    Notes: Abstract Mutantt haplotypes derived from thet 6 haplotype were typed forH-2. The mutantt h2 that arose fromt 6 due to crossing over in the region betweenT andtf had, as expected, lost theH-2 haplotype characteristic oft 6. The haplotypest h17,t h18, andt p1, which also arose by recombination, but which represent the complementary crossover products, including the distal part of thet 6 haplotype, carried the sameH-2 type ast 6. This suggests that crossing over betweentf andH-2 is suppressed int h17 andt 18. This in turn suggests that mutantt haplotypes suppress crossing over for that part of thet chromatin that they still retain. The origin oft h7, which apparently did not include any crossover distal toT, and which retains the crossover-suppressing property oft 6, retains thet 6 H-2 type. Unexpectedly, J h20 , which expressestf and was at first thought to have arisen due to crossing over, also retains theH-2 type oft 6. This provides part of the evidence thatt h20 arises fromt 6 not by crossing over, but by a small deletion, and hence that duplication and deletion are possible modes of origin of mutantt haplotypes.

    Notes: Summary Genetic studies on Searle'S X-autosome translocation of Mus musculus have shown that a segment of the X carrying the loci for two sex-linked genes, Ta and Blo, has been translocated to an autosome. Females heterozygous for Tabby normally have variegated hair structures, with transverse Ta bands visible against a background of wildtype fur. When the wild-type allele of Ta or Ta itself was located on the translocated X, it was capable of full expression in heterozygous females. Thus, the Tx+/+Ta genotype was completely wild-type, while TxTa/++ genotype showed the phenotype of the Ta hemizygote (Searle 1962; Lyon, Searle, Ford, and Ohno 1964). The present study on meiotic figures of the male carrying this translocation revealed that the reciprocal translocation was one-sided in favor of the autosome. While the at became nearly twice the size of the original autosome, the Xt became almost half the size of the Xn. The study of somatic prophase figures of the female carrying this translocation further showed that the Xn manifested positive heteropycnosis in 90 per cent of the cells. Thus, genes on the normal X should be inactivated in most somatic cells of the female, allowing full expression to the genes on the translocated X.

    Notes: The t haplotypes of mouse chromosome 17 bear a number of interesting mutations and rearrangements, some of which map close to the H-2 complex. Since there are many H-2 class I genes of unknown function, we have investigated their arrangement in t haplotypes using genomic Southern blots. We present a detailed chart of the H-2w30(tw12) complex, and compare it with the arrangement in other t haplotypes and standard mouse haplotypes. The chart shows duplications, deletions, and reshuffling of conserved and divergent regions. The two major features of the t arrangement-large deletions in the Qa and Tla regions–have analogues in some standard strains, so it is unlikely that these deletions are responsible for t-specific phenotypes. The differences between t and standard mouse strains are similar, in nature and in degree, to those between different standard strains.

    Notes: The chapter describes some personal reminiscences of various stages in the growth of knowledge of the mouse genome in the past 50 years. Initially mapping was done by crossing new mutants with linkage testing stocks, a slow and laborious method. In the 1950s major mutagenesis experiments led to spin-offs in terms of new mutants, new knowledge of phenomena including sex determination and X-chromosome inactivation, and further understanding of the t-complex. The 1970s saw the development of recombinant inbred (RI) strains and the use of biochemical variants for mapping. In addition the linkage groups were assigned to chromosomes. Techniques of embryo surgery were developed, leading to work with embryonic stem (ES) cells and hence to the identification of gene functioning by knockouts and transgenesis. Another major advance in the 1970s and 1980s was the beginning of comparative mapping, which is now so important. With the advent of DNA technology, progress in mapping increased considerably. Progress became even faster with the use of interspecific backcrosses and with the development of microsatellite markers. The completion of the mouse DNA sequence is now imminent, opening fascinating prospects for the analysis of gene function.
