    Notes: The synchrotron Laue technique has been applied to high-resolution structure refinement of the ribotoxin, restrictocin [Yang & Moffat (1996). Structure, 4, 837–852]. By employing carefully designed data-collection strategies and the data-reduction algorithms incorporated in the software system LaueView [Ren & Moffat (1995a). J. Appl. Cryst. 28, 461–481; Ren & Moffat (1995b). J. Appl. Cryst. 28, 482–493], a set of high-resolution Laue data with a completeness and accuracy comparable to excellent monochromatic data was obtained. Through detailed comparison with the monochromatic data and electron-density maps derived from the Laue data, optimum data-collection and reduction strategies were identified and the application of Laue diffraction techniques to conventional crystallographic refinement was demonstrated.

    Notes: The A1 station on the CHESS wiggler beamline has been the workhorse for most macromolecular crystallographic experiments. This station is equipped with a fixed energy focusing germanium (111) monochromator and a focusing total reflection mirror. Our macromolecular crystallographers made full use of the high flux of more than 1012 photons/s/mm2 and the stable beam conditions, both in position and energy resolution. As a result, the A1 station was heavily oversubscribed. CHESS is presently expanding its capabilities and a new diffraction station for macromolecular crystallography is under construction. This beamline will be powered by a 24-pole hybrid permanent magnet wiggler with a critical energy of 25 keV. A focusing monochromator, which handles a specific heat load of 10 W/mm2, will have a range of tunability which covers all relevant absorption edges from 7 to 15 keV using a Ge(111) crystal. The energy resolution and the focusing properties remain constant within a factor of 2 over the entire tunability range. We expect a brilliance of about 1013 photons/s/mm2/mrad2/0.1% bandpass. The diffraction station will be equipped with an oscillation camera which can be used with x-ray film of 5×5 or 8×10 in. size or alternatively with Kodak storage phosphors. A wide variety of clamp-on accessories, like crystal coolers, fast shutters, helium pathways, polarimeter, etc. are available. The station will contain a beampipe system, which can also be used for small angle scattering experiments with sample-to-detector distances of up to 3000 mm. The entire diffraction station, its control area, a biological preparation area, and a darkroom are to be embedded in a biological safety containment of the level BL3. This will allow diffraction studies of virulent strains of viruses and other biohazards, which could not previously be studied at synchrotron radiation sources before without causing major disruption to the normal laboratory procedure.

    Notes: We describe a single crystal microspectrophotometer for optical monitoring of protein crystals during time-resolved x-ray diffraction experiments. It is a single beam device with a diode array detector for simultaneous multiwavelength measurement of optical spectra. The time resolution of the microspectrophotometer is 2.5 ms, which matches the ms time resolution of the x-ray experiments obtained at existing synchrotron sources. Optical densities of single crystals can be reliably measured up to 2.5 OD, with the linear range extending to 2 OD. The tight focusing of the light beam allows monitoring of crystals as small as 30–50 μm, an important feature since successful reaction initiation and optical monitoring of crystals in time-resolved x-ray diffraction experiments often require very small crystals. The compact design of the microspectrophotometer and the long objective-to-crystal distance make it compatible with the crowded environment of time-resolved x-ray diffraction experiments.

    Notes: X-ray radiation damage of a lysozyme single crystal by an intense monochromatic beam from a third-generation radiation source at the Advanced Photon Source has been studied. The results show that primary radiation damage is linearly dependent on the X-ray dose even when the crystal is at cryogenic temperatures. The existence of an upper limit for the primary radiation damage was observed. Above the threshold of approximately 1 × 107 Gy, excessive damage of the crystal develops which is interpreted as the onset of secondary and/or tertiary radiation damage. This upper limit of X-ray dose is compared with Henderson's limit [Henderson (1990). Proc. R. Soc. London, B241, 6–8], and its implication for the amount of useful X-ray diffraction data that can be obtained for crystals of a given scattering power is also discussed.

    Notes: A renewed interest in the Laue diffraction technique has been brought about by the development of new, more intense and brilliant synchrotron sources along with their insertion devices such as wigglers and undulators, and by the prospect of using these sources to study structural dynamics by time-resolved crystallography. Theoretical studies during the past decade have identified unique features of the polychromatic diffraction geometry and greatly improved our understanding of the Laue method. This led to innovative approaches to Laue data processing and its software implementation. Most of the problems in Laue data processing, considered for a long time to limit the applicability of the technique, have been solved. Significant advances have also been made in the development of synchrotron sources, beamline optics and instrumentation, and the X-ray detectors. Static Laue experiments yield structure amplitudes that equal those from monochromatic data in quality. When coupled with careful consideration of data-collection strategies and reaction initiation in crystals, a series of successful time-resolved Laue experiments on biological systems have been conducted. These have revealed information on structural dynamics inaccessible to any other conventional diffraction method. These static and time-resolved experiments demonstrate that the Laue method is coming of age. They also suggest avenues for future improvements: a correct treatment of finite mosaic spread and the associated energy width of Laue spots; consideration of diffuse scattering; and determination of intermediate structures in time-resolved experiments in which those intermediates do not attain a high concentration.

    Notes: Wavelength normalization is an essential part of processing of Laue X-ray diffraction data and is critically important for deriving accurate structure-factor amplitudes. The results of wavelength normalization for Laue data obtained in nanosecond time-resolved experiments at the ID09 beamline at the European Synchrotron Radiation Facility, Grenoble, France, are presented. Several wiggler and undulator insertion devices with complex spectra were used. The results show that even in the most challenging cases, such as wiggler/undulator tandems or single-line undulators, accurate wavelength normalization does not require unusually redundant Laue data and can be accomplished using typical Laue data sets. Single-line undulator spectra derived from Laue data compare well with the measured incident X-ray spectra. Successful wavelength normalization of the undulator data was also confirmed by the observed signal in nanosecond time-resolved experiments. Single-line undulators, which are attractive for time-resolved experiments due to their high peak intensity and low polychromatic background, are compared with wigglers, based on data obtained on the same crystal.
