    Description: Whereas case rates for some childhood diseases (chickenpox) often vary according to an almost regular annual cycle, the incidence of more efficiently transmitted infections such as measles is more variable. Three hypotheses have been proposed to account for such fluctuations. (i) Irregular dynamics result from random shocks to systems with stable equilibria. (ii) The intrinsic dynamics correspond to biennial cycles that are subject to stochastic forcing. (iii) Aperiodic fluctuations are intrinsic to the epidemiology. Comparison of real world data and epidemiological models suggests that measles epidemics are inherently chaotic.     Description: Bacteria adapt to environmental stimuli by adjusting their transcriptomes in a complex manner, the full potential of which has yet to be established for any individual bacterial species. Here, we report the transcriptomes of Bacillus subtilis exposed to a wide range of environmental and nutritional conditions that the organism might encounter in nature. We comprehensively mapped transcription units (TUs) and grouped 2935 promoters into regulons controlled by various RNA polymerase sigma factors, accounting for ~66% of the observed variance in transcriptional activity.     Description: Adaptation of cells to environmental changes requires dynamic interactions between metabolic and regulatory networks, but studies typically address only one or a few layers of regulation. For nutritional shifts between two preferred carbon sources of Bacillus subtilis, we combined statistical and model-based data analyses of dynamic transcript, protein, and metabolite abundances and promoter activities. Adaptation to malate was rapid and primarily controlled posttranscriptionally compared with the slow, mainly transcriptionally controlled adaptation to glucose that entailed nearly half of the known transcription regulation network. Interactions across multiple levels of regulation were involved in adaptive changes that could also be achieved by controlling single genes.     Description: The molecular organization of eukaryotic nuclear volumes remains largely unexplored. Here we combined recent developments in cryo-electron tomography (cryo-ET) to produce three-dimensional snapshots of the HeLa cell nuclear periphery. Subtomogram averaging and classification of ribosomes revealed the native structure and organization of the cytoplasmic translation machinery. Analysis of a large dynamic structure-the nuclear pore complex-revealed variations detectable at the level of individual complexes. Cryo-ET was used to visualize previously elusive structures, such as nucleosome chains and the filaments of the nuclear lamina, in situ.     Description: The application of thermo-sensitive tracers is a promising technique for evaluating the thermal state of geothermal reservoirs. To extend the compound spectrum for hydrolyzable compounds to reservoir temperatures between 100 and 200 °C carboxamides were studied. The kinetic parameters of 17 self-synthesized amides were determined in hydrothermal batch and autoclave experiments. The influence of the molecular structure and the role of pH/pOH on hydrolysis kinetics were studied. Additionally, the thermal stabilities of the hydrolysis products were evaluated. The results demonstrate the high potential of tracers based on amide hydrolysis for use in medium enthalpy reservoirs.

    Notes: Measurements of the electric fields, E in the DIII-D tokamak divertor region [J. C. Luxon and L. G. Davis, Fusion Technology 8, Part 2A, 441 (1985)] are quantitatively consistent with recent computational modeling establishing that E×BT circulation is the main cause of changes in divertor plasmas with the direction of the toroidal magnetic field, BT. Extensive two-dimensional measurements of plasma potential in the DIII-D tokamak divertor region are reported for the first time. The resulting E×BT/B2 drift particle flux is calculated for standard (ion ∇BT drift toward divertor X-point) and reversed BT direction and for low (L) and high (H) confinement modes. Perpendicular field strengths of up to E∼5 kV/m are observed at the separatrix between the divertor private region and the scrape-off layer (SOL). The E×BT drift, which reverses with reversal of BT, creates a poloidal circulation pattern in the divertor that convects 25%–40% of the total ion flow to the divertor target. The circulation strongly couples the various regions of the divertor and SOL and fuels the X-point region. An outward shift of the profiles is seen in reversed BT. © 2000 American Institute of Physics.

    Notes: Intermittent plasma objects (IPOs) featuring higher pressure than the surrounding plasma, and responsible for ∼50% of the E×BT radial transport, are observed in the scrape off layer (SOL) and edge of the DIII-D tokamak [J. Watkins et al., Rev. Sci. Instrum. 63, 4728 (1992)]. Conditional averaging reveals that the IPOs, produced at a rate of ∼3×103 s−1, are positively charged and also polarized, featuring poloidal electric fields of up to 4000 V/m. The IPOs move poloidally at speeds of up to 5000 m/s and radially with E×BT/B2 velocities of ∼2600 m/s near the last closed flux surface (LCFS), and ∼330 m/s near the wall. The IPOs slow down as they shrink in radial size from 4 cm at the LCFS to 0.5 cm near the wall. The IPOs appear in the SOL of both L and H mode discharges and are responsible for nearly 50% of the SOL radial E×B transport at all radii; however, they are highly reduced in absolute amplitude in H-mode conditions. © 2001 American Institute of Physics.

    Notes: The radiation of divertor heat flux on DIII-D [J. Luxon et al., in Proceedings of the 11th International Conference on Plasma Physics and Controlled Nuclear Fusion (International Atomic Energy Agency, Vienna, 1987), p. 159] is shown to greatly exceed the limits imposed by assumptions of energy transport dominated by electron thermal conduction parallel to the magnetic field. Approximately 90% of the power flowing into the divertor is dissipated through low-Z radiation and plasma recombination. The dissipation is made possible by an extended region of low electron temperature in the divertor. A one-dimensional analysis of the parallel heat flux finds that the electron temperature profile is incompatible with conduction-dominated parallel transport. Plasma flow at up to the ion acoustic speed, produced by upstream ionization, can account for the parallel heat flux. Modeling with the two-dimensional fluid code UEDGE [T. Rognlien, J. L. Milovich, M. E. Rensink, and G. D. Porter, J. Nucl. Mater. 196–198, 347 (1992)] has reproduced many of the observed experimental features. © 1998 American Institute of Physics.
