    Notes: Abstract Cytogenetic preparations from young, adult, random bred female guinea pig oocytes were made on days 5–8 of vaginal cycles with an in vitro technique. First meiotic metaphase was harvested at 8 hours and second meiotic metaphase at 14 hours. The mean chiasmata count per bivalent was 1.035. A microscopic appearance suggesting lampbrushing was seen in some metaphase I bivalents.

    Notes: Abstract Cytological examination of pachytene stages of oogenesis and spermatogenesis revealed the presence of diagnostic loops in cells heterozygous for each one of six radiation induced lethal albino alleles in the mouse. Such loops were never found in any of the normal sibs or other controls. Such evidence supports the contention that all six of these mutations are deletions as indicated previously for five of them by complementation tests. An incidental finding was that of a pleiomorphic region on a medium sized pachytene bivalent in both sexes. This was not restricted to mutant cells but was also found in control cells. Whereas the pachytene deletion loops characterize lethal albino allele carriers, the observation of the pleiomorphic region reveals an apparently normal Cytological phenomenon in the mouse, the significance of which remains to be determined.

    Notes: Abstract A map of the mouse oocyte has been constructed utilizing early and mid/late pachytene stages. Each bivalent was clearly identified. At the mid/late stage 195 chromomeres corresponded directly to precursor chromomeres in the early stage, whilst 84 chromomeres were derived from two chromomeres from the early stage. The X and 13 bivalents were found to have a single chromomere in the early stage which later yielded two sites. There were approximately twice as many total chromomeres as mitotic bands, with good correspondence between pachytene chromomeres and major mitotic bands. Application of the use of mapping to a previously reported bivalent bearing a pleiomorphic region is noted.

    Notes: Abstract Pachytene chromomere maps of early and mid/late mouse spermatocytes have been prepared which permit exact identification of each bivalent. The average total number of chromomeres on the autosomal bivalents was 248 in the early cells and 184 in the mid/late. There was close correspondence between early and mid/late chromomeres in 122 locations. Comparisons of early pachytene chromomeres with published prometaphase dark G bands revealed 1.6 more chromomeres in the meiotic autosomal bivalents, with close correspondence of larger chromomeres and major mitotic bands. Fewer chromomeres were found in pachytene spermatocytes than had been seen in a previous study of pachytene oocytes. Comparisons of chromomeres of spermatocytes and oocytes revealed several differences.

    Notes: Summary Small, nucleolus-like structures were demonstrated in the nuclei of human diplotene oocytes. At least some of these bodies were shown to be true micronucleoli by virtue of their ability to bind rRNA during RNA-DNA hybridization in situ.

    Notes: Summary The effects of caffeine on meiosis of mouse ova in vitro and in vivo have been studied. Concentrations of 200 μg/cc or more suppressed entry of ova into in vitro meiosis without production of any numerical or structural abnormalities at dose levels above or below that. Acute superovulation by the standard technique was prevented at a dose of 0.25 mg/g/body weight and examination of the intrafollicular ova revealed presistence of the germinal vesicle stage. Chronic administration of the same dose (0.25 mg/g/bw) throughout the period from weaning to puberty did not induce any cytologically detectable meiotic abnormalities in ova studied in vitro at the end of that period. It is suggested that the reduced fertility seen by some previous authors may have been due to the suppression of entry into meiosis of the ova with diminished numbers of ova being ovulated.

    Notes: The concept of Byskov which hypothesizes a glycoprotein inducer of meiosis for the mammalian fetal ovary has been tested by examining the incorporation of the specific precursor 3H-fucose into the rete ovarii of day 14 mouse ovaries. Semiquantitation of grains in autoradiographs has documented a concentration over the rete ovarii which exceeded that over the oogonia, ovarian fibroblasts, or surface epithelia. These data support the concept that the cytoplasm of mouse rete ovarii is capable of synthesizing a glycoprotein that may be the meiosis-inducing substance.

    Notes: It has been suggested that the production of estrogens by the fetal ovary may modulate the entry or progression of meiosis in the female mammalian fetus. In the present study the possibility that the site of this steroid synthesis is the rete ovarii system was explored in the fetal mouse of gestational ages 12.5 to 18 days. The method of ultracytochemical localization of 3β hydroxy-steroid ferricyanide was used. Reaction product was found in the cytoplasm of the rete ovarii (prefollicular) cells as early as day 14 with increasing amounts seen at later gestational ages. The presence of this essential enzyme system in cells closely applied to oogonia and oocytes during an active meiotic period must be considered in developing concepts of meiotic entry.

    Notes: A quantitative analysis of mitochondrial populations during the meiotic prophase of mouse oogenesis was carried out. The mean absolute area occupied by mitochondria and the mean number of mitochondria per cell increases in a linear fashion from pachytene through dictyate. The mean area occupied by mitochondria increases at pachytene and thereafter. Both small and large aggregations of mitochondria are seen, particularly during the later stages of prophase. Vacuolated mitochondria are present from preleptotene through pachytene. Mitochondria show major dynamic changes throughout fetal mouse oogenesis, which may suggest significant functional activities yet to be elucidated.

    Notes: A combination of autoradiography and air-dried techniques was used to calculate the duration of the major meiotic stages in the first wave of spermatogenesis in the newborn mouse. The data indicated that the entry into meiosis occurred asynchronously over 2 days, and the time required for each stage and the total cycle was constant. These time intervals were nearly identical with those estimated for adult animals in the present study and by other authors.
