    Notes: Campylobacter jejuni is a significant cause of bacterial enteritis in humans. Three of seven C. jejuni isolates examined were found to contain fragmented 23S rRNA. The occurrence of fragmented 23S rRNA correlated with the presence of an intervening sequence (IVS) within the 23S rRNA genes. The IVS is 157 nucleotides in length and replaces an eight nucleotide sequence in the 23S rRNA genes of C. jejuni isolates that contain intact 23S rRNA. The two ends of the IVS share 31 bases of complementarity that could form a stem-loop structure. Fragmentation of the 23S ribosomal RNA results from the excision of the IVS from the transcribed RNA; the 3’ cleavage site maps within the putative stem-loop formed by the IVS. Southern hybridization analysis revealed that the IVS is not present in the genomes of isolates that contain intact 23S rRNA, suggesting that the IVS is not derived from Campylobacter chromosomal sequences. The C. jejuni IVS is located at a position analogous to that of the IVSs found in both Salmonella and Yersinia spp.

    Notes: Summary The maximum specific growth rates (μmax) of 2 μ-plasmid-free ([cir°]) segregants of three haploid and one diploid strain of Saccharomyces cerevisiae have been determined and compared with the μmax of their 2 μ-plasmid-containing ([cir +]) progenitors. Two classes of [cir°] strains have been examined: those induced by transformation with a 2 μ-based recombinant plasmid according to the method of Dobson et al. (1980) and those isolated as spontaneous [cir°] segregants from glucose-limited continuous cultures. The μmax of the spontaneous [cir°] segregants was not found to differ significantly from that of their [cir +] parents. In all cases, however, the induced [cir°] strains had a μmax which was significantly less than that of their [cir +] counterparts. This effect was particularly marked in the case of the diploid strain where a 34% reduction in μmax was observed. The implications of these results are discussed in terms of the effect of the transformation process on host yeast cells.

    Notes: Abstract The empirical Cole-Cole distribution is an analytical three parameter model of the relaxation spectra that provides accurate fits to experimental dynamic viscosity data for many systems of commercial linear backbone flexible polymers. We demonstrate that for disparate systems of polyethylenes, the three Cole-Cole model parameters have simple power law relationships to moments of the molecular weight distribution enabling direct molecular interpretation of the mechanical relaxation spectrum. A simple relationship between the Cole-Cole distribution and the Cross model for the non-linear flow curve can be deduced utilizing the empirical Cox-Merz rule. Accurately representing the linear viscoelastic material functions with empirical analytical relaxation spectra containing relatively few fitting parameters that can be readily interpreted is a major advance in polymer characterization. The three Cole-Cole parameters effectively replace single point material characterizations such as melt flow index. Development of higher resolution polymer characterization methods is imperative with the advent of metallocene catalyst technology, which enables the molecular weight and backbone architecture to be carefully controlled.

    Notes: Summary The designation of the yeast 2 μ circle as a “selfish” DNA molecule has been confirmed by demonstrating that the plasmid is lost with exponential kinetics from haploid yeast populations grown in continuous culture. We show that plasmid-free yeast cells have a growth rate advantage of some 1.5%–3% over their plasmid-containing counterparts. This finding makes the ubiquity of this selfish DNA in yeast strains puzzling. Two other factors probably account for its survival. First, the rate of plasmid loss was reduced by allowing haploid populations to enter stationary phase periodically. Second, it was not possible to isolate a plasmid-free segregant from a diploid yeast strain. Competition experiments demonstrated that stability in a diploid is conferred at the level of segregation and that plasmid-free diploid cells are at a selective advantage compared with their plasmid-containing counterparts. Yeast cells in nature are usually homothallic and must frequently pass through both diploid and stationary phases. The 2 μ plasmid appears to have evolved a survival strategy which exploits these two features of its host's life cycle.

    Notes: Experiments measuring the orientation angle and birefringence in startup and double-step strain rate flows were conducted on a 3.0 wt % 8.42 × 106 molecular weight polystyrene solution in a Couette flow cell. A phase-modulated flow birefringence apparatus was used to noninvasively probe the sample. Upon startup from rest, the orientation angle undershoots its final steady-state value, as seen by earlier investigators. When the shear rate undergoes a step increase from one nonzero value to another, the amplitude of this undershoot is decreased. However, a more significant effect is a shorter time scale overshoot in the orientation angle that is highly counterintuitive in the sense that an increase of shear rate initially produces a rotation of chain segments away from the flow direction. Similarly, a step decrease in shear rate yields an initial transient rotation toward the flow direction. In both cases, the height of the peaks depends upon the magnitude of the shear rate jump, and the width of the peaks is a function of the final shear rate. The longer time transients in the startup and step increase experiments reflect an apparent change in the relaxation time for segment orientation, which we tentatively attribute to a combination of tube dilation and convective constraint release. The shorter time scale over- and undershoots in the orientation angle appear to be qualitatively explained by considering the differences in extension or contraction of segments along the polymer chain. © 1998 John Wiley & Sons, Inc.     Notes: The glucose oxidase gene (god) from Aspergillus niger was expressed in Hansenula polymorpha using the methanol oxidase promoter and transcription termination region and the MF-α leader sequence from Saccharomyces cerevisiae to direct secretion. The expression cassette was cloned into the S. cerevisiae vector YEp13 and used to transform H. polymorpha strain A16. In the initial transformants plasmid replication was unstable, but was stabilized by a growth regime consisting of alternating cycles of selective and non-selective growth. The stabilized strain was grown to high cell density by fed-batch fermentation. Upon induction of the MOX promoter, glucose oxidase synthesis was initiated. At the end of the fermentation, the culture density was 76 g dry weight/1 and 108 IU/ml (0·5 g/l or 0·65% dry weight) glucose oxidase was found in the culture medium; a further 86 IU/ml (0·43 g/l or 0·56% dry weight) was recovered from the cell lysate. A plate assay was used to monitor glucose oxidase levels in individual colonies. This was then used to isolate mutants which showed abnormal regulation of god expression or which showed an altered pattern of secretion. One mutant, which showed increased production of glucose oxidase, was grown to high cell density by fed-batch fermentation (100·6 g/l) and produced 445 IU/ml (2·25 g/l or 2·2% dry weight) extracellularly and 76 IU/ml (0·38 g/l or 0·4% dry weight) intracellularly. The mutant thus not only increased total production but exported 83% of the total enzyme made compared to 55% in the parent strain.
