    Description: New magnetostratigraphic results from a 3300 m-thick section across the syntectonic fluvial sediments of the Campodarbe Formation (Upper Eocene–Oligocene) in the Ebro foreland basin (NE Spain) are presented. The new data allow the top of the Campodarbe Formation to be correlated to Chron 7r (Chattian), younger than previously stated (C10r), therefore shifting the age of significant palaeogeographical changes in the foreland basin. The deformation in the southern front produces the cannibalization in the piggyback basin of 1300 m of sediments spanning c. 3.7 Myr. Average accumulation rates are lower in the Ebro foreland basin than in the piggyback basin and decrease from 35 to 27 cm kyr –1 by the time the San Felices thrust sheet activity decelerates (at c. 28 Myr). Shifts of accumulation rates result from accommodation space changes, which occur locally and are linked to the activity of the San Felices thrust, while the sediment supply occurs at orogenic scale (source of sediments is c. 200 km to the NE). Finally, sequence boundaries previously considered isochronous in the continental record of the Cenozoic Pyrenean basins are revealed to be 1.8–1 Myr older in the piggyback basin than in the Ebro foreland basin.

    Description: Genotyping microarrays are an important resource for genetic mapping, population genetics, and monitoring of the genetic integrity of laboratory stocks. We have developed the third generation of the Mouse Universal Genotyping Array (MUGA) series, GigaMUGA, a 143,259-probe Illumina Infinium II array for the house mouse ( Mus musculus ). The bulk of the content of GigaMUGA is optimized for genetic mapping in the Collaborative Cross and Diversity Outbred populations, and for substrain-level identification of laboratory mice. In addition to 141,090 single nucleotide polymorphism probes, GigaMUGA contains 2006 probes for copy number concentrated in structurally polymorphic regions of the mouse genome. The performance of the array is characterized in a set of 500 high-quality reference samples spanning laboratory inbred strains, recombinant inbred lines, outbred stocks, and wild-caught mice. GigaMUGA is highly informative across a wide range of genetically diverse samples, from laboratory substrains to other Mus species. In addition to describing the content and performance of the array, we provide detailed probe-level annotation and recommendations for quality control.

