    Notes: Summary Treatment of 3T3 cell plasma membranes with glycosidase enzymes decreased their ability to inhibit cell growth and also decreased their binding to 3T3 cells. This suggests that carbohydrate is required for complete function of inhibitory activity and that inhibition is associated with membrane adhesion.

    Notes: Abstract  The biocontrol agent Pseudomonas fluorescens F113, which produces the antimicrobial compound 2,4-diacetylphloroglucinol and inhibits soil-borne phytopathogenic fungi, might also affect non-target resident microorganisms involved in the cycling of soil nutrients. Analyses of soil nutrients and crop foliage were used to assess possible residual effects of the pseudomonad on soil fertility. The Bandon field site was sown in 1994 with sugarbeet seeds that were either untreated, inoculated with the spontaneous rifampicin-resistant mutant F113Rif, or treated with chemical fungicides. The field was cropped with uninoculated red clover in 1995. The inoculant was recovered at only 2.0 log CFU/root system at the first harvest of clovers, and the F113Rif treatment influenced 5 of 17 levels of readily available nutrients determined by electro-ultrafiltration (EUF) analysis of soil. However, this effect was essentially negligible since it was (1) small in magnitude and of little agronomic significance when compared with reference EUF levels, (2) similar in part to those of the commercial seed treatment and other farming practices followed at Bandon, and (3) not statistically significant when data were analysed at the plot level rather than using results from individual soil samples within plots. Clover leaf composition was investigated at two harvests. The F113Rif treatment had no effect on the 20 foliage parameters studied except a small (15%) transient decrease in foliage chlorine at the first harvest. EUF soil analysis and foliage analysis will be useful in future risk assessment studies of genetically modified derivatives of F113.

    Notes: Abstract Indigenous strains of R. trifolii, originating from different geographical regions, were compared using total DNA restriction analysis, plasmid DNA profiles and intrinsic antibiotic resistance patterns. The results of strain relatedness using 2 of these techniques (DNA restriction and antibiotic resistance patterns) were similar, and indicated that these strains formed a heterogeneous group. A large variation in plasmid DNA profiles was observed among the R. trifolii isolates investigated, indicating heterogeneity of plasmid DNA pools. This variation occurred even in some strains that were shown to be related on the basis of their antibiotic resistance patterns. The application of these techniques to differentiate indigenous R. trifolii strains is discussed in this report.

    Notes: This study describes the macromixing within a hydro-dynamic vortex separator and considers its potential as a disinfectant contact tank. It is typically used in the sedimentation process during the treatment of sewage. The macromixing was investigated by conducting tracer experiments from which the retention time distribution was obtained and interpreted to characterize the mixing. The separator was operated in a continuous mode with no baseflow, and the retention-time distribution was obtained using a pulse input for a range of flow-rates. The method of moments was used to obtain various functions and axial dispersion and tanks-in-series model parameters.

    Notes: In fry, 1- and 2-year old juvenile Atlantic salmon, relatively small superficial red muscle fibres staining well for glycogen and succinic dehydrogenase (SDH) but with little myofibrillar ATPase, were evident on either side of the lateral line, in June. Well differentiated relatively large white fibres contained lower concentrations of glycogen than the red fibres, higher ATPase and no SDH, except traces in the larger 1- and 2-year-olds. Intermediate size pink fibres, which were also intermediate between red and white fibres in their staining properties, occurred in a thin diffuse layer along the red-white boundary, thickest at the apex near the vertebral column, and most evident in the younger fish.

    Notes: Both red and white muscle fibre numbers in juvenile Atlantic salmon increased gradually with fish length throughout the freshwater growth period. Mean fibre area increased as fish grew to 6.5 cm f.l., but thereafter was unrelated to fish length. Hyperplasia was most obvious when fish were growing fastest, and was the dominant growth process in fish over 6.5 cm f.l. Hypertrophy was most important when growth was slow, as in autumn and winter.Mean white fibre area was significantly smaller in deep muscle than at medial and superficial sites. Total cross-sectional area of red, white and total trunk muscle increased with fish length. The ratio of red: white cross-sectional area increased with fish length to a plateau at about 10% after 6.5 cm f.l.

    Notes: The investigation established and validated methods to measure multiple biochemical indices of condition (whole body total lipids, whole body triglycerides, muscle RNA : DNA ratio and muscle protein) simultaneously in the same individual juvenile fish. It also provided examples of their application using a species comparison (salmonids and cyprinids) of the degree of change in these indices after food deprivation. The results showed that juvenile rainbow trout Oncorhynchus mykiss were much more susceptible to a decline in all biochemical indices of condition than juvenile fathead minnows Pimephales promelas upon food deprivation. The combination of these techniques can be used to accurately assess condition and growth potential of individuals from wild fish populations.
