Abstract: Since their development a decade ago, human induced pluripotent stem cells (iPSC) have revolutionized the study of human disease, given rise to regenerative medicine technologies, and provided exceptional opportunities for pharmacologic research. These cells provide an essentially unlimited supply of cell types that are difficult to obtain from patients, such as neurons or cardiomyocytes, or are difficult to maintain in primary cell culture. iPSC can be obtained from patients afflicted with a particular disease but, in combination with recently developed gene editing techniques, can also be modified to generate disease models. Moreover, the new techniques of 3 Dimensional printing and materials science facilitate the generation of organoids that can mirror organs under disease conditions. These properties make iPSC powerful tools to study how diseases develop and how they may be treated. In addition, iPSC can also be used to treat conditions in which the target cell population has been lost and such regenerative approaches hold great promise for currently untreatable diseases, including cardiac failure or photoreceptor degenerations.

Description / Table of Contents: Contents After deriving the basic equations for the magnetic vector potential for a simple arrangement consisting of a three-phase current sheet, produced on a cylindrical surface and an external shield tube, the vector potential and field equations are given. A complex screening factor is defined. It is shown that for calculation of multiregion systems the property of every cylindrical region is determined by a four-pole network. At the terminals of this network exist electrical and magnetic field intensities. The coaxial arrangement of several cylindrical regions leads to a recurrent network. As a practical example for the application of the theory the calculation of the electromagnetic field and of the power in a turbogenerator with superconducting field winding is demonstrated.

Description / Table of Contents: Contents Explained in the introduction are the problems and the facilitating assumptions on which the presented theory is based. This is followed by a brief survey of the required equations of the magnetic scalar potential, the magnetic vector potential and a special vector potential. Beginning with a double Fourier analysis of the inverse fundamental stator current density the magnetic fields, the eddy currents and the eddy current losses are calculated in three dimensions. Because the thickness of the outer screen is assumed as much larger than the depth of penetration the rotor can be considered as a solid cylinder.

Notes: Abstract The activity of Na−K-ATPase was measured in crude homogenates prepared from various organs (leg muscle, pincer muscle, heart, testes, digestive gland, hypodermis, gills 1–9) of shore crabs, Carcinus maenas L., acclimated to salinities ranging between 10 and 50‰ S (in steps of 10‰ S). In all salinities tested, Na−K-ATPase activity was highest in posterior gills 7–9 (10–12 μmol Pi mg protein-1 h-1), followed by anterior gills 1–6 (ca. 2.5 μmol Pi mg protein-1 h-1) and the other organs (in most cases far below 2μmol Pi mg protein-1 h-1). In gills only, Na−K-ATPase activity was salinity-dependent, with the highest values in the lowest salinities and vice versa. In gills 7–9, Na−K-ATPase activity was increased more than threefold following a reduction in salinity from 50 to 10‰ S. Na−K-ATPase activity, expressed as percentage of total ATPase activity, amounted to 60–80% in gills, about 60% in hypodermis and 20–40% in the other organs. Ouabain, a specific inhibitor of Na−K-ATPase activity, reduced serum osmolalities in crabs kept at 9–10‰ S only when injected into the hemolymph (1 and 5 · 10-5 M), but had no effect when dissolved in ambient water (10-4 M). The results obtained underline that crustacean gills are the main organs for ionic regulation, and confirm the hypothesis of the central role of the Na−K-ATPase in active Na uptake as the basic mechanism of hyperregulation in dilute media. Reduction of serum osmolalities following injection of ouabain into the hemolymph confirms previous reports on localization of the sodium pump in the basolateral parts of epithelial cells.

Notes: Summary The acute central haemodynamic and neuroendocrine effects of intravenous flosequinan were studied in a group of 10 patients with severe heart failure. Flosequinan improved cardiac output by a maximum of 1.59 l·min−1, it reduced pulmonary capillary wedge pressure by 11.9 mm Hg and it also caused a reduction in right atrial pressure by a maximum of 7.2 mm Hg. It tended to cause a fall in plasma adrenaline levels but not in plasma noradrenaline. There was little fall in blood pressure in response to flosequinan and no patient developed an adverse event. Intravenous flosequinan may be a useful candidate drug for controlled clinical studies in patients with severe heart failure.

Notes: Summary The clinical efficacy of a new slow release preparation of the calcium antagonist gallopamil was assessed in 20 patients by diary cards and treadmill exercise tests. A single blind phase of two week periods of placebo, gallopamil 100 mg o.d. and gallopamil 100 mg b.i.d., blinded to the patient, was followed in 18 patients by a double blind comparison of gallopamil 100 mg od versus 100 mg b.i.d. Angina frequency and trinitrin consumption per week were both significantly less on high dose (2.7 and 1.7) than on low dose (5.4 and 3.4) respectively. Treadmill total exercise time was longer on high dose (523 s) than low dose (449 s). Other exercise test parameters showed similar improvements on high dose treatment. Gallopamil was well tolerated. PR interval correlated best with plasma gallopamil level, while exercise test parameters correlated best with the log plasma level of its major metabolite nor-gallopamil.