    Description: G-protein-coupled receptors (GPCRs) are seven-transmembrane proteins that mediate most cellular responses to hormones and neurotransmitters. They are the largest group of therapeutic targets for a broad spectrum of diseases. Recent crystal structures of GPCRs have revealed structural conservation extending from the orthosteric ligand-binding site in the transmembrane core to the cytoplasmic G-protein-coupling domains. In contrast, the extracellular surface (ECS) of GPCRs is remarkably diverse and is therefore an ideal target for the discovery of subtype-selective drugs. However, little is known about the functional role of the ECS in receptor activation, or about conformational coupling of this surface to the native ligand-binding pocket. Here we use NMR spectroscopy to investigate ligand-specific conformational changes around a central structural feature in the ECS of the beta(2) adrenergic receptor: a salt bridge linking extracellular loops 2 and 3. Small-molecule drugs that bind within the transmembrane core and exhibit different efficacies towards G-protein activation (agonist, neutral antagonist and inverse agonist) also stabilize distinct conformations of the ECS. We thereby demonstrate conformational coupling between the ECS and the orthosteric binding site, showing that drugs targeting this diverse surface could function as allosteric modulators with high subtype selectivity.     Description: The evolution of insect resistance threatens the effectiveness of Bacillus thuringiensis (Bt) toxins that are widely used in sprays and transgenic crops. Resistance to Bt toxins in some insects is linked with mutations that disrupt a toxin-binding cadherin protein. We show that susceptibility to the Bt toxin Cry1Ab was reduced by cadherin gene silencing with RNA interference in Manduca sexta, confirming cadherin's role in Bt toxicity. Native Cry1A toxins required cadherin to form oligomers, but modified Cry1A toxins lacking one alpha-helix did not. The modified toxins killed cadherin-silenced M. sexta and Bt-resistant Pectinophora gossypiella that had cadherin deletion mutations.     Description: Opium is one of the world's oldest drugs, and its derivatives morphine and codeine are among the most used clinical drugs to relieve severe pain. These prototypical opioids produce analgesia as well as many undesirable side effects (sedation, apnoea and dependence) by binding to and activating the G-protein-coupled micro-opioid receptor (micro-OR) in the central nervous system. Here we describe the 2.8 A crystal structure of the mouse micro-OR in complex with an irreversible morphinan antagonist. Compared to the buried binding pocket observed in most G-protein-coupled receptors published so far, the morphinan ligand binds deeply within a large solvent-exposed pocket. Of particular interest, the micro-OR crystallizes as a two-fold symmetrical dimer through a four-helix bundle motif formed by transmembrane segments 5 and 6.     Notes: Abstract. Lanthanum-modified lead titanate thin films have been prepared by a diol-based sol-gel method from precursor solutions with a growing excess of PbO. The films were crystallized with thermal treatments at 650 °C and heating rates of 10 °C  min-1 and higher than 500 °C min-1 by direct insertion in a pre-heated furnace. The structure, microstructure, and composition of the films were studied by grazing-incidence X-ray diffraction, electron microscopies, and energy-dispersive X-ray spectroscopy. A 20 mole % excess of PbO must be included in the precursor solution in order to compensate PbO volatilization occurring during the thermal treatment and, thus, obtain single-phase perovskite-type structure films. Non-textured porous films were obtained when a 10 °C min-1 rate is used, whereas [001]/[100] oriented films without porosity were obtained when rapid heating was used. Dielectric permittivity, ac current density, hysteresis loops, and switching curves were measured in the films. Values of remanent polarization and coercive field are comparable to those reported for similar films. The effects on the ferroelectric properties of an electrical and a post-crystallization thermal treatments were characterised.

    Notes: Abstract. Pb0.88La0.08TiO3 films were processed on Si-based substrates by a diol-based sol-gel route from solutions with variable content of PbO excess. Crystallisation was performed at heating rates of 10 °C min-1 and higher than 500 °C min-1 (rapid heating). The pyroelectric coefficient was measured after poling the samples by two methods: applying a sinusoidal wave and applying a train of square pulses, with the latter showing a higher poling efficiency. The piezoelectric d33 coefficient was determined by double-beam interferometry. Strain vs. field measurements provided evidence of 90° domain orientation in these films. Those crystallised at 10 °C min-1 showed the highest functional properties (γ=1.7×10-2 μC cm-2 K-1 and d33=57 pm V-1). This is a consequence of the higher stability of the 90° domains oriented during poling, caused by the lower tensile stress arising during preparation. The voltage responsivity of these films also benefited from the lower permittivity arising from their higher porosity. These films are good candidates for applications in infrared detectors and microelectromechanical devices.

    Notes: Although characterization of piezoceramics from resonance is a customary technique, theworks dealing with the determination of the ten elastic, dielectric and piezoelectric coefficients thatare needed in the full matrix characterization of such 6mm symmetry materials are rather scarce.Even more, if the complex characterization is foreseen, thus accounting with the three types oflosses, few are the methods avaliable to obtain the material linear coefficients. This work deals withsuch complex characterization by the use of Alemany et al. automatic iterative method. Thismethod has been applied to the four modes of resonance that are sufficient for the purpose: (1) thelength extensional mode of long rods, length poled, (2) the thickness extensional mode and (3) theradial mode of a thin disk, thickness poled, and (4) the thickness shear mode of a thin plate. Recentwork of the authors has shown the limits in characterizing materials that arise from the use of theStandard shear geometry and, therefore, and alternative geometry is used here.This work presents the matrix characterization of some piezoceramics and the Finite ElementAnalysis (FEA) simulation based on such characterization, of the samples used as a reliabilitycriteria of the results obtained by comparison of the experimental and simulated values at resonanceof the electrical parameters
