    Description: We used a controlled CO2 perturbation experiment to test hypotheses about changes in diversity, composition and structure of soft-bottom intertidal macrobenthic assemblages, under realistic and locally relevant scenarios of seawater acidification. Patches of undisturbed sediment were collected from 2 types of intertidal sedimentary habitat in the Ria Formosa coastal lagoon (South Portugal) and exposed to 2 levels of seawater acidification (pH reduced by 0.3 and 0.6 units) and 1 unmanipulated (control) level. After 75 d the assemblages differed significantly between the 2 types of sediment and between field controls and the ex situ treatments, but not among the 3 pH levels tested. The naturally high values of total alkalinity buffered seawater from the changes imposed on carbonate chemistry and may have contributed to offsetting acidification at the local scale. Observed differences on biota were strongly related to the organic matter content and grain-size of the sediments, particularly to the fractions of medium and coarse sand. Soft-bottom intertidal macrofauna was significantly affected by the stress of being held in an artificial environment, but not by CO2-induced seawater acidification. Given the previously observed variations in the sensitivities of marine organisms to seawater acidification, direct extrapolations of the present findings to different regions or other types of assemblages do not seem advisable. However, the contribution of ex situ studies to the assessment of ecosystem-level responses to environmental disturbances could generally be improved by incorporating adequate field controls in the experimental design.

    Description: Sites with naturally high CO2 conditions provide unique opportunities to forecast the vulnerability of coastal ecosystems to ocean acidification, by studying the biological responses and potential adaptations to this increased environmental variability. In this study, we investigated the bivalve Ervilia castanea in coastal sandy sediments at reference sites and at volcanic CO2 seeps off the Azores, where the pH of bottom waters ranged from average oceanic levels of 8.2, along gradients, down to 6.81, in carbonated seawater at the seeps. The bivalve population structure changed markedly at the seeps. Large individuals became less abundant as seawater CO2 levels rose and were completely absent from the most acidified sites. In contrast, small bivalves were most abundant at the CO2 seeps. We propose that larvae can settle and initially live in high abundances under elevated CO2 levels, but that high rates of post-settlement dispersal and/or mortality occur. Ervilia castanea were susceptible to elevated CO2 levels and these effects were consistently associated with lower food supplies. This raises concerns about the effects of ocean acidification on the brood stock of this species and other bivalve molluscs with similar life history traits.

    Notes: Summary In order to delineate the spectrum and the relative abundance of β-globin gene defects causing thalassaemia in the Portuguese population, a representative sample was analysed including 51 β-thalassaemia carriers along with 26 patients representing different clinical phenotypes. Seven mutations were identified, four of which [codon 39 (C→T), 39%; intervening sequence (IVS)1 nucleotide (nt) 1 (G→A), 26%; IVS1 nt 110 (G→A), 17%; IVS1 nt6 (T→C), 15%] account for 97% of 93 β-thalassaemia chromosomes. Two previously undescribed mutations, namely a C→T substitution at position — 90 in the proximal CACCC box, and the deletion of nucleotides 4 and 5 (AG) in IVS 2 were identified. The uncommon, though ubiquitous, G→T transversion at codon 121 was found once upon haplotype V. Direct prenatal diagnosis can be offered to 95% of couples at risk of bearing a thalassaemic child.

    Notes: Four sciaenid species Johnieops dussumieri, Kathala axillaris, Pennahia macropthalmus and Otolithes ruber were analysed electrophoretically for genetic variation at 18 loci (16 in P. macropthalmus and O. ruber). Twelve loci were polymorphic in J. dussumieri, 10 in K. axillaris, three in P. macropthalmus and 12 in O. ruber (P〈0·99). Average heterozygosity ranged from 0·033 ± 0·100 to 0·070 ± 0·122. The allele frequencies of 14 loci were used to estimate Nei's genetic distance (D̄). The D̄ values ranged from 0·334 to 0·612. Three isozyme loci (LDH-B*, MDH-2* and G3PDH-1*) were found to be the most reliable species-specific markers.

    Notes: We present experimental results on the growth and structure of Fe overlayers deposited on Cu84Al16(100) and discuss the correlation between the structural and magnetic properties of this system. Fe films 1–6 monolayers (ML) thick were grown under molecular beam epitaxy conditions onto the clean substrate at 160 K. Electron diffraction was applied to investigate the structure of the Fe films. The magnetic properties were investigated in situ by surface magneto-optical Kerr effect in the longitudinal geometry. Our results show that the onset of in-plane ferromagnetism around 3.5 ML of Fe coincides with structural changes that suggest a transformation within the Fe films from a fcc-like (100) to a bcc-like (110) structure with increasing Fe thickness, starting from 2.5 ML. © 2001 American Institute of Physics.

    Notes: Rutilus alburnoides complex is a common and widely distributed Iberian cyprinid, whose natural populations include mainly diploid and triploid forms. The Guadiana populations of R. alburnoides were studied to determine whether habitat segregation and morphological differences exist between these forms. The ploidy level of each specimen was determined by measuring erythrocyte DNA content using flow cytometry. Evidence of spatial segregation between diploid males and the two female forms was found. Diploid males were best represented in the River Degebe, which was shallow, with higher temperatures (especially during the spring and summer), and silt and sandy substrate. Diploid females were found in deeper water, on steeper gradients and coarse substrata, while triploid females preferred higher current velocity and a high proportion of instream cover, especially during the spring. The ecological differences may reduce competitive interactions, and should promote a stable coexistence of the different forms. Morphological distinction between fish of different ploidy levels was not established, but differences were found between the males and females. Discriminant analysis allowed, with a 10% error, the separation of both sexes through six morphological characteristics that could be recorded in the field.
