    Notes: Abstract Phage reactivation systems in Bacteroides fragilis were induced by far-UV irradiation, O2 and H2O2. These three treatments also induced the synthesis of 3, 6, and 4 protein bands, respectively, which were easily detectable by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Two proteins with apparent Mrs of approx. 90 000 and 70 000 were induced by all three treatments. Caffeine completely inhibited UV- and O2-induced phage reactivation and prevented the synthesis of the Mr 90 000 and Mr 70 000 proteins. The results suggest that these two proteins may be involved in phage reactivation processes induced by UV, O2 and H2O2 in B. fragilis.

    Notes: Abstract Reactivation of UV-irradiated phage b-1 was induced by H2O2 and UV in Bacteroides fragilis. The characteristics of H2O2 and UV induced phage reactivation differ from a previously reported oxygen induced reactivation system. The survival of B. fragilis cells after UV irradiation was also increased by pretreatment with H2O2. DNA synthesis was not inhibited in the host cells exposed to H2O2 concentrations which induced phage reactivation. The pattern of DNA degradation and synthesis after UV irradiation with and without H2O2 differed from the effect of O2 on DNA synthesis in irradiated B. fragilis cells.

    Notes: Abstract Bacteroides fragilis Bf-2 cells were more sensitive to far-UV radiation, N-methyl-N′-nitrosoguanidine, ethylmethane sulphonate, acriflavine and mitomycin C under aerobic conditions than under anaerobic conditions. The opposite effect was observed with H2O2-treated cells and exposure to O2 enhanced the survival of H2O2-treated cells. Pretreatment of cells with sublethal concentrations of H2O2 also increased the survival of H2O2-treated cells. Reactivation of UV- and X-irradiated and methylmethane sulphonate and H2O2-treated phage b-1 was induced by O2 and H2O2 in B. fragilis.

    Notes: Here, the proposal is investigated that protein tertiary structure prediction methods and threading methods in particular might be applied to the problem of solving a protein structure by X-ray crystallography, thus reducing the need for the more traditional experimental intensity methods of data phasing, such as heavy-metal isomorphous replacement and anomalous scattering methods, and without reference to a very closely related protein of known structure. If this kind of approach were to become successful and reliable, this would represent a significant advance in protein structure determination, offering an easy and accessible method for the initial data phasing for proteins' crystal structures, utilizing the vast amount of structural data, deposited in the Brookhaven PDB, that has been accumulated over the past 30 years of crystallographic structural studies. In the light of the ongoing structural genomics initiatives, the successful development of this kind of approach would be of enormous benefit.

    Notes: Summary A minimal medium was used to investigate the triggers regulating the initiation of solvent production and differentiation in Clostridium acetobutylicum P262. The accumulation of acid end-products caused the inhibition of cell division and the initiation of solvent production and cell differentiation. Initiation only occurred with a narrow pH range. Glucose or ammonium limited cultures failed to achieve the necessary threshold of acid end-products and solvent production and differentiation were not initiated. The addition of acid end-products or ammonium to cultures containing suboptimal levels of glucose or nitrogen respectively, enhanced solvent production. Resuspension of cells in media containing the threshold level of acid end-products and residual glucose induced endospore formation. Glucose or ammonium limitation did not induce sporulation and there was a requirement for glucose and ammonium during solventogenesis and endospore formation. Initiation of solvent production and clostridial stage formation were essential for sporulation. The induction of endospore formation in C. acetobutylicum P262 differs from that in the aerobic endospore forming bacteria where sporulation is initiated by nutrient starvation.
