Description: National-scale emissions of carbon tetrachloride (CCl4) are derived based on inverse modeling of atmospheric observations at multiple sites across the United States from the National Oceanic and Atmospheric Administration’s flask air sampling network. We estimate an annual average US emission of 4.0 (2.0–6.5) Gg CCl4 y−1 during 2008–2012, which is...

Description: Alcohol dependence is a major public health challenge in need of new treatments. As alcoholism evolves, stress systems in the brain play an increasing role in motivating continued alcohol use and relapse. We investigated the role of the neurokinin 1 receptor (NK1R), a mediator of behavioral stress responses, in alcohol dependence and treatment. In preclinical studies, mice genetically deficient in NK1R showed a marked decrease in voluntary alcohol consumption and had an increased sensitivity to the sedative effects of alcohol. In a randomized controlled experimental study, we treated recently detoxified alcoholic inpatients with an NK1R antagonist (LY686017; n = 25) or placebo (n = 25). LY686017 suppressed spontaneous alcohol cravings, improved overall well-being, blunted cravings induced by a challenge procedure, and attenuated concomitant cortisol responses. Brain functional magnetic resonance imaging responses to affective stimuli likewise suggested beneficial LY686017 effects. Thus, as assessed by these surrogate markers of efficacy, NK1R antagonism warrants further investigation as a treatment in alcoholism.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉George, David T -- Gilman, Jodi -- Hersh, Jacqueline -- Thorsell, Annika -- Herion, David -- Geyer, Christopher -- Peng, Xiaomei -- Kielbasa, William -- Rawlings, Robert -- Brandt, John E -- Gehlert, Donald R -- Tauscher, Johannes T -- Hunt, Stephen P -- Hommer, Daniel -- Heilig, Markus -- Intramural NIH HHS/ -- New York, N.Y. -- Science. 2008 Mar 14;319(5869):1536-9. doi: 10.1126/science.1153813. Description: Circulating tumour cells (CTCs) shed into blood from primary cancers include putative precursors that initiate distal metastases. Although these cells are extraordinarily rare, they may identify cellular pathways contributing to the blood-borne dissemination of cancer. Here, we adapted a microfluidic device for efficient capture of CTCs from an endogenous mouse pancreatic cancer model and subjected CTCs to single-molecule RNA sequencing, identifying Wnt2 as a candidate gene enriched in CTCs. Expression of WNT2 in pancreatic cancer cells suppresses anoikis, enhances anchorage-independent sphere formation, and increases metastatic propensity in vivo. This effect is correlated with fibronectin upregulation and suppressed by inhibition of MAP3K7 (also known as TAK1) kinase. In humans, formation of non-adherent tumour spheres by pancreatic cancer cells is associated with upregulation of multiple WNT genes, and pancreatic CTCs revealed enrichment for WNT signalling in 5 out of 11 cases. Thus, molecular analysis of CTCs may identify candidate therapeutic targets to prevent the distal spread of cancer.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3408856/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3408856/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Yu, Min -- Ting, David T -- Stott, Shannon L -- Wittner, Ben S -- Ozsolak, Fatih -- Paul, Suchismita -- Ciciliano, Jordan C -- Smas, Malgorzata E -- Winokur, Daniel -- Gilman, Anna J -- Ulman, Matthew J -- Xega, Kristina -- Contino, Gianmarco -- Alagesan, Brinda -- Brannigan, Brian W -- Milos, Patrice M -- Ryan, David P -- Sequist, Lecia V -- Bardeesy, Nabeel -- Ramaswamy, Sridhar -- Toner, Mehmet -- Maheswaran, Shyamala -- Haber, Daniel A -- 5K12CA87723-09/CA/NCI NIH HHS/ -- 5R01EB008047/EB/NIBIB NIH HHS/ -- CA129933/CA/NCI NIH HHS/ -- P01 CA117969/CA/NCI NIH HHS/ -- R01 CA129933/CA/NCI NIH HHS/ -- U01 EB012493/EB/NIBIB NIH HHS/ -- Howard Hughes Medical Institute/ -- England -- Nature. 2012 Jul 26;487(7408):510-3. doi: 10.1038/nature11217.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Massachusetts General Hospital Cancer Center, Harvard Medical School, Boston, Massachusetts 02114, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/22763454" target="_blank"〉PubMed〈/a〉

Description: Compared with pure metals and ionic salts, covalent solids such as silicon are hard and brittle because dislocations do not move in them except at high temperatures. A satisfactory explanation for this behavior has been lacking in spite of its great importance for the mechanics of materials and structures. It is shown here that the critical atomic process leading to the observed brittleness is analogous to a chemical substitution reaction. Analysis of this analogy with the aid of a correlation diagram yields the observed high resistive stress and high activation energy. When a kink on a dislocation line moves, it breaks the atomic bonding symmetry, a forbidden process.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Gilman, J J -- New York, N.Y. -- Science. 1993 Sep 10;261(5127):1436-9.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/17745356" target="_blank"〉PubMed〈/a〉

Description: The novel intemal structures of metallic glasses lead to exceptional strength, corrosion resistance, and ease of magnetization. Combined with low manufacturing costs, these properties make glassy ribbons attractive for many applications. These materials also have scientific fascination because their compositions, structures, and properties have unexpected features.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Gilman, J J -- New York, N.Y. -- Science. 1980 May 23;208(4446):856-61.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/17772809" target="_blank"〉PubMed〈/a〉
