    Description: The transcriptional coactivator p300/CBP (CREBBP) is a histone acetyltransferase (HAT) that regulates gene expression by acetylating histones and other transcription factors. Dysregulation of p300/CBP HAT activity contributes to various diseases including cancer. Sequence alignments, enzymology experiments and inhibitor studies on p300/CBP have led to contradictory results about its catalytic mechanism and its structural relation to the Gcn5/PCAF and MYST HATs. Here we describe a high-resolution X-ray crystal structure of a semi-synthetic heterodimeric p300 HAT domain in complex with a bi-substrate inhibitor, Lys-CoA. This structure shows that p300/CBP is a distant cousin of other structurally characterized HATs, but reveals several novel features that explain the broad substrate specificity and preference for nearby basic residues. Based on this structure and accompanying biochemical data, we propose that p300/CBP uses an unusual 'hit-and-run' (Theorell-Chance) catalytic mechanism that is distinct from other characterized HATs. Several disease-associated mutations can also be readily accounted for by the p300 HAT structure.     Description: Ocean dynamics, land motion, and changes in Earth’s gravitational and rotational fields cause local sea level change to deviate from the rate of global mean sea level rise. Here, we use observations and simulations of spatial structure in sea level change to estimate the likelihood that these processes cause sea level trends in the longest and highest-quality tide gauge records to be systematically biased relative to the true global mean rate. The analyzed records have an average 20th century rate of approximately 1.6 mm/yr, but based on the locations of these gauges, we show the simple average underestimates the 20th century global mean rate by 0.1   ±  0.2 mm/yr. Given the distribution of potential sampling biases, we find  〈 1% probability that observed trends from the longest and highest-quality TG records are consistent with global mean rates less than 1.4 mm/yr.

    Description: Recent decadal sea surface height (SSH) variability across the Equatorial and North Indian Ocean (ENIO, north of 5°S) is spatially coherent and related to a reversal in basin-scale, upper-ocean-temperature trends. Analysis of ocean and forcing fields from a data-assimilating ocean synthesis (ECCOv4) suggests that two equally-important mechanisms of wind-driven heat redistribution within the Indian Ocean account for a majority of the decadal variability. The first is the Cross-Equatorial Cell (CEC) forced by zonal-wind-stress curl at the equator. The wind-stress curl variability relates to the strength and position of the Mascarene High, which is influenced by the phase of the Indian Ocean Subtropical Dipole. The second mechanism is deep (700 m) upwelling related to zonal wind stress at the equator that causes deep, cross-equatorial overturning due to the unique geometry of the basin. The CEC acts to cool the upper ocean throughout most of the first decade of satellite altimetry, while the deep upwelling delays and then amplifies the effect of the CEC on SSH. During the subsequent decade, reversals in the forcing anomalies drive warming of the upper ocean and increasing SSH, with the effect of the deep upwelling leading the CEC.     Description: Interannual to decadal sea level variability on the North Atlantic western boundary is surprisingly coherent over substantial distances stretching from the Caribbean to Nova Scotia. The physical mechanisms responsible for this basin-scale, low-frequency coherence are explored in a diagnosis of simulated ocean fields from GECCO, which reproduces the observations to good approximation. Coastal sea level variability on the western boundary is known to be influenced by meridional divergence in the boundary current resulting in a geostrophic tilting of the sea surface. This mechanism is found to be of leading order along some stretches of the boundary, but it does not account for the coherence spanning the western North Atlantic. Instead, the coherence along the entire boundary is accounted for by vertical divergence resulting in the uniform rise and fall of the sea surface west of the 295°E meridian. The vertical divergence is found to be due to net vertically integrated zonal transport across this meridian resulting from meridional variation in the Sverdrup transport over the basin interior.
