the thrombin-directed activity of all target molecules and revealed that posttranslational sulfation of specific tyrosine residues crucially modulates potency. Importantly, access to this homogeneously modified protein library not only enabled the determination of key structure–activity relationships and the identification of potent anticoagulant leads, but also revealed subtleties in the mechanism of thrombin inhibition, between and within the families, that would be impossible to predict from the amino acid sequence alone. The synthetic platform described here therefore serves as a highly valuable tool for the generation and thorough characterization of libraries of related peptide and/or protein molecules (with or without modifications) for the identification of lead candidates for medicinal chemistry programs.
