Interactions of the Nonfunctional Coenzyme Binding Site in Dehydrogenases with [Nicotinamide-ribofuranosyl] [ω-(Adenine-9-yl)-n-alkyl] PyrophosphatesThe [nicotinamide-ribofuranosyl] [ω-(adenine-9-yl)-n-alkyl] pyrophosphates 1-41) are obtained by condensation of ω-(adenine-9-yl)-n-alkyl phosphates with nicotinamide mononucleotide. The coenzyme analogues show nearly identical physical and chemical properties as nicotinamide-adenine dinucleotide (NAD). The intramolecular interaction between adenine and the quaternary nicotinamide system is less compared to NAD⊕ and depends on the length of the hydrocarbon chains which in the models connect the adenine ring to the pyrophosphate group. The coenzyme models 1-4 were reduced enzymatically to the dihydro derivatives, the physical and chemical properties of which are accordant with those of NADH. The fluorescence spectra of the reduced coenzyme analogues resemble those of NADH. In the presence of dehydrogenases they show little or no increase in fluorescence intensity. In enzymatic tests with alcohol dehydrogenase from yeast and horse liver, lactate dehydrogenase and supernatant or mitochondrial malate dehydrogenase the oxidized coenzyme models and their dihydro derivatives are able to act as hydrogen acceptors as well as hydrogen donators. Compared to NAD⊕ and NADH they show high Michaelis constants. The catalytic constants are smaller than those of natural coenzymes. Tested with alcohol dehydrogenase from horse liver the coenzyme models 2, 3 and 4 show high catalytic constants.

Die [Nicotinamid-ribofuranosyl]-[ω-(adenin-9-yl)-n-alkyl]-pyrophosphate 1-41) werden durch Kondensation der ω-(Adenin-9-yl)-n-alkyl-phosphate mit Nicotinamidmononucleotid gewonnen. Die Coenzymanalogen zeigen nahezu gleiche physikalische und chemische Eigenschaften wie Nicotinamid-adenin-dinucleotid (NAD). Die intramolekulare Wechselwirkung zwischen Adenin- und quartärem Nicotinamidsystem ist kleiner als im nativen NAD⊕ und ist von der Länge der Kohlenwasserstoffkette abhängig, die in den Modellen den Adeninring mit der Pyrophosphatgruppe verknüpft. Die Coenzymmodelle 1-4 lassen sich enzymatisch zu den Dihydroderivaten reduzieren, deren physikalische und chemische Eigenschaften mit denen des NADH übereinstimmen. Die Fluoreszenzspektren der reduzierten Coenzymmodelle gleichen denen des NADH. In Gegenwart von Dehydrogenasen zeigen sie keine oder nur geringe Steigerung der Fluoreszenzintensität. Die oxidierten Coenzymanalogen und ihre Dihydroderivate sind im Test mit Alkohol-Dehydrogenase aus Hefe und Pferdeleber, Lactat-Dehydrogenase, cytoplasmatischer und mitochondrialer Malat-Dehydrogenase enzymatisch als Wasserstoffakzeptoren bzw. Wasserstoffdonatoren wirksam. Sie zeigen große Michaeliskonstanten verglichen mit NAD⊕ und NADH. Die maximalen Umsatzzahlen sind kleiner als die der natürlichen Coenzyme. Mit Alkohol-Dehydrogenase aus Pferdeleber zeigen die Analogen 2, 3 und 4 hohe Umsatzzahlen.
