α-Connected Di- and Trisaccharides of D-Ribofuranose1β-Halogeno-ribofuranuronic acid ester 1 and mono-and disaccharides with primary respectively secondary free hydroxylic groups highly stereoselectively yield α-connected di- and trisaccharides (2a-9a) under Koenigs-Knorr conditions. 1is a synthon to effect conveniently and in high yield the synthesis of α-(1→5) connected oligosaccharides in two steps (formation of the glycosidic bond, specific reduction of the ester group). The 2,3-O-isopropylidene protective group is hydrolyzed more readily with α-glycosides than with β-glycosides. The protective groups of 2a-9a are partly removed by hydrogenolysis and hydrolysis; thereby the unprotected di- and trisaccharides 3f, 5f, and 8f are obtained. The anomeric configuration of O-glycosides of ribo- and lyxofuranose and derivatives is easily determined from the 1H-NMR chemical shift of the isopropylidene groups.

Aus dem 1β-Halogen-ribofuranuronsäureester 1 und Mono- und Disacchariden mit primären bzw. sekundären freien Hydroxygruppen werden unter Koenigs-Knorr-Bedingungen hoch-stereoselektiv α-verknüpfte Di- und Trisaccharide (2a-9a) synthetisiert. Mit 1 als Baustein werden in zwei Schritten (Knüpfung der glycosidischen Bindung, spezifische Reduktion der Estergruppe) α-(l→5)-verknüpfte Oligosaccharide bequem und in hoher Ausbeute erhalten, bei welchen die 2,3-O-Isopropylidenschutzgruppe leichter abgespalten wird als bei β-Verknüpfung. Durch Hydrogenolyse und Hydrolyse werden die Schutzgruppen von 2a-9a teilweise oder ganz entfernt und u. a. die ungeschützten Di- und Trisaccharide 3f, 5f und 8f hergestellt. Die Anomerenkonfiguration von O-Glycosiden der Ribo- und Lyxofuranose und Derivaten kann aus der chemischen Verschiebung der 1H-NMR-Signale der Isopropylidengruppen bequem bestimmt werden.
