    Notes: Abstract We examined the chromosomal localization of the telomeric sequence, (TTAGGG)n, in seven species of the lemurs and one greater galago, as an outgroup, using the primed in-situ labeling (PRINS) technique. As expected, the telomeric sequence was identified at both ends of all chromosomes of the eight prosimians. However, six species showed a signal at some pericentromeric regions involving constitutive heterochromatin as well. The pericentromeric region of chromosome 1 of Verreaux's sifaka (Propithecus verreauxi verreauxi) was labeled with a large and intense signal. The range of the signal considerably exceeded the area of DAPI positive heterochromatin. On the other hand, in the five lemurs, a large signal was detected also in the short arm of acrocentric chromosomes. Acquisition of the large block of the telomeric sequence into the acrocentric short arm might be interpretable in terms of the tandem growth of the heterochromatic short arm and the reciprocal translocation between heterochromatic short arms involving the telomeric sequence. Subsequently, it was postulated that meta- or submetacentric chromosomes possessing the telomeric sequence at the pericentromeric region could be formed by centric fusion between such acrocentric chromosomes.

    Notes: Abstract During exploration of chromosome polymorphisms in Japanese macaques, a heteromorphic polymorphism was found in a population in the Zigokudani monkey park. The population consisted of three troops (social units). Of 36 monkeys examined, five females showed heterozygotic 'marker' chromosome (chromosome 9). The polymorphism was a tandem duplication of the nucleolus organizer region (NOR) of the short arm of chromosome 9, which was found for the first time in the genus Macaca. FISH and fibre-FISH using human 18S rDNA and sequential silver nitrate staining revealed that the duplicated region included a part of the euchromatic satellite and the stalk and that the euchromatic block (intercalary satellite) divided the NOR into two parts (distal and proximal). Furthermore, it showed that the distal region possessed much more rDNA than the proximal region, and that the duplications might have been introduced via a mechanism of gene amplification (inverted duplications associated with over-replication and recombination events). As the tandem duplication was observed sporadically in four maternal pedigrees in two troops and the mothers of the variants all had normal chromosomes, the variation might have been introduced from another population's gene pool by a solitary male immigrant.

Two phenotypes, “a” and “b,” were distinguished in homozygotes. It is suggested that these are controlled by autosomal codominant alleles. In 10 laboratory strains, only the IS strain showed the a type. This allele found in the IS strain was broadly distributed in Japanese wild rats. It is considered to be derived from a wild rat in Japan. Linkage relationship betweenTf andAlp-1 was not established.

    Notes: Abstract Cleavage patterns of mitochondrial DNAs (mtDNAs) by 15 restriction endonucleases were analyzed for 10 swamp and 13 river types of domestic water buffaloes. Digestions with nine enzymes exhibited polymorphisms giving two or three kinds of cleavage patterns. Five mtDNA types were identified, three types in the swamp buffaloes of the Philippines, Vietnam, and Indonesia (S-types) and two types in the river buffaloes of Bangladesh and Pakistan (R-types). Nucleotide diversities ranged from 0.2 to 0.6% within the S- and R-types and from 1.9 to 2.4% between the R-types and the S-types. These values indicated that R-type and S-type mtDNAs differentiated at thesubspecific level of other mammalianspecies reported. The possibility of polyphyletic domestication in different places is discussed for the origin of two distinct types of domestic water buffaloes.

    Notes: Abstract The cytochromeb genes of all living species ofBubalus, including the river type and the swamp type of domestic buffaloes (Bubalus bubalis), were sequenced to clarify their phylogenetic relationships. These sequences were compared together with the African buffalo (Syncerus caffer) and banteng (Bos javanicus) sequences as an outgroup. Phylogenetic trees ofBubalus species based on the DNA sequences of the cytochromeb gene demonstrated that the tamaraw (Bubalus mindorensis), endemic to the Philippines, could be classified into the subgenusBubalus, not the subgenusAnoa. The divergence time between the lowland anoa (B. depressicornis) and the mountain anoa (B. quarlesi) was estimated at approximately 2.0 million years (Myr), which is almost the same as the coalescence time for theBubalus sequences. This large genetic distance supports the idea that the lowland anoa and the mountain anoa are different species. An unexpectedly large genetic distance between the river and the swamp type of domestic buffaloes suggests a divergence time of about 1.7 Myr, while the swamp type was noticed to have the closest relationship with the tamaraw (1.5 Myr). This result implies that the two types of domestic buffaloes have differentiated at the full species level.

    Notes: Abstract The monkeys on the island of Sulawesi (Celebes), Indonesia, comprise seven species ofMacaca, that isM. maura, M. tonkeana, M. hecki, M. nigrescens, M. nigra, M. ochreata, andM. brunnescens. Hemoglobins from 248 individuals of these seven species were analyzed by isoelectric focusing electrophoresis (IEFE) and by starch gel electrophoresis in the presence of urea (USGE). Eighteen phenotypes consisting of eight molecular types were identified by IEFE analysis. The speciestonkeana inhabiting the central part of the island revealed 11 phenotypes, while peripheral species such asnigrescens andbrunnescens carried only 3 and 2 phenotypes, respectively. On USGE, three α chains and three β chains were identified and named α1, α2, and α6, and β1, β3, and β5, respectively. The α1 chain has the same mobility as the α chains of other macaques, while the α2 chain is less positively charged than α1, and α6 is the least positive among these α chains. The α2 chain is widely distributed in the Sulawesi macaques as the major component. Four species,ochreata, tonkeana, maura, andnigrescens, carried the α1 and α6 chains as minor components. The electrophoretic mobility of β1 was the same as that of other macaques, while β3 and β5 were more positively charged and less positively charged than β1, respectively. All of the Sulawesi species had β3 in high or low gene frequencies and inmaura, tonkeana, andbrunnescens, this type was most abundant. β5 chain existed in the species of the northern peninsula, as the major type. The subordinate type was β3 innigra andnigrescens and β1 inhecki. On the other hand, β1 was most frequently observed inochreata.

    Notes: Abstract Genetic variability in individual troops of the Japanese macaque (Macaca fuscata fuscata) was quantified by the proportion of polymorphic loci and the average heterozygosity per individual from the results of starch-gel electrophoreses of blood proteins controlled by 32 independent genetic loci. The former averaged 9.2% and the latter 1.3%, the values being remarkably lower than those estimated for other animal populations. Geographical distribution of the genetic variations was not uniform in the whole species but the variants occurred only in limited areas. Assuming the selective neutrality of segregating alleles and the two-dimensional stepping-stone model of population structure, the genetic migration rate between the local demes per generation could be estimated to average less than inverse of average effective deme size. Here, the local deme is not a troop itself, but it consists of several troops tightly connected with each other by frequent exchanges of reproductive males. Analyses of correlation between geographic and genetic distances between troops revealed that the gene constitutions of two troops apart more than 100 km on an island could be regarded as practically independent of each other. These results suggest that the population structure of the Japanese macaque species has a tendency to split into a number of local subpopulations in which the effect of random genetic drift is prevailing.

    Notes: Abstract The degree of genetic differentiation among three genera of cercopithecines,Macaca, Papio andCercopithecus, was quantified by the electrophoretic analyses of blood protein variations controlled by 29 genetic loci. The estimates ofNei's standard genetic distance between the pair of species belonging to different genera were in a range of 0.48–0.90. It was found that the degree of genetic differentiation between different genera of cercopithecines was comparable to those between congeneric species of other animal groups. This work failed, however, to determine definitely the branching order of the lineages of the three genera.
