    Notes: Summary The general problem of determining the figure of the earth leads to the solution of the geodetic boundary value problem. By its discrete approximation we obtain ‘the discrete disturbing potential’ that maintains all properties of the original problem. Thus, the discrete approximation of the disturbing potential can be used in studying the behaviour of the earth's gravity field outside the disturbing masses. The deflections of the vertical are one of the quantities describing the behaviour of the earth's gravity field. A method for their computation from the discrete solution of the geodetic boundary value problem is put forth and estimates for its accuracy are given.

    Notes: Summary In the present paper the gravity field of the earth in the neighbourhood of the local disturbing masses is studied. The object of the method presented consists of the approximation of the disturbing potentialT h , which fulfils Laplace's equation outside disturbing masses, on the earth's surface the fundamental boundary value condition of gravity and in infinity it is to be regular by the approximation of the disturbing potential (or by the discrete disturbing potential)T h , which fulfils the respective finite difference approximation of Laplace's equation and the boundary value conditions in infinity and on the earth's surface. It is also shown that the approximation of the disturbing potentialT h has the same properties as the disturbing potentialT. The method under consideration will be derived quite generally without any hypothesis about the distribution of the mass between the earth's surface and the geoid. It commences from the gravity data related to the earth's surface only-from the given geodetic measurements.

    Notes: We conducted a transplant experiment to elucidate the effects of different levels of grazing pressure, nutrient availability, especially phosphorus, and the impact of viruses on the changes in the structure of bacterioplankton assemblage in a meso-eutrophic reservoir. A sample taken from the nutrient-rich inflow part of the reservoir was size-fractionated and incubated in dialysis bags in both inflow and dam area. The structure of bacterial assemblage was examined by fluorescence in situ hybridization using oligonucleotide probes with different levels of specificity. In terms of the relative proportions of different bacterial groups, we found very few significant changes in the bacterioplankton composition after transplanting the treatments to the nutrient-poor dam area. However, we observed marked shifts in morphology and biomass towards the development of filaments, flocs and “vibrio-like” morphotypes of selected probe-defined groups of bacteria induced by increased grazing pressure. Despite the very high abundances of viruses in all the treatments, their effects on bacterioplankton were rather negligible.

    Notes: The line-intercept method was adopted for quantification of aquatic filamentous microorganisms. The cumulative length of filaments in a sample is calculated from the number of intercepts between filaments and test bars of known length. The product of the cumulative length and of filament diameter reveals the total biovolume of filaments. The method is suitable for reliable and statistically correct quantification of long or dense filaments immeasurable individually. The combination of epifluorescent microscopy and image analysis speeds up sample processing and supports differentiation between heterotrophic and autotrophic filaments. Parallel testing of the line-intercept method in Utermöhl sedimentation chambers revealed tight correlation (r2=0.89) of both methods for cyanobacterial filaments.

    Notes: We documented permanent presence of heterotrophic filaments in three acidified lakes (pH〈5) in the Bohemian Forest. Due to acidification, crustacean zooplankton were absent in all but one lake. In terms of carbon flow, microorganisms were thus almost exclusive players in the pelagic food webs. Variety of extremely long (〉100 μm) heterotrophic filaments occurred in the lakes. The filaments usually accounted for 〉50% of total heterotrophic microbial biomass in the pelagic zone (medians of the total biomass: 82–108 μg C l−1), except for anoxic bottom layers and for episodic appearance of Daphnia longispina in one lake. Seasonal filament formation was mainly induced and maintained by grazing of mixotrophic flagellates (Dinobryon spp.) on small unicellular bacteria in the absence of cladoceran filtration. The filaments were less active than unicellular suspended bacteria. Fluorescence in situ hybridisation revealed that usually 〈50% of total bacteria were targeted with oligonucleotide probes for Eubacteria.

    Notes: 1. Phosphorus limitation was studied along the eutrophic, canyon-type Římov reservoir (Czech Republic) during a spring phytoplankton bloom. Concentration of soluble reactive phosphorus (SRP), C:P molar ratio in seston, extracellular alkaline phosphatase activity (APA), and P limitation (bioassay) were used as indices for phosphorus deficiency in the phytoplankton.2. SRP, C:P, APA, and P limitation indicated a moderate P deficiency in the downstream, but not upper, part of the reservoir.3. Significant correlations between these parameters were found in the downstream part. Chlorophyll a concentration correlated with APA and P limitation in the upper part.4. APA was significantly enhanced in the phosphorus-deficient phytoplankton. However, APA was apparently not related to total biomass or species composition of the phytoplankton.5. Generally, APA was closely correlated with pH in the reservoir. However, extracellular alkaline phosphatases, with a pH optimum above 9.0, were induced and active only during the phytoplankton bloom, whereas low background activity of extracellular phosphatases was found at low chlorophyll a concentrations (winter, clear-water phase).

    Notes: 〈list xml:id="l1" style="custom"〉1 Seston elemental composition (C, N, P) and the orthophosphate uptake rate constant, k(upt), were measured in the epilimnion of the eutrophic Římov Reservoir in the period March-August 1989.2 Seasonal series of log C/P, log N/P and log [k(upt)] values were highly correlated (r2 in the range 0.84–0.93), but, based on literature-derived criteria, k(upt) values indicated stronger P deficiency than did the C/P values.3 The results showed moderate to strong phosphorus deficiency in summer, moderate deficiency at the end of the spring peak of phytoplankton, and no deficiency early in the spring or during the spring clear-water period.
