    Description: Multiple studies have confirmed the contribution of rare de novo copy number variations to the risk for autism spectrum disorders. But whereas de novo single nucleotide variants have been identified in affected individuals, their contribution to risk has yet to be clarified. Specifically, the frequency and distribution of these mutations have not been well characterized in matched unaffected controls, and such data are vital to the interpretation of de novo coding mutations observed in probands. Here we show, using whole-exome sequencing of 928 individuals, including 200 phenotypically discordant sibling pairs, that highly disruptive (nonsense and splice-site) de novo mutations in brain-expressed genes are associated with autism spectrum disorders and carry large effects. On the basis of mutation rates in unaffected individuals, we demonstrate that multiple independent de novo single nucleotide variants in the same gene among unrelated probands reliably identifies risk alleles, providing a clear path forward for gene discovery.     Description: Brain development and function depend on the precise regulation of gene expression. However, our understanding of the complexity and dynamics of the transcriptome of the human brain is incomplete. Here we report the generation and analysis of exon-level transcriptome and associated genotyping data, representing males and females of different ethnicities, from multiple brain regions and neocortical areas of developing and adult post-mortem human brains. We found that 86 per cent of the genes analysed were expressed, and that 90 per cent of these were differentially regulated at the whole-transcript or exon level across brain regions and/or time. The majority of these spatio-temporal differences were detected before birth, with subsequent increases in the similarity among regional transcriptomes. The transcriptome is organized into distinct co-expression networks, and shows sex-biased gene expression and exon usage. We also profiled trajectories of genes associated with neurobiological categories and diseases, and identified associations between single nucleotide polymorphisms and gene expression.     Notes: Abstract – There has been little investigation of the winter ecology of adult trout during winter, especially in regard to concealment behavior. We compared day vs night underwater counts of adult rainbow trout and brook trout from four streams. At water temperatures between 1°C and 9°C, daytime counts accounted for 44% and 16% of nighttime snorkeling counts for rainbow trout and brook trout adults, respectively. As winter progressed, nighttime counts declined more so for brook trout than rainbow trout, but the decline was not significant for either species. Nocturnalism of both species was higher in streams with colder water temperatures. We observed few fish within concealment structure; however, by electrofishing concealment habitat during the day, we captured 10 times more adult trout than we counted immediately beforehand by snorkeling. Adult trout were concealed in cobble-boulder substrate and woody debris during the day./〉

    Notes: Summary One-dimensional expansion waves in 4 different shock tube driver geometries are investigated and compared. These are the plane wave in a straight tube and its reflection from a co-planar wall, the cylindrical wave in a wedge shaped duct and the spherical wave in a cone. Characteristics and pressure histories are computed for all cases. Pressure measurements in truncated cones are presented to check the predicted pressure histories for the spherical wave.

    Description: To better understand the molecular and cellular differences in brain organization between human and nonhuman primates, we performed transcriptome sequencing of 16 regions of adult human, chimpanzee, and macaque brains. Integration with human single-cell transcriptomic data revealed global, regional, and cell-type–specific species expression differences in genes representing distinct functional categories. We validated and further characterized the human specificity of genes enriched in distinct cell types through histological and functional analyses, including rare subpallial-derived interneurons expressing dopamine biosynthesis genes enriched in the human striatum and absent in the nonhuman African ape neocortex. Our integrated analysis of the generated data revealed diverse molecular and cellular features of the phylogenetic reorganization of the human brain across multiple levels, with relevance for brain function and disease.

    Description: Context. Sub-stellar objects exhibit photometric variability, which is believed to be caused by a number of processes, such as magnetically-driven spots or inhomogeneous cloud coverage. Recent sub-stellar models have shown that turbulent flows and waves, including internal gravity waves, may play an important role in cloud evolution. Aims. The aim of this paper is to investigate the effect of internal gravity waves on dust nucleation and dust growth, and whether observations of the resulting cloud structures could be used to recover atmospheric density information. Methods. For a simplified atmosphere in two dimensions, we numerically solved the governing fluid equations to simulate the effect on dust nucleation and mantle growth as a result of the passage of an internal gravity wave. Furthermore, we derived an expression that relates the properties of the wave-induced cloud structures to observable parameters in order to deduce the atmospheric density. Results. Numerical simulations show that the density, pressure, and temperature variations caused by gravity waves lead to an increase of the dust nucleation rate by up to a factor 20, and an increase of the dust mantle growth rate by up to a factor 1.6, compared to their equilibrium values. Through an exploration of the wider sub-stellar parameter space, we show that in absolute terms, the increase in dust nucleation due to internal gravity waves is stronger in cooler (T dwarfs) and TiO2-rich sub-stellar atmospheres. The relative increase, however, is greater in warm (L dwarf) and TiO2-poor atmospheres due to conditions that are less suited for efficient nucleation at equilibrium. These variations lead to banded areas in which dust formation is much more pronounced, similar to the cloud structures observed on Earth. Conclusions. We show that internal gravity waves propagating in the atmosphere of sub-stellar objects can produce banded clouds structures similar to that observed on Earth. We propose a method with which potential observations of banded clouds could be used to estimate the atmospheric density of sub-stellar objects.
