    Notes: The aim of this work was to define the metabolic factors which regulate the respiratory pathways in trangenic potato tubers. We previously found that respiration is enhanced in transgenic tubers which express a yeast invertase and a glucokinase from Zymomonas mobilis. In this study we investigated glycolysis in three further transgenic potato lines with profound changes in the mobilization of sucrose. We studied antisense ADPglucose pyrophosphorylase lines which are characterized by a reduction in starch accumulation and a significant build up of sucrose and related metabolic intermediates. We also report the generation of two novel double transgenic lines where the yeast invertase is expressed specifically in tubers of the ADPglucose pyrophosphorylase antisense line, targeted to either the cytosol or apopolast. We evaluated whether the localization of sucrose cleavage had an impact on the glycolytic induction, and assessed if invertase expression in the high-sucrose background had any further effects on glycolysis. We found that induction of the glycolytic enzymes only occurs when the invertase is targeted to the cytosol, and that the extent of this induction was comparable in the wild type and antisenseADPglucose pyrophosphorylase backgrounds. We conclude that the signal regulating glycolysis is directly linked to cytosolic sucrose hydrolysis.

    Notes: Summary The basal swellings on the processes of the cone cells of Pteronemobius include conspicuous “bodies” consisting of a double unit membrane as well as of many granules (β-glycogen?). These membranes are infoldings of the cell membrane. According to their site these structures seem to be a continuation of the rhabdome to which they lie very close distally. These “bodies” probably could—like the rhabdome-act as light guides, and together with pigment prevent an intrusion of light into deeper regions of the head.

    Description / Table of Contents: Summary Nineteen long-term individual- and group-feeding experiments with 180 male calves, 338 growing bulls, 302 heifers, and 344 dairy cows were carried out in order to measure the influence of feeding (straw, silages or green fodder as roughages) and different vitamin-A supplies (0–40 000 IU per 100 kg body weight per day in growing cattle or 0–120 000 IU per dairy cow per day) on liver vitamin-A concentration. All together, 2 127 biopsies from livers were taken for retinol analysis. At the end of six growth experiments animals were slaughtered. Liver vitamin-A concentration of calves depends on their term at birth and is associated with the carotene intake of their mothers. The carotene content of feeds and the vitamin-A supply are the most important influencing factors on liver vitamin-A concentration of growing and lactating cattle. On the average, livers of calves fed with colostrum contained 100–200 IU, those of growing cattle fed with grass and legumes or with silages contained 200–300 or 100–200 IU resp., and those of cows fed with green fodder or silage contained 300–600 or 100–300 IU vitamin A resp. per g fresh liver. There were also values outside of the variations mentioned above. The vitamin-A storage capacity of liver and the effects of oral and parenteral vitamin-A supply to depleted calves and growing cattle were also tested.
    Notes: Zusammenfassung In insgesamt 19 langfristigen Einzel- und Gruppenfütterungsversuchen mit 180 männlichen Kälbern, 338 Jungmastbullen, 302 weiblichen Jungrindern und 344 Milchkühen wurde der Einfluß der Fütterung (Stroh, Silagen oder Grünfutter als Grundfutter) und unterschiedlich hoher Vitamin-A-Zulagen (0–40 000 IE Vitamin A je 100 kg Lebendmasse und Tag bei wachsenden Rindern bzw. 0–120 000 IE je Milchkuh und Tag) auf die Leber-Vitamin-A-Konzentration ermittelt. Im Rahmen der Untersuchungen wurden insgesamt 2 127 Leberbioptate auf ihren Vitamin-A-Gehalt untersucht. Am Ende der Wachstumsversuche erfolgte teilweise eine Schlachtung der Tiere; die Vitamin-A-Bestimmung wurde in den Bioptaten und der Gesamtleber vorgenommen. Die Leber-Vitamin-A-Konzentration der Kälber zu Versuchsbeginn hängt vor allem vom Geburtstermin und damit der Karotinversorgung ihrer Mutter ab. Der Karotingehalt der Grundration und die Vitamin-A-Zulage zu den Rationen haben bei wachsenden und laktierenden Rindern wesentlichen Einfluß auf die Höhe der Leber-Vitamin-A-Konzentration. Im Mittel enthielten die Lebern der Kälber nach Kolostralmilchgabe 100–200, der wachsenden Rinder bei Grünfutter-(Gras, Leguminosen) 200–300 und bei Silagefütterung (Grassilage, Maissilage) 100–200 und der Milchkühe nach Grünfutter- bzw. Silageeinsatz 300 bis 600 bzw. 100–300 IE Vitamin A je g Frischmasse, wobei auch Meßwerte außerhalb dieser Variationsbereiche auftraten. In verschiedenen Depletionsversuchen mit Kälbern und wachsenden Rindern wurden die Vitamin-A-Speicherkapazität der Leber getestet sowie die Wirkungen oraler und parenteraler Zulagen verglichen.

    Description / Table of Contents: Summary The influence of an increasing protein supply in combination with soybean oil upon the IGF-I concentration in the serum in correlation with growth was measured on 8×10 male Wistar rats. With a casein content of 0 % in the food, the IGF-I level was 0.13±0.02 rU/ml. An IGF-I plateau of 0.74±0.07 rU/ml was reached at some 15 % casein. The additional application of 3 % soybean oil increased the IGF-I concentration significantly (P〈0.01) up to 0.95±0.16 rU/ml. The investigations show a specific increase of the IGF-I synthesis by the addition of oil, which is paralleled by a further stimulation of the growth of the rats. The nutrition-dependent IGF production in the peripheral tissues (mainly liver) represents the connection link between the growth hormone axis (genetically potential growth) and the growth realizable depending on the supply with nutrients.
    Notes: Zusammenfassung An 8×10 männlichen Wistar-Ratten wurde der Einfluß einer steigenden Proteinversorgung in Kombination mit Sojaöl auf die IGF-I-Konzentration im Serum in Korrelation zum Wachstum gemessen. Bei einem Caseingehalt von 0 % im Futter betrug der IGF-I-Spiegel 0,13±0,02 rU/ml. Ein Plateau wurde bei etwa 15 % Casein erreicht (0,74±0,07 rU/ml). Die zusätzliche Applikation von 3 % Sojaöl erhöhte die IGF-I-Konzentration signifikant (P〈0,01) auf 0,95±0,16 rU/ml. Die Untersuchungen zeigen eine spezifische Steigerung der IGF-I-Synthese durch die Ölzulage, der parallel eine weitere Stimulation des Wachstums der Ratten folgt. Die ernährungsabhängige IGF-Produktion in den peripheren Geweben (bes. Leber) stellt die Schaltstelle zwischen der Wachstumshormon-Achse (genetisch mögliches Wachstum) und dem in Abhängigkeit von der Nährstoffversorgung realisierbaren Wachstum dar.
