    Notes: Abstract Cauliflower (Brassica oleracea L. ssp. botrytis) protoplasts with Ogura male sterile and fertile B. oleracea cytoplasms were fused, producing plants with an array of organellar types. Plants with Ogura mitochondria were male sterile; those with B. oleracea chloroplasts were cold tolerant. In some fusions, unfused parental protoplasts were eliminated by double inactivation with iodoacetate and gamma-irradiation; in others, fused protoplasts were physically isolated by micromanipulation or by cell sorting. Double inactivation fusions produced the most plants, including many which were male sterile, female fertile, cold tolerant and diploid.

D'Arcy, sowie auf der Kreuzung L. esculentum Mill. und L. pennellii, war deutlich grösser als auf L. esculentum. Das mechanische Verfangen der Läuse war nicht der Hauptgrund der Resistenz von L. pennellii. Wenige tote Läuse wurden in dem klebrigen Sekret der Typus IV Trichome gefunden. Auf L. pennellii siedelten sich die Läuse in viel geringerer Zahl an als auf L. esculentum. Dies führte zum Schluss, dass Verhungern eine der Ursachen der hohen Mortalität der Läuse war. Im Vergleich zum Saugverhalten auf L. esculentum war das Saugverhalten auf L. pennellii, wie auch auf F1, durch Folgendes gekennzeichnet 1) Verspätung des ersten Stichversuchs, 2) Verminderung der Stichversuche pro Zeiteinheit und 3) Verminderung des Zeitanteils, der zum Saugen verwendet wurde. Die Entfernung des Sekrets der Typus IV Trichomen auf L. pennellii verursachte 1) eine kürzere Zeitspanne vor dem ersten Stichversuch, 2) eine Vergrösserung der Anzahl Stichversuche pro Zeiteinheit, 3) eine Verlängerung der Saugzeit. Die Uebertragung des Sekretes von L. pennellii auf Blätter von L. esculentum verbesserte deren Resistenz gegen Blattläuse gemessen mit den genannten drei Kriterien.
    Notes: Abstract Mortality of the potato aphid, Macrosiphum euphorbiae (Thomas), on Lycopersicon pennellii (Corr.) D'Arcy and its F1 hybrid with Lycopersicon esculentum Mill. was significantly greater than that on L. esculentum. Physical entrapment was not the sole mechanism of resistance in L. pennellii since few late instar aphids were found trapped in the sticky glandular exudate of the type IV trichomes; entrapment could, however, affect survival of early instars. Aphid settling on L. pennellii was dramatically less than that on L. esculentum, suggesting that starvation may have contributed to high mortality. Compared to L. esculentum, aphid feeding behavior on L. pennellii and the F1 was characterized by a delay in the time to first probe, a reduction in the number of probes, and a decrease in the total proportion of time spent feeding. Removal of the glandular exudate of the type IV trichomes from L. pennellii resulted in a decrease in preprobe time and an increase in both the number of probes and the percent of time spent probing. Transfer of glandular trichome exudate of L. pennellii to leaflets of L. esculentum resulted in an increase in resistance as measured by these three parameters.

    Notes: Abstract Mature green tomato fruit (Lycopersicon esculentum Mill) of cv. ‘Rutgers’ and the line ‘Alcobaca-red’ were vacuum infiltrated with solutions of polyamines, their precursors and metabolites, and other compounds which might affect ripening and/or storage duration. Putrescine (1,4-diaminobutane), spermidine, spermine, diaminopropane, γ-aminobutyric acid and methionine were found to increase the storage life of these fruit after vacuum infiltration of the test compounds and storage of fruit in darkness. Polyamines probably play a role in the normal ripening/overripening process and may prove commercially valuable in the extension of fruit shelf life.

