    Notes: Abstract The fragile X mental retardation syndrome is caused by an expansion of a trinucleotide repeat (CGG)n in the FMR-1 gene. Molecular genetic study of fragile X provides accurate diagnosis and facilitates genetic counseling in families with affected members. We present here the molecular study of 59 Spanish fragile X syndrome families using probe StB 12.3 and the polymerase chain reaction (PCR) of the (CGG)n repeat sequence of the FMR-1 gene. The results obtained have allowed us to characterize 455 individuals, including eight prenatal diagnoses. The clinical diagnosis of fragile X in 89 affected males was confirmed, 137 female carriers were identified (48 of whom were mentally retarded), 176 individuals “at risk” were found not to have the expansion, and 12 cases of normal transmitting males (NTM) were detected. In the sample studied, no de novo mutations were detected, nor any mutation different from that described for the (CGG)n expansion. One nonmentally retarded male was detected as having an unmethylated CpG island for the FMR-1 gene, but with more than 200 CGG repeats (high functioning male). The analysis of the (CGG)n repeat in 208 normal chromosomes gave an allele distribution similar to that in other Caucasoid population groups, with alleles of 29 and 30 CGG repeats accounting for 46% of the chromosomes. The combination of Southern analysis and PCR of the (CGG)n repeat is highly efficient for diagnosis, compared with cytogenetic techniques, especially in the detection of female carriers, NTMs, and prenatal diagnosis, enabling accurate genetic counseling to be provided in all cases.

    Notes: A full review for technological processes used today, in dental prostheses industry for titanium casting,both in Europe and in United States, reveal that, there is no equipment available on the market able tocast small titanium parts in secondary inert vacuum by induction melting and centrifugal casting. Allthese advanced processes together contribute to an original concept of the Denticast System. Thecomputer controlled cast sequence is another important factor contributing to a robust system, able toreproduce the achieved technological quality in a reliable manner.Mechanical testing (tensile and hardness) shows that the different alloys used in Denticast project andcast using the prototype, are in most of the results superior to those obtained with two differentcommercial systems (pressure-vacuum and centrifugal systems). The samples are usable for dentalprosthesis without any risk for the patient health

    Notes: The objective of the study was to evaluate the grazing behaviour by sheep in hill country paddocks in New Zealand which had received two long-term fertilization and stocking rate treatments [high fertility–high stocking rate (HH); low fertility–low stocking rate (LL)]. Herbage accumulation and selective grazing were evaluated within low slope (LS), medium slope (MS) and high slope (HS) categories. Transects lines were placed and tillers of Agrostis capillaris and Lolium perenne in the LS category; A. capillaris, Anthoxanthum odoratum and L. perenne in the MS category; and A. capillaris and A. odoratum in the HS category were marked. The leaf length of each marked tiller was measured and used to determine selective grazing over 3 weeks during each season. The highest herbage accumulation rates were during spring and the lowest in summer and winter. The LS category showed the highest herbage accumulation rates and HS the lowest. Except for autumn, the marked tillers were more frequently grazed in the HH than in the LL paddock. During summer, autumn and spring, grazing frequency in the slope categories was in the order LS 〉 MS 〉 HS. During winter sheep did not discriminate between slope category. During summer, autumn and spring, sheep did not selectively graze the species studied but this was not the case during winter. Overall, sheep selectively grazed L. perenne. In all the seasons L. perenne consistently had the longest leaves but within species there was no consistent relationship between leaf length and probability of being grazed. Selective grazing changed through the year according to herbage accumulation rate. Sheep concentrated grazing in the category LS when herbage accumulation rate was high, but they did not discriminate between slope categories in winter when herbage accumulation rate was low.

    Notes: Two commercial shrimp farms in south Texas were evaluated for influent and effluent water quality from June to October 1994. The intensive farm, Taiwan Shrimp Village Association (TSV) had an average annual yield of 4630 kg ha−1 while the semi-intensive farm, Harlingen Shrimp Farm (HSF), had a yield of 1777 kg ha−1. The study had three objectives: (1) to compare influent and effluent water from the intensive and semi-intensive shrimp farms, (2) to show which effluent water-quality indicators exceeded allowable limits, (3) to indicate inherent problems in farms operated with water exchange and summarize how findings from this study led to changes in farms' management that limited potential negative impact on receiving streams. Water samples were collected and analysed twice a week for the TSV farm and once a week for the HSF farm. Samples were analysed for dissolved oxygen (DO), salinity, pH, ammonia-nitrogen (NH3-N), nitrite-nitrogen (NO2-N), nitrate-nitrogen (NO3-N), total phosphorus (TP), total reactive phosphorus (TRP), five-day carbonaceous biochemical oxygen demand (cBOD5), total suspended solids (TSS) and settleable solids (SettSols). Most of the effluent constituents showed fluctuations throughout the sampling period often related to harvest activity. Effluent pH at TSV was lower than influent values but within the regulatory requirements set by Texas Commission of Environmental Quality (TCEQ), formerly known as Texas Natural Resource Conservation Commission (TNRCC). HSF effluent pH values were higher than its influent, but still within TCEQ limits. Effluent DO mean levels were generally below the regulatory daily mean requirement, with values at TSV often below those for influent. Effluent nutrient concentrations and net loads were generally higher at the intensive shrimp farm, with NH3-N mean concentrations above the daily mean set by the TCEQ on several occasions. Effluent TSS concentrations were higher than influent for both farms, with daily mean values above the TCEQ limit. The two farms presented similar TSS concentrations despite their different stocking densities. However, TSS total net load and net load per hectare were higher at the intensive farm. The semi-intensive farm presented higher cBOD5 concentrations and net loads despite its lower stocking density, with daily mean values above the TCEQ limit. The cBOD5 net load at TSV presented negative values indicating higher load at the influent than at the effluent. Analyses showed no evidence of self-pollution between influent and effluent at the two farms. The high feed conversion ratio (FCR) values (2.3 and 2.7 for the intensive and the semi-intensive farm respectively) suggest that better feed management is needed to reduce nutrient and solid net loads release from the two farms. The data obtained from this study resulted in several modifications in design and management of the two farms that reduced the potential negative impact on receiving streams. A brief summary of the improvement in selected effluent water-quality indicators at the intensive shrimp farm is provided.
