Notes: Abstract Following injections of the insoluble portion of disrupted yeast cells ofBlastomyces dermatitidis animals exhibited a biphasic pyrogenic response, a decrease in total serum proteins, and there was no release of interferon.

Notes: Abstract Soluble and particulate spherule antigens fixed complement in tests with anti—C. immitis sera. However, the soluble antigen proved more active than the particulate one. Cross reactions were noted between the spherule antigens and anti—H. capsulatum and anti—B. dermatitidis sera. Following enzymatic treatment of the particulate preparation a soluble antigen was obtained which formed two bands with anti—C. immitis serum and one band with anti-H. capsulatum serum in an agar gel double diffusion test.

Notes: Conclusions Of the several morphological and physiological criteria that have been proposed to identifyTrichophyton rubrum andTrichophyton mentagrophytes differentially, only the hair invasion test agrees with the immunological results reported here. A typical strain ofT. rubrum andT. mentagrophytes was obtained from the Centraalbureau voor Schimmelcultures (Netherlands) and was used as reference strain. All isolates that did not invade hair shafts (in vitro) and that produced three precipitate lines in agargel diffusion tests against anti-T. rubrum serum (refence strain, Tr-1) were placed in one group. These isolates all also produced a red undersurface pigment and macroconidia with long parallel sides. They were labledT. rubrum. Other isolates that did invade hair shafts, and produced only two precipitate lines against anti-T. rubrum serum were placed in another group and were labeledT. mentagrophytes. These isolates had variable characteristics of pigment production, colony and conidial morphology. The antigenic studies thus corroborate the validity of the hair invasion test. The hair invasion test will probably remain in wider use because of its simplicity.

Notes: Summary The results of this study indicated that antigens prepared from the three morphological phases ofCoccidioides immitis differed in their complement fixing activity with anti-Histoplasma capsulatum pooled serum. Spherule antigens were serologically less active in tests with the anti-H. capsulatum pooled serum than antigens prepared from arthrospores and from mycelium. Antigenic determinants which are common toC. immitis andH. capsulatum appeared to be located on the intact arthrospore cellular surface but not on the surface of spherule cells.

Notes: Abstract Immunogens were found associated with particular fractions prepared from spherules ofCoccidioides immitis (Kong, Levine &Smith, 1963) and from yeast cells ofHistoplasma capsulatum (Salvin &Ribi, 1955). However,Blastomyces dermatitidis, another dimorphic systemic fungal pathogen was shown to elicit a minimal immunogenic response in experimental animals (Kong &Levine, 1967). It was therefore deemed pertinent to study factors which might enhance the resistance of mice to infection withB. dermatitidis.

Notes: Abstract Blastomyces dermatitidis, a dimorphic fungus, can infect man and cause chronic, suppurative, and granulomatous lesions in any part of the body. AlthoughSalfelder (1965) andDiSalvo &Denton (1963) commented on differences in virulence observed between isolates of this fungus, host factors pertaining to susceptibility have not been elucidated. BecauseConant et al. (1954) reported that males contract the natural disease 9 times more often than females, the present work was undertaken to study factors pertaining to this difference in susceptibility.
