Isolation and characterization of an amyloplast envelope-enriched fraction from immature maize endosperm.A 10000–100000 g pellet obtained by centrifugation of homogenates from immature (25 days after pollination) de-embryonated maize (Zea mays L., cv. W64A-normal and a typical hybrid) kernels was further fractionated by sedimentation on discontinuous sucrose density gradients. Particles with the highest carotenoid content (0.68% by weight carotenoids based upon total lipid) sedimented at densities of 1.083-1.106 g ml-1, coincident with the plastid envelope marker enzyme, galacto-syltransferase (EC 2.4.1.46). Lipids extracted from the carotenoid-rich fraction were mainly digalactosyldiacylglycerols, monogalactosyldiacylglycerols, phosphatidylcholines, phosphatidylinositols and phosphatidylglycerols, in order of molar abundance. With increasing particle density (〉1.106 g ml1) the phospholipid and neutral lipid content increased, and the proportion of carotenoids and galactolipids decreased. Electron micrographs of the carotenoid-rich fraction revealed vesicles ranging in size from 〈 0.1 to 0.5 um, as well as smaller granular membranes. The carotenoid-rich membrane fraction was progressively more difficult to isolate as the endosperm matured, and freezing the immature endosperm prevented subsequent isolation. The lipid and enzyme composition and ultrastructural characteristics of the isolated fraction suggest that it is composed of amyloplast envelope vesicles.

    Notes: Imbibing ‘Grand Rapids’ lettuce (Lactuca saliva L.) seeds take up 14C-kinetin, and metabolize this cytokinin to the 5′-nucleotide. The identity of the labeled nucleotide in seed extracts was verified by Sephadex LH-20 column chromatography, paper and thin layer chromatography, and high voltage paper electrophoresis.Incubations with kinetin in the presence of abscisic acid lead to an apparent specific inhibition of kinetin nucleotide formation. ABA has no effect on kinetin uptake, and does not inhibit kinetin nucleotide synthesis in vitro by a cell-free preparation from lettuce seeds. Additionally, ABA does not inhibit adenylate synthesis from exogenously supplied adenine. These results represent a specific cytokinin-ABA interaction, which might play a significant role in the hormonal regulation of lettuce seed germination.

    Notes: Exogenously applied 8-14C-kinetin is rapidly taken up by seeds of lettuce (Lactuca sativa L. cv. Grand Rapids). Radioactive metabolites were extracted and purified by solvent fractionation, column and paper chromatography. The primary metabolite was identified as the 9-riboside-5′-monophosphate. As germination proceeds, some kinetin is released from this bound storage form, giving a maximum level of free kinetin at 12 hours after imbibition. After this time the concentration of ribotide increases while the concentration of free base decreases. Other metabolites are the 9-riboside, AMP and IMP.It is suggested that a required amount of free base cytokinin is necessary by 12 hours after imbibition. This concentration of free cytokinin may act as a physiological trigger for later events during germination.

    Notes: A plasmid, pWEH1, was constructed containing a fusion of the DNA encoding the signal sequence of the Escherichia coli outer-membrane protein A to the 5′-end of a glyceraldehyde-3-phosphate dehydrogenase cDNA from Ricinus communis. When expressed in E. coli, the fusion protein was secreted by the normal membrane-potential-dependent pathway. Processing by signal peptidase was inhibited by low concentrations of phenethyl alcohol. Quantitative cell fractionation was used to show that the mature plant protein was associated with the bacterial outer membrane. The protein could not be released from the membrane by washing with alkaline sodium carbonate. Radioactivity from [U-14C]-palmitate was incorporated into the heterologous protein. These results suggest that the sequence of this normally cytoplasmic enzyme contains a cryptic lipid-modification site, and the combination of a signal sequence plus a lipid-modification sequence results in specific targeting to the bacterial outer membrane.
