    Notes: Summary We have studied the disposition of perhexiline and its two major metabolites, M1 and M3, in healthy volunteers and in patients with biliary T-tube drains after cholecystectomy. In healthy volunteers the genetic control for impaired hepatic oxidation is identical for debrisoquine, sparteine, and perhexiline. Poor metabolizers demonstrate markedly reduced production and excretion of the major metabolite, M1. Their production of M3 is also reduced, but to a lesser degree than for M1, confirming substrate stereoselectivity by hepatic oxidases. Biphasic urinary elimination of M1 and M3 is seen in intact extensive oxidizers, whereas only the first phase is apparent in patients with biliary T-tube drainage. This suggests the possibility of enterohepatic recycling of these compounds, which may account for their prolonged elimination More than 90% of an ingested dose of perhexiline maleate remains unaccounted for at 24h after ingestion, even in extensive metabolizers. A careful, radiolabelled tissue-distribution study is warranted to elucidate the complicated metabolic fate of perhexiline.

    Notes: Abstract Two-year-old blue tilapia, Oreochromis aureus (Steindachner), were stocked into five replicate breeding hapas (1·2 m × 1·2 m × 2·7 m) at a density of six males and six females per hapa. At stocking, males and females averaged 415 and 225g respectively. During the 50-day trial (July-August), 62900 seed (eggs and sac-fry) representing 78 spawns, were collected from 30 females. In comparative treatments, prestocking surgical removal of the male's maxillary bone did not influence hatchery productivity. However, removal of the female's maxillary bone significantly reduced frequency of repeat spawning and clutch size (P 〈 0·05). Removal of the maxillary bone also influenced weight gain of broodstock. It is hypothesized that these significant responses were due to the surgical removal of the maxillary bone and the subsequent breakdown in dominance hierarchies.

    Notes: Abstract. The origin, sequence and location of molecular damage arising during sub-lethal drought is described in 4-week-old wheat (Triticum aestivum L.). Short episodes of water deprivation increased the uptake of iron within 6h of drought. Superoxide, precursor of the highly reactive hydroxyl radical in the presence of iron, was formed in the thylakoid membranes. Further draughting promoted destruction of chlorophylls (but not carotenoids), which was detectable after 14 h and preceded significant losses in relative water content. Peroxidation of lipids and a decreased protein content occurred after 26h of cumulative water deprivation but was confined to the shoots. In contrast, there was no significant loss of protein nor peroxidation of lipids in the root tissue, despite quantitatively larger, more rapid, losses in water content. These results indicate that damage arising in the early phases of drought appears to be initiated within the chloroplast and to be largely confined to the photosynthetic tissue. The pattern of drought-induced damage in 11 other non-xerophytes of the north temperate region was qualitatively similar to wheat, although the response to drought of the three cereals was more extreme than in the indigenous grasses and less extreme in ecologically stress-tolerant species. A method for the simultaneous quantification of carotenoids in the presence of chlorophylls is provided.

    Notes: Abstract  Drought is a major abiotic stress of upland rice, and good root growth has been associated with drought avoidance. We report on the genetic mapping of root growth traits in an F2 population derived from two drought-resistant rice varieties, ‘Bala’ and ‘Azucena’. Restriction fragment length polymorphism (RFLP) between the parents was 32%, and a molecular map with 71 marker loci and 17 linkage groups covering 1280 cM was produced. Quantitative trait loci (QTLs) for eight root growth characteristics were mapped using phenotype data obtained in a hydroponic screen previously described in a companion paper. Using a significance threshold of LOD 2.4, we observed one QTL for maximum root length after 28 days growth on chromosome 11. It had a LOD score of 6.9, explained nearly 30% of the variation and appeared to be largely additive in effect. QTLs for maximum root length after 3, 7, 14 and 21 days of growth were also revealed. Some root-length QTLs, including that on chromosome 11, varied greatly with developmental stage. One QTL for root volume and two QTLs for adventitious root thickness were detected. No QTLs were detected for the length of cells in the mature (fully expanded) zone of adventitious root tips. The results obtained are discussed in the context of previous reports on mapping root growth parameters in rice.

    Notes: Abstract  Root growth is an important component of the adaptation of rice to drought-prone environments. A hydroponic screen was used to study root growth of 28 rice varieties. Both maximum root length and adventitious root thickness varied widely between varieties. In general, japonica varieties had larger root systems than indica varieties. Two F2 populations involving the thick- and long-rooted upland japonica variety ‘Azucena’ and two poor-rooting varieties, namely the upland indica‘Bala’ and the Italian japonica‘Maratelli’, were made and screened in hydroponics. Generation means analysis revealed significant additive and dominance main effects for the root length traits with a prevalence of dominance gene effects in both crosses. The dominance×dominance type of non-allelic interactions were important for maximum root length from day 7 to day 28, root volume, root thickness and root cell length in the cross ‘Bala’בAzucena’. The heritability (broad-sense) estimates varied from low to high for the traits and displayed differences between populations. This suggested that recombinant lines with improved root traits can be developed from the two crosses with selection methods that involve some form of progeny evaluation. In a companion paper, we report the mapping of quantitative trait loci (QTLs) for root growth traits in the ‘Bala’בAzucena’ population using restriction fragment length polymorphisms (RFLPs).
