    Description: A role for B-cell-receptor (BCR) signalling in lymphomagenesis has been inferred by studying immunoglobulin genes in human lymphomas and by engineering mouse models, but genetic and functional evidence for its oncogenic role in human lymphomas is needed. Here we describe a form of 'chronic active' BCR signalling that is required for cell survival in the activated B-cell-like (ABC) subtype of diffuse large B-cell lymphoma (DLBCL). The signalling adaptor CARD11 is required for constitutive NF-kappaB pathway activity and survival in ABC DLBCL. Roughly 10% of ABC DLBCLs have mutant CARD11 isoforms that activate NF-kappaB, but the mechanism that engages wild-type CARD11 in other ABC DLBCLs was unknown. An RNA interference genetic screen revealed that a BCR signalling component, Bruton's tyrosine kinase, is essential for the survival of ABC DLBCLs with wild-type CARD11. In addition, knockdown of proximal BCR subunits (IgM, Ig-kappa, CD79A and CD79B) killed ABC DLBCLs with wild-type CARD11 but not other lymphomas. The BCRs in these ABC DLBCLs formed prominent clusters in the plasma membrane with low diffusion, similarly to BCRs in antigen-stimulated normal B cells. Somatic mutations affecting the immunoreceptor tyrosine-based activation motif (ITAM) signalling modules of CD79B and CD79A were detected frequently in ABC DLBCL biopsy samples but rarely in other DLBCLs and never in Burkitt's lymphoma or mucosa-associated lymphoid tissue lymphoma. In 18% of ABC DLBCLs, one functionally critical residue of CD79B, the first ITAM tyrosine, was mutated. These mutations increased surface BCR expression and attenuated Lyn kinase, a feedback inhibitor of BCR signalling.     Description: Control of intracellular reactive oxygen species (ROS) concentrations is critical for cancer cell survival. We show that, in human lung cancer cells, acute increases in intracellular concentrations of ROS caused inhibition of the glycolytic enzyme pyruvate kinase M2 (PKM2) through oxidation of Cys(358). This inhibition of PKM2 is required to divert glucose flux into the pentose phosphate pathway and thereby generate sufficient reducing potential for detoxification of ROS. Lung cancer cells in which endogenous PKM2 was replaced with the Cys(358) to Ser(358) oxidation-resistant mutant exhibited increased sensitivity to oxidative stress and impaired tumor formation in a xenograft model.     Description: The electrical conductivity of gabbro was investigated at 623–1173 K, 0.5–2.0 GPa and oxygen fugacities controlled by three solid buffers (Cu + CuO, Ni + NiO and Mo + MoO 2 ) using a YJ-3000t multianvil press and Solartron-1260 impedance spectroscopy analyzer within the frequency range 10 –1 –10 6 Hz. The measured electrical conductivity shows an Arrhenius increase with temperature and increases with increasing pressure. The activation energy and activation volume of charge carriers have been determined as 0.72±0.01 eV and –6.8 ± 0.69 cm 3 /mol, respectively. The hydrogen-assisted electrical conductivity in gabbro decreases with increasing oxygen fugacity. Furthermore, it is inferred that some hydrogen-related defects with minor concentration ( e.g. , H' M or H • ) are the main charge carriers in the sample and provide reasonable explanations for the behavior of electrical conductivity of gabbro at high pressure.

    Description: Centromeres are defined by the presence of CENH3, a variant of histone H3. Centromeres in most plant species contain exclusively highly repetitive DNA sequences, which has hindered research on structure and function of centromeric chromatin. Several maize centromeres have been nearly completely sequenced, providing a sequence-based platform for genomic and epigenomic research of plant centromeres. Here we report a high resolution map of CENH3 nucleosomes in the maize genome. Although CENH3 nucleosomes are spaced ~190 bp on average, CENH3 nucleosomes that occupied CentC , a 156-bp centromeric satellite repeat, showed clear positioning aligning with CentC monomers. Maize centromeres contain alternating CENH3-enriched and CENH3-depleted subdomains, which account for 87% and 13% of the centromeres, respectively. A number of annotated genes were identified in the centromeres, including 11 active genes that were located exclusively in CENH3-depleted subdomains. The euchromatic histone modification marks, including H3K4me3, H3K36me3 and H3K9ac, detected in maize centromeres were associated mainly with the active genes. Interestingly, maize centromeres also have lower levels of the heterochromatin histone modification mark H3K27me2 relative to pericentromeric regions. We conclude that neither H3K27me2 nor the three euchromatic histone modifications are likely to serve as functionally important epigenetic marks of centromere identity in maize.

    Description: Perovskite-structured Ba 1− x La x SnO 3 ( x  = 0–0.10) films have been directly grown on (0001) sapphire substrates by a sol-gel method. Optical properties and bandgap energy of the films have been investigated by transmittance spectra from 10 K to 450 K. It indicates that these films exhibit a high transmission of more than 80% in the visible region. With increasing temperature, there is a significant bandgap shrinkage of about 0.5 eV for lightly La doping ( x  ≤ 0.04) films. For heavily La doping concentration ( x  ≥ 0.06), the bandgap remains nearly stable with the temperature and La composition. This is due to the fact that the lattice expansion caused by La doping is close to the saturation for the film doped with x  = 0.06. Moreover, temperature dependent conductivity behavior shows a similar pattern, which suggests that the doping concentration of La-doped BaSnO 3 (BLSO) films has a saturated state. The La introduction can modify the Sn 5 s -O 2 p antibonding state and the nonbonding O 2 p orbital, which remarkably affect the electronic bandgap of the BLSO films.

    Description: Raman scattering of (K 0.5 Na 0.5 )NbO 3 -0.05LiNbO 3 -yMnO 2 (y = 0% and 1.0%) single crystals have been reported in the temperature range from 300 to 800 K. The spectra exhibit a competition between a soft mode and a relaxation mode upon heating across the diverse transitions. The progressive change in the conflicting displacive mechanism (soft mode) and order-disorder (relaxation mode) can explain the origin of the successive orthorhombic-tetragonal-cubic phase transitions. Moreover, the polymorphic phase transition between orthorhombic and tetragonal structures can be confirmed through the observation of thermotropic phase boundaries for MnO 2 -doped (K 0.5 Na 0.5 )NbO 3 -0.05LiNbO 3 single crystals.
