    Description: We have localized cathepsin K in rat osteoclasts and within exposed resorption pits by immuno-fluorescence microscopy. Intracellular staining using an antibody raised against recombinant mouse cathepsin K was vesicular and uniformly distributed throughout the cell. Confocal microscopy analysis did not reveal an accumulation of cathepsin K containing vesicles opposing the ruffled border and the resorption lacuna. Exposed resorption pits exhibited a uniform distribution of cathepsin K, and no differences were observed between the edges and the centers of the pits. The immunostaining of resorption pits with anti-cathepsin K antibodies demonstrates that the protease is secreted into the sub-osteoclastic compartment.Cathepsin K-specific inhibition using peptidyl vinyl sulfones as selective cysteine protease inactivators reduced bone resorption by 80% in a dose-dependent manner at sub-micromolar concentrations. No reduction of bone resorption was observed at those low concentrations using a potent cathepsin L, S, B-specific inhibitor. That the inhibition of bone resorption can be attributed to cathepsin K-like protease inhibition was corroborated by the selective inhibition of the osteoclastic Z-Gly-Pro-Arg-MβNA hydrolyzing activity by the cathepsin K, L, S, B-inhibitor, but not by the cathepsin L, B, and S inhibitor. Z-Gly-Pro-Arg-MβNA is efficiently hydrolyzed by cathepsin K but only poorly by cathepsins L, S, and B. On the contrary, the intracellular hydrolysis of the cathepsin B-specific substrate, Z-Arg-Arg-MβNA, was prevented by both types of inhibitors.The identification of cathepsin K in resorption pits and the inhibition of bone resorption and intracellular cathepsin K activity by selective vinyl sulfone inhibitors indicate the critical role of the protease in osteoclastic bone resorption.

    Description: Described from oak apple galls and fallen buds of Quercus robur in the Netherlands and recently synonymized with Ciborinia candolleana, Sclerotinia bresadolae is reinstated as an independent species after studying Rick’s type material in Oudemans’s herbarium and fresh collections on oak apples from England and Germany. According to Boudier’s figures and published descriptions, and the study of two English collections, Ciborinia hirtella seems to be a synonym.

    Description: The 31 collections of Nidulariaceae in Persoon\xe2\x80\x99s Herbarium, bearing 23 names in all, are revised into 8 species: Crucibulum laeve (Bull. ex DC.) Kambly, Cyathus microsporus Tul., C. olla (Batsch) ex Pers., C. poeppigii Tul., C. stercoreus (Schw.) De Toni, C. striatus (Huds.) ex Pers., C. triplex Lloyd and Nidularia farcta (Roth ex Pers.) Fr. Nine names are unpublished and hence nomina nuda. Six specimens appear to be of West Indian origin and two from Mauritius. As one West Indian specimen was named Nidularia Domingensis\xe2\x80\x9d, the type of Cyathus microsporus var. domingensis Tul., which is the type variety of the species, was examined and the two collections agree in most respects. However, the spores of C. microsporus are larger than is given in the literature. The spores of both C. striatus and C. triplex are reported to have a proximal notch which does not appear to have been previously reported.

    Description: The authentic collections of Geastrum pectinatum Pers., G. plicatum Berk. and G. tenuipes Berk, are redescribed. Persoon\xe2\x80\x99s collection in the Rijksherbarium, Leiden, is designated as the Neotype of G. pectinatum. Geastrum plicatum and G. tenuipes are considered as probable synonyms, although observations on freshly expanded specimens are still required. Comparisons are made between freshly collected and dried material in the British Museum (Nat. Hist.), the Hartley Botanical Laboratories, the Rijksherbarium and Herb. Kew, and the literature is discussed.