    Notes: Abstract Two novel tandemly-oriented open reading frames (ORFs) with homology to alcohol dehydrogenase (ADH) were isolated from tomato. The predicted amino acid composition for each of the two tandem adh genes indicates the presence of 22 and 21, respectively, of 22 amino acids conserved in ADH proteins from plants and animals. However, comparison to known plant adh genes reveals a significantly lower similarity indicating that they belong to a novel class of ADHs. According to mapping data, the adh-homologous ORFs do not represent either of the previously studied adh1 or adh2 genes of tomato. The tandem genes, termed adh3a and adh3b, mapped to a distal region of the long arm of chromosome 4, unlike adh1, which maps closer to the centromere. Adh3a and adh3b have over 90% similarity to each other at the nucleotide and putative peptide levels. The adh3a gene has ten exons and nine introns with the transcription initiation site 57 bp upstream of the translation start. A putative TATA box and polyadenylation site have been identified. Adh3a is transcribed and, according to cDNA sequence analysis, fully processed in the late stages of anther development. According to transformation analysis, tissue-specific regulatory elements reside within the -448 to +724 region. The termination codon of adh3a is separated from the putative adh3b translation start site by 789 bp of intervening sequence. The 5′ untranscribed sequences of each gene contain a stretch of 68 bp with 78% similarity. Within this stretch are sequences which are homologous to sequences found in anaerobically-induced or pollenexpressed genes from various plant species.

    Notes: Abstract Factors affecting the division of cells derived from leaf and cotyledon protoplasts from Brassica oleracea L. var. italica (‘Green Comet’ hybrid broccoli) were examined to optimize conditions for plant regeneration and to determine whether there was a genetic basis for improved regeneration from protoplasts derived from plants previously regenerated from tissue cultures [15]. When leaf protoplasts from different plants grown from hybrid seed were isolated and cultured simultaneously, division efficiencies of 1–95% were obtained. Cells from some plants showed high division efficiencies in consecutive experiments while cells from other plants had consistently low division rates. More plants from hybrid seed gave high division efficiencies when cotyledon protoplasts were used. However, cotyledon or leaf protoplasts from selfed progeny of regenerated plants produced more vigorous calli and more shoots than protoplasts from hybrid seed. These results suggest that there may be a genetic component to the increased totipotency of Brassica oleracea protoplasts.

    Notes: Abstract Settling of the potato aphid,Macrosiphum euphorbiae, on feeding membranes was deterred by methanolic leaf rinses ofLycopersicon pennellii, or of its F1 with tomato,L. esculentum. The active compounds in theL. pennellii rinsates were identified as 2,3,4-tri-O-acylglucoses bearing short to medium chain length fatty acids. These compounds are localized in the glandular exudate of the type IV trichomes and may accumulate to levels in excess of 400 μg/cm2. In choice assays, purified glucose esters fromL. pennellii reduced aphid settling at concentrations as low as 25 μg/cm2; at concentrations of 150 μg/cm2 or more, all aphids avoided treated areas. Glucose esters were also active in deterring aphid settling in no-choice assays. At 100 μg/ cm2, these esters resulted in increased levels of mortality after 48 hr.

    Notes: Abstract Acylsugars exuded from type IV trichomes mediate the multiple pest resistance found in the wild tomato species, Lycopersicon pennellii. A marker-assisted selection breeding program was used to attempt the transfer of the ability to accumulate acylsugars to cultivated tomato. RFLP and PCR-based markers were used through three backcross generations to select plants containing 5 target regions associated by QTL analysis with acylsugar accumulation. The BC1F1 plant selected possessed all 5 target regions and accumulated acylsugars at a moderate level similar to that of the interspecific F1 control. The BC2F1 and BC3F1 selections contained complementary subsets of the 5 target regions and did not accumulate acylsugars. BC3F1 plants with complementary subsets of the 5 target regions were intermated to produce populations segregating for the 5 target regions. From 1000 BC3F1-intermated plants, three plants were found which accumulated acylsugars at low levels and contained 3 to 5 of the target regions. The recovery of acylsugar accumulation in progeny of the intermated BC3F1 plants supports the involvement of at least some of the 5 target regions in acylsugar biosynthesis. However, since the levels of acylsugars accumulated by these plants were lower than that of the interspecific F1, it is likely that another, as of yet unidentified, region is necessary for accumulation of higher levels of acylsugars.
