    Description: 〈p〉The ability of naturally occurring proteins to change conformation in response to environmental changes is critical to biological function. Although there have been advances in the de novo design of stable proteins with a single, deep free-energy minimum, the design of conformational switches remains challenging. We present a general strategy to design pH-responsive protein conformational changes by precisely preorganizing histidine residues in buried hydrogen-bond networks. We design homotrimers and heterodimers that are stable above pH 6.5 but undergo cooperative, large-scale conformational changes when the pH is lowered and electrostatic and steric repulsion builds up as the network histidine residues become protonated. The transition pH and cooperativity can be controlled through the number of histidine-containing networks and the strength of the surrounding hydrophobic interactions. Upon disassembly, the designed proteins disrupt lipid membranes both in vitro and after being endocytosed in mammalian cells. Our results demonstrate that environmentally triggered conformational changes can now be programmed by de novo protein design.〈/p〉

    Description: The performance of a QPSK (quadrature phase-shift keying) lock detector is described, taking into account the degradation due to carrier phase jitter. Such an analysis is necessary for accurate performance prediction purposes in scenarios where both the loop SNR is low and the estimation period is short. The derived formulas are applicable to several QPSK loops and are verified using computer simulations.

    Description: When a digital phase-locked loop with a long loop update time tracks a signal with high Doppler, the demodualtion losses due to frequency mismatch can become very significant. One way of reducing these Doppler-related losses is to compensate for the Doppler effect using some kind of frequency-rate estimator. The performance of the fixed-window least-squares estimator and the Kalman filter is investigated; several Doppler compensating techniques are proposed. It is shown that the variance of the frequency estimator can be made as small as desired, and with this, the Doppler effect can be effectively compensated. The remaining demodulation losses due to phase jitter in the loop can be less than 0.1 dB.

    Description: Four different arraying schemes that can be used by the Deep Space Network are functionally discussed and compared. These include symbol stream combining (SSC), baseband combining (BC), carrier arraying (CA), and full spectrum combining (FSC). In addition, sibeband aiding (SA) is also included and compared even though it is not an arraying scheme, since it uses a single antenna. Moreover, combinations of these schemes are discussed, such as carrier arraying with sideband aiding and baseband combining (CA/SA/BC) or carrier arraying with symbol stream combining (CA/SSC). Complexity versus performance is traded off and the benefits to the reception of existing spacecraft signals are discussed. Recommendations are made as to the best techniques for particular configurations.

    Description: The results are presented for an investigation of ground antenna pointing errors which are caused by fluctuations of the receiver AGC signal due to thermal noise and a spinning spacecraft. Transient responses and steady-state errors and losses are estimated using models of the digital Conscan (conical scan) loop, the FFT, and antenna characteristics. Simulation results are given for the on-going Voyager mission and for the upcoming Galileo mission, which includes a spinning spacecraft. The simulation predicts a 1 sigma pointing error of 0.5 to 2.0 mdeg for Voyager, assuming an AGC loop SNR of 35 to 30 dB with a scan period varying from 128 to 32 sec, respectively. This prediction is in agreement with the DSS 14 antenna Conscan performance of 1.7 mdeg for 32 sec scans as reported in earlier studies. The simulation of Galileo predicts 1 mdeg error with a 128 sec scan and 4 mdeg with a 32 sec scan under similar AGC conditions.

    Description: The subcarrier demodulation digital loop is part of the baseband assembly. The subcarrier demodulator is a fourth-order Costas-type loop corresponding to a type 2 analog loop in terms of steady state response. The expected value and the variance of the error signal are determined as functions of the input SNR. A Nyquist sampling rate of the input signal is assumed. From the integro-difference equations a mixed s/z domain block diagram is obtained. From the loop's transfer function a set of gains for the loop filter is obtained. Also, a set of state equations is presented for future reference. Finally, the noise-equivalent bandwidths are calculated for normalized computation times of 0, 0.25 and 0.5. The subcarrier demodulator analyzed tracks a parabolic phase input with finite steady state error. Since at each update instant the loop gains are adjusted to compensate for the variations in SNR of the input signal, the noise-equivalent bandwidth is maintained constant.

    Description: An s/z hybrid model for a general phase locked loop is proposed. The impact of the loop filter on the stability, gain margin, noise equivalent bandwidth, steady state error and time response is investigated. A specific digital filter is selected which maximizes the overall gain margin of the loop. This filter can have any desired number of integrators. Three integrators are sufficient in order to track a phase jerk with zero steady state error at loop update instants. This filter has one zero near z = 1.0 for each integrator. The total number of poles of the filter is equal to the number of integrators plus two.

Each noncatastrophic state of the spacecraft is indicated by the transmission of a specific signal to the ground station.     Description: Cost efficiency is becoming a major driver in future space missions. Because of the constraints on total cost, including design, implementation, and operation, future spacecraft are limited in terms of their size power and complexity. Consequently, it is expected that future missions will operate on marginal space-to-ground communication links that, in turn, can pose an additional risk on the successful scientific data return of these missions. For low data-rate and low downlink-margin missions, the buffering of the telemetry signal for further signal processing to improve data return is a possible strategy; it has been adopted for the Galileo S-band mission. This article describes techniques used for postprocessing of buffered telemetry signal segments (called gaps) to recover data lost during acquisition and resynchronization. Two methods, one for a closed-loop and the other one for an open-loop configuration, are discussed in this article. Both of them can be used in either forward or backward processing of signal segments, depending on where a gap is specifically situated in a pass.
