    Notes: We have isolated an H-2 deficient cell line (LR.4) from the T-cell lymphoma L5178Y which grew without restrictions in the peritoneal cavity of different inbred strains of mice. The use of polyclonal anti-H-2 antiserum and complement indicated that LR.4 cells did not express class I determinants on the cell membrane. Southern blots of genomic DNA of LR.4 cells showed that B2m genes were severely damaged and that class I H-2 genes were extensively methylated. Consequently, LR.4 cells failed to transcribe mRNAs for both B2m and class I H-2 genes. On the other hand, specific immunity to LR.4 was demonstrated in C57BL/6J mice since, in subsequent challenges with either LR.4 or EL4.4, LR.4 did not grow, whereas EL4.4 grew and killed the mice. In C57BL/6J mice, rejection of LR.4 was accompanied by the production of cytotoxic antibodies. The immune response induced in C57BL/6J mice was determined by non-H-2 antigenic determinants in LR.4 cells.

    Notes: Summary A region of the mitochondrial genome associated with cytoplasmic male sterility (CMS) in Phaseolus vulgaris was flanked by two different repeated sequences designated x and y. The DNA sequence of the CMS-unique region and a portion of each flanking repeat was determined. Repeat x contained a complete coding copy of the F1 ATPase subunit A (atp A) gene, as well as an open reading frame (orf) predicting a protein of 209 amino acids. The TGA termination codon of the atpA gene and the ATG initiation codon of orf209 were overlapping. These reading frames were oriented with their 3′ ends proximal to the CMS-unique region. The CMS-unique region of 3736 nucleotides contained numerous orfs. The longest of these predicted proteins being of 239, 98 and 97 amino acids. The 3′ coding and 3′ flanking regions of orf98 were derived from an internal region of the higher plant chloroplast tRNA alanine intron. The region of repeat y immediately adjacent to the CMS-unique region contained the 111 carboxy-terminal coding residues of the apocytochrome b (cob) gene. This segment was oriented with its 5′ end proximal to the CMS-unique region, but cob gene sequences were not fused to an initiation codon within the unique region.

    Notes: Summary Genetic variation in Phaseolus vulgaris L. (P. vulgaris) was investigated at the isozyme and DNA levels. We constructed a library of size-selected Pst I clones of P. vulgaris nuclear DNA. Clones from this library were used to examine 14 P. vulgaris accessions for restriction fragment length polymorphisms (RFLPs). DNAs from each accession were analyzed with three restriction enzymes and 18 single copy probes. The same accessions were also examined for variability at 16 isozyme loci. Accessions included four representatives of the T phaseolin group and five representatives each of the C and S phaseolin groups. One member of the S group (the breeding line XR-235-1-1) was derived from a cross between P. vulgaris and P. coccineus. Isozymes and RFLPs revealed very similar patterns of genetic variation. Little variation was observed among accessions with C and T phaseolin types or among those with the S phaseolin type. However, both isozyme and RFLP data grouped accessions with S phaseolin separately from those accessions with C or T phaseolin. The highest degree of polymorphism was observed between XR-235-1-1 and members of the C/T group. RFLP markers will supplement isozymes, increasing the number of polymorphic loci that can be analyzed in breeding, genetic, and evolutionary studies of Phaseolus.

    Notes: Abstract Studies were performed to define tissue culture techniques and culture conditions for morphogenesis, callus culture and plantlet culture of sweet orange (Citrus sinensis (L.) Osb.), citron (C. medica L.) and lime (C. aurantifolia) (Christm. Swing). The optimal concentrations of NAA to induce root formation on stem segments were 10 mg l-1 for sweet orange and lime, and 3 mg l-1 for citron. The optimal BA concentration for shoot and bud proliferation was 3 mg l-1 for sweet orange and citron, and 1 mg l-1 for lime. Callus initiation was accomplished in a culture medium containing 10 mg l-1 NAA and 0.25 mg l-1 BA. Callus was maintained by periodical subculture into the same medium supplemented with 10% (v:v) organge juice. In vitro plantlets of the three species were obtained by rooting of shoots developed from bud cultures, and of citron and lime by development of shoots from root cultures. The plants were successfully established on soil.

