    Notes: The performance of serological tests for Helicobacter pylori infections is hampered by the persistence of antibodies after eradication therapy or spontaneous healing. Detection of different antigens or immunoglobulin classes might have an impact on the validity of serodiagnosis. The aim of this study was to assess the decrease in IgA and IgG antibody levels after eradication of H. pylori. Serum samples of 242 patients with active duodenal ulcer were tested with the ELISA and the immunoblot (IB) techniques for H. pylori-specific IgA and IgG antibodies before therapy and 1 year after successful eradication. From a total of 81 patients paired sera were available. At the end of the follow-up period ELISA antibody titres from the IgA class had decreased from a mean value of 6.69 to 4.26 units (P=0.0001), and IgG class antibody titres from a mean value of 21.9 to 12.1 units (P=0.0001). Regarding seroreversion, from 34 initially IgA positive sera 16 (47%), and from 74 IgG positive sera 18 (24%), had definitively reverted to ‘negative’. One year after eradication, when tested with the immunoblot, the antibody responses against specific antigens of 37% IgA-positive sera (23/62) and 8% IgG-positive sera (6/78) reverted to ‘negative’, compared to a seroreversion rate of 27% of the anti-CagA IgA-positive sera (18/67) and of 9% of the anti-CagA IgG-positive sera (7/79). In conclusion, despite an overall significant decrease of H. pylori antibodies, both tests cannot be recommended for monitoring treatment success.

    Notes: Investigations of 3d transition metal (TM) compounds by means of linear dichroism in TM K-NEXAFS will be reported. At this, the decomposition of the angular-dependent NEXAFS into orbital projected components are important problems. A survey is given on methods how to extract information on the geometrical, electronic and magnetic structure from linearly polarized NEXAFS and EXAFS spectra. New results on CuO and CuPc (Copper Phthalocyanine) are reported. A reference is given to the novel method for spin-resolving of NEXAFS by analysis of resonantly scattered X-ray core lines.

    Notes: Polarized Cu K edge X-ray absorption near-edge structure (XANES) spectra of CuO are analyzed. Partial spectral components reflecting both dipole and quadrupole transitions are resolved from the experiment. Theoretical spectra were obtained using the real-space multiple-scattering technique and by calculating the band structure via the pseudopotential method. We demonstrate that the pre-peak is of a quadrupole character and find its decomposition into individual d components. The self consistent pseudopotential calculation, free from any constraints on the form of the potential, improves the agreement between theory and experiment in those areas where real-space calculation, based on non-self-consistent muffin-tin potential, fails. Therefore we argue that the most significant contributions to the Cu K edge XANES come from one-electron processes.

    Notes: New diagnostic and prognostic markers are needed in colorectal cancer. They can be found by differential analysis at DNA, RNA or protein level. The accuracy of phenotypic comparisons of tumor and normal tissues depends on the purity of the samples. We present an effective method to identify and isolate proteins that are differentially expressed under altered conditions, and a two-dimensional reference protein map of the normal human colonic epithelium Normal colonic mucosa, primary tumors and liver metastases were prepared in the operating room. After washing in an ice-cold medium containing protease inhibitors, crypts were isolated by mechanical preparation without using metalloproteinases. Epithelial cells were then selected using Ber-EP4 Dynabeads. The samples were denaturated before processing for immobilized pH gradient two-dimensional polyacrylamide gel electrophoresis according to SWISS-2DPAGE standards. The samples contained more than 95% epithelial cells as confirmed by fluorescence-activated cell sorting using pan-anticytokeratin antibodies. Cell surfaces were not damaged, as assessed by scanning electronic microscope. A protein reference map of the normal colonic epithelium was defined. Using gel matching, N-terminal sequencing and/or immunoblotting techniques, 60 polypeptides - including proteins specifically expressed in colorectal epithelium - have now been identified. This reproducible method of sample preparation permits the comparison of protein patterns found in various pathological states with the present reference map (http://www.expasy.ch). Some of these patterns might provide diagnostic or prognostic markers, or even molecular targets for therapy in the future.

