    Description: The ATP-dependent chromatin assembly and remodelling factor (ACF) functions to generate regularly spaced nucleosomes, which are required for heritable gene silencing. The mechanism by which ACF mobilizes nucleosomes remains poorly understood. Here we report a single-molecule FRET study that monitors the remodelling of individual nucleosomes by ACF in real time, revealing previously unknown remodelling intermediates and dynamics. In the presence of ACF and ATP, the nucleosomes exhibit gradual translocation along DNA interrupted by well-defined kinetic pauses that occurred after approximately seven or three to four base pairs of translocation. The binding of ACF, translocation of DNA and exiting of translocation pauses are all ATP-dependent, revealing three distinct functional roles of ATP during remodelling.     Description: Evenly spaced nucleosomes directly correlate with condensed chromatin and gene silencing. The ATP-dependent chromatin assembly factor (ACF) forms such structures in vitro and is required for silencing in vivo. ACF generates and maintains nucleosome spacing by constantly moving a nucleosome towards the longer flanking DNA faster than the shorter flanking DNA. How the enzyme rapidly moves back and forth between both sides of a nucleosome to accomplish bidirectional movement is unknown. Here we show that nucleosome movement depends cooperatively on two ACF molecules, indicating that ACF functions as a dimer of ATPases. Further, the nucleotide state determines whether the dimer closely engages one or both sides of the nucleosome. Three-dimensional reconstruction by single-particle electron microscopy of the ATPase-nucleosome complex in an activated ATP state reveals a dimer architecture in which the two ATPases face each other. Our results indicate a model in which the two ATPases work in a coordinated manner, taking turns to engage either side of a nucleosome, thereby allowing processive bidirectional movement.     Description: Although substantial progress has been achieved in the structural analysis of exporters from the superfamily of adenosine triphosphate (ATP)-binding cassette (ABC) transporters, much less is known about how they selectively recognize substrates and how substrate binding is coupled to ATP hydrolysis. We have addressed these questions through crystallographic analysis of the Atm1/ABCB7/HMT1/ABCB6 ortholog from Novosphingobium aromaticivorans DSM 12444, NaAtm1, at 2.4 angstrom resolution. Consistent with a physiological role in cellular detoxification processes, functional studies showed that glutathione derivatives can serve as substrates for NaAtm1 and that its overexpression in Escherichia coli confers protection against silver and mercury toxicity.     Notes: We demonstrate a simple calibration method of a rf magnetic probe by utilizing a two port network analysis method. The sensitivity (V/G) of the probe is determined by measuring the scattering parameters of S11 and S21. The magnetic field generated by the Helmholtz coil is calculated from S11 and the probe sensitivity is obtained from S21. The measured probe sensitivity is 0.1–0.6 V/G in the frequency range of 1–15 MHz. © 1999 American Institute of Physics.

    Notes: A frequency modulated interferometer is developed, which uses a 94 GHz Gunn oscillator and a single sideband (SSB) upconverter as a frequency modulator. An SSB upconverter makes it possible to use a heterodyne phase measurement technique with one mm-wave source. In addition, the influence of frequency drift and phase noise of a mm-wave source on the phase measurement can be reduced. In this work, the performance of a SSB upconverter as a frequency modulator is examined. Also, overall performance of the mm-wave interferometer with a SSB upconverter and line density measurement results in a mirror device are described. © 1999 American Institute of Physics.

    Notes: The flux-surface-averaged linearized resistive magnetohydrodynamic (MHD) boundary-layer equations including the compressibility, diamagnetic drift, and neoclassical viscosity terms are derived in toroidal geometry. These equations describe the resistive layer dynamics of resistive MHD modes over the collisionality regime between the banana plateau and the Pfirsch–Schlüter. From the resulting equations, the effects of neoclassical viscosity on the stability of the tearing and resistive ballooning modes are investigated numerically. Also, a study is given for the problem of how the neoclassical resistive MHD mode is generated as the collisionality is reduced. It is shown that the neoclassical viscosity terms give a significant destabilizing effect for the tearing and resistive ballooning modes. This destabilization comes mainly from the reduction of the stabilizing effect of the parallel ion sound compression by the ion neoclassical viscosity. In the banana-plateau collisionality limit, where the compressibility is negligible, the dispersion relations of the tearing and resistive ballooning modes reduce to the same form, with the threshold value of the driving force given by Δc=0. On the other hand, with the finite neoclassical effect it is found that the neoclassical resistive MHD instability is generated in agreement with previous results. Furthermore, it is shown that this later instability can be generated in a wide range of the collisionality including near the Pfirsch–Schlüter regime as well as the banana-plateau regime, suggesting that this mode is a probable cause of anomalous transport.

    Notes: The characteristics of electron cyclotron resonance (ECR) plasmas at the plug chamber of the "Hanbit" plasma facility have been investigated. The chamber length and diameter are 2.25 m and 1.5 m, respectively, and three pairs of solenoids are used to produce a variety of magnetic field configurations. Two magnetrons of 5 kW maximum output power each are used to produce the ECR plasma, and the microwaves are launched in extraordinary mode (X-mode) and/or right-hand circular polarized mode (R-mode). The dependence of the radial plasma density profile and the energy distribution of end loss ions upon the magnetic field strength and geometry, gas pressure, microwave power, and the mode of microwave are investigated. The plasma parameters are measured by using a position-controllable electrostatic probe, an x-ray spectrometer, and a multigrid ion energy analyzer. Transitions, occurred by threshold magnetron powers between low and high density modes, are found on certain conditions of magnetic field geometry and gas pressure. The density profile and the ion energy distribution are strong functions of magnetic field geometry and gas pressure, respectively. © 1998 American Institute of Physics.

    Notes: The diamagnetic effect is measured by using the diamagnetic loop with a compensation loop in the central cell of the Hanbit magnetic mirror device. To ensure reliable measurement, the influence of nearby conductors on the diamagnetic loop signal is investigated, and the output voltages during a pulsed vacuum field are measured to calibrate the loops installed inside the central cell. The diamagnetic measurement is carried out during rf discharges with a frequency of 3.75 MHz and heating power of up to 150 kW. In the measurement, the vacuum field of 0.126 T is applied in the central cell. The results from the experimental measurements are presented. © 2001 American Institute of Physics.