    Notes: Abstract Both incubation temperature and photosynthetic radiation affected morphogenesis, callus culture and plantlet culture of sweet orange (Citrus sinensis) cultured in vitro. Bud culture from nodal stem segments, regeneration of shoots and buds from internode stem segments and induction of primary callus were near optimal at incubation temperatures between 21–30°C. The optimal temperature for root formation was 27°C with temperatures above and below being clearly deleterious. Incubation in the dark or under low photosynthetic photon flux density (PPFD) was beneficial for callus induction and growth and also favored the production of rooted plantlets from bud cultures. Incubation in the dark improved considerably the regeneration of shoots and buds from internode segments and the recovery of whole plants. No off-types, as determined by protein and isoenzyme analysis, were observed among plantlets recovered from bud cultures or from regeneration of shoots from internode stem segments.

    Description / Table of Contents: Abstract Using rabbits, we have studied the effect of reinfection of T. mentagrophytes var granulosum and T. rubrum into lesions that were previously infected and resolved. Clinical mycological and histopathological studies were done for 16 weeks. Timentagrophytes produced a more severe infection than T. rubrum. The clinical lesions produced by reinoculation were less intense and long lasting.
    Notes: Résumé On réalise une étude expérimentale sur des lapins inoculés et réinoculés par Trichophyton mentagrophytes var. granulosum et Trichophyton rubrum, pour connaitre l'évolution clinique, mycologique et histopathologique pendant 16 semaines. Au cours de la premiere inoculation, les lésions cliniques, ainsi que le patron histopathologique produit par T. mentagrophytes var. granulosum, furent beaucoup plus grandes en intensité et durée que celles dûes á T. rubrum. Au cours de la réinoculation, les lésions cliniques produites para les deux dermatophytes ont eu une intensité et une durée moindres.

    Description: When ductile alloys are subject to sliding wear, small increments of plastic strain accumulate into severe plastic deformation and mechanical alloying of the surface layer. The authors constructed a simple coaxial tribometer, which was used to study this phenomenon in wrought Al-Sn and cast Cu-Mg-Sn alloys. The first class of materials is ductile and consists of two immiscible phases. Tribological modification is observed in the form of a transition zone from virgin material to severely deformed grains. At the surface, mechanical mixing of both phases competes with diffusional unmixing. Vortex flow patterns are typically observed. The experimental Cu-Mg-Sn alloys are ductile for Mg-contents up to 2 wt% and consist of a- dendrites with a eutectic consisting of a brittle Cu 2 Mg-matrix with α-particles.     Description: 〈span〉〈div〉Summary〈/div〉The marine magnetotelluric (MT) method is a useful tool for offshore studies aimed at, for example, hydrocarbon exploration and the understanding of Earth's tectonics. Marine MT data are often distorted by coastlines because of the strong resistivity contrast between the conductive ocean and the resistive land. At mid ocean ridges, the resistivity of Earth's structure can be assumed to be two-dimensional (2D), which allows MT data to be decomposed into a transverse electrical (TE) mode, with electric current flowing approximately along the ridge, and a transverse magnetic (TM) mode, with electric current flowing perpendicular to the ridge. We collected marine MT data at the middle Atlantic Ridge which exhibited highly negative TM-mode phases, as large as -180°, at relatively high frequencies (0.1 to 0.01 Hz). Similar negative phases have been observed in other marine MT datasets, but have not been the subject of study. We show here that these negative phases are caused by a newly distinguished coast effect. The TM-mode coast effect is not only a galvanic effect, as previously understood, but also includes inductive distortions. TM-mode negative phases are caused by the turning of the Poynting vector, the phase change of electromagnetic fields, and vertically flowing currents in the sea floor. The findings provide a new understanding of the TM-mode coast effect, which can guide our ability to fit the field data with the inclusion of coastlines, and reduce misinterpretation of the data in offshore studies. The study also shows that the TM-mode coast effect is sensitive to the depth and conductivity of the asthenosphere, an important feature of the Earth's interior that was the object of our Atlantic Ocean study.〈/span〉