    Notes: Large tissue samples from ten patients operated for colorectal cancer were prepared in the operating room in iced phosphate buffered saline, containing ethylene diaminetetraacetic acid and protease inhibitors. After cutting the specimens into small fragments, the tissues were gently pressed through a steel mesh. Membranes were permeabilized in chilled ethanol 70% to allow cytosolic fluoresceine isothiocyanate labeling, performed with anti-cytokeratin (CAM 5.2) antibodies. Samples were quantitatively sorted with a fluorescence activated cell sorter (FACS) and denatured before processing separation by two-dimensional electrophoresis on polyacrylamide gels. This procedure made it possible to sample about 4 × 107 viable normal and tumoral cells before fixation, and up to 4 × 106 cells after FACS. The gels run before and after fixation showed no major differences. The rate of cytokeratin-positive cells in the samples was the following (mean, Cl 5-95%): mucosa 29.5% (8.9-66.7%), tumor 44.3% (6.6-94.8%). The epithelial cell content in colorectal cancer and normal mucosa shows important intersample variations. This is important for any comparison of fresh samples, whether at DNA, RNA, or at the protein level. We propose a method allowing the preparation of pure epithelial cell samples from normal and tumoral colonic fresh mucosa.

    Description: La famille de Darius, tableau peint en 1660-1661, marque le début de la carrière de Charles Le Brun comme peintre du roi, elle marque également le début du gouvernement personnel de Louis XIV. Cette œuvre ouvre une nouvelle époque de la peinture en France, elle montre l'importance du dialogue avec la théorie de l'art moderne d’origine italienne, et incarne également une politique royale qui poursuit l’idée d’intégrer l’art moderne à ses stratégies. Pour mieux en saisir la signification complexe, cette étude analyse l’œuvre sous divers angles, elle s’attache au point de vue de la théorie politique, comme à celui de l’historiographie ou encore à l’histoire de la psychologie.

    Description: Le portrait est sans conteste le genre artistique le plus fécond du début des Temps modernes. Dans la France du XVIIe siècle, qui se distingue par une mobilité sociale inconnue jusqu’alors, le portrait permet précisément d’appuyer la revendication d’un nouveau statut social ou d’assurer un rang acquis, mais désormais remis en question. Le portrait se fait également l’écho de la discussion capitale concernant le rapport entre le corps et l’âme. Pourtant, les sources écrites parvenues jusqu’à nous, qui s’intéressent au portrait, sont étonnamment parcimonieuses. L’Académie royale de peinture et de sculpture fondée en 1648, en particulier, est presque totalement muette à ce sujet. Et ce, bien que nombre de ses membres gagnent leur vie comme portraitistes et que le genre voie son importance s’accroître au cours du Grand Siècle : les portraits se multiplient, tandis que leur prix augmente constamment. Il semble que l’Académie ait sciemment passé sous silence le portrait et les débats afférents, afin de mieux célébrer comme sa véritable mission la peinture d’histoire, sur laquelle l’institution nous a laissé d’innombrables témoignages. La présente étude reconstitue les discours autour du portrait dans la France du XVIIe siècle et dévoile une discussion d’une vivacité surprenante, où d’aucuns se sont même demandé si le portrait ne méritait pas – plutôt que la peinture d’histoire – d’occuper la première place dans la hiérarchie des genres.

    Description: Lorsque Henri IV a décidé, au tournant du XVIIe siècle, d’utiliser l’art à des fins politiques, ses projets étaient peu spectaculaires d’un point de vue strictement artistique. Ce n’est que petit à petit que l’État absolutiste grandissant a aspiré à une nouvelle forme de représentation susceptible de répondre à des attentes artistiques plus élevées. Ainsi, les successeurs de Henri IV, Louis XIII et Louis XIV, ont eu de plus en plus recours à la peinture d’histoire, considérée comme le genre artistique le plus noble. Les aspirations artistiques et politiques se rejoignirent dans la recherche d’une peinture capable de transposer des histoires à narration complexe. Or, en France, un tel genre restait à élaborer et c’est la politique qui en fut à l’origine. Cette union entre l’art et la politique artistique se révéla profitable aussi bien pour l’État que pour l’art lui-même : si l’État a ainsi pu disposer d’un moyen de représentation convaincant, l’art y a trouvé une forme d’expression jusqu’alors inconnue en France et qui devint vite un modèle pour d’autres pays. Mais les deux chemins devaient rapidement diverger. Ne pouvant plus répondre aux exigences toujours plus fortes de la politique, l’art s’est soustrait doucement à son influence. La voie était alors tracée pour la quête d’un art moderne et libre, objet de tant de discussions au XVIIIe siècle.
